    Notes: TETRAMETHOPRIM (9) and PENTAMETHOPRIM (10), both prepared from the corresponding benzaldehydes 3 and 6 by conventional procedures, did not exhibit noteworthy antibacterial activity in vitro. A single crystal X-ray analysis of 9 and 10 provided evidence that the out-of-plane methoxy groups in the two compounds created a completely different topographical situation to that present in TRIMETHOPRIM, and less ideal for the binding to bacterial dihydrofolate reductase.

    Notes: Bromination of (-)-4-hydroxy-N-methylmorphinan-6-one (3), prepared from natural morphine, with 1 mol of bromine in acetic acid, afforded the 1-bromo ketone 5. The structure of 5 was assigned by 13C-NMR.spectroscopy, and confirmed by X-ray diffraction analysis of its hydrobromide salt. It is suggested that monobromination of synthetic (±)-2,4-dihydroxy-N-formylmorphinan-6-one (7) takes in principle a similar course, although the 13C-NMR.spectrum of the primary reaction product 9 could not be measured because of insolubility in commonly used solvents. Monobromination of (-)-4-acetoxy-N-formylmorphinan-6-one (12) of the natural series, and of (±)-2,4-diacetoxy-N-formylmorphinan-6-one (8) of the synthetic series, followed by treatment of the monobrominated ketones with potassium carbonate in methanol resulted in closure of the O-bridge, and afforded after acid hydrolysis, the corresponding 4,5-epoxy-morphinan-6-ones (-)-16 and (±)-17 respectively. This variation of the ring closure reaction represents a novel and convenient method to convert 4-hydroxymorphinan-6-ones into their corresponding 4,5-epoxymorphinan-6-ones, without involving aromatic bromination and with only 1 mol of bromine.

    Notes: The packing of peptide helices in crystals of the leucine-rich decapeptide Boc-Aib-Leu-Aib-Aib-Leu-Leu-Leu-Aib-Leu-Aib-OMe provides an example of ladder-like leucylleucyl interactions between neighboring molecules. The peptide molecule forms a helix with five 5→1 hydrogen bonds and two 4→1 hydrogen bonds near the C terminus. Three head-to-tail NH ċ O = C hydrogen bonds between helices form continuous columns of helices in the crystal. The helicial columns associate in an antiparallel fashion, except for the association of Leu ċ Leu side chains, which occurs along the diagonal of the cell where the peptide helices are parallel. The peptide, with formula C56H102N10O13, crystallizes in space group P212121 with Z = 4 and cell parameters a = 16.774(3) Å, b = 20.032(3) Å and c = 20.117(3) Å; overall agreement factor R = 10.7% for 2014 data with |Fobs| 〈 3σ(F); resolution 1.0 Å.

    Notes: The crystal structures of two helical peptides Boc-Val-Ala-Leu-Aib-Val-Ala-Leu-OMe (VALU-7) and Boc-Val-Ala-Leu-Aib-Val-Ala-Leu-Aib-OMe (VALU-8) have been determined to a resolution of 1.0 and 0.9 Å, respectively. Both the seven and eight residue peptides crystallize with two conformers per asymmetric unit. The VALU-8 conformers are completely helical and differ only at the C-terminus by a sign reversal of the φ, ψ angles of the last residue. One of the VALUE-7 conformers occurs as a normal α-helix, whereas in the other, the N(7)=O(3) α-type hydrogen bond is ruptured by the entry of a water molecule (W) into the helix, which in turn makes hydrogen bonds N(7)⃛W = 2.97 Å and ⃛O(3) = 2.77 Å. The other side of the water molecule is surrounded by a hydrophobic pocket. These two conformers give a static representation of a step in a possible helix unwinding or folding process. In the value-8 crystal the helices aggregate in a parallel mode, whereas the aggregation is antiparallel in the VALU-7 crystal. The crystal parameters are VALUE-7 crystal. The crystal parameters are VALUE-7, P21, a = 10.203 (3) Å, b = 19.744 (6) Å, c = 22.561 (6) Å, α = 96.76°, Z = 4, C38, H69N7O10·0.5 H2O, R = 6.65% for 3674 reflections observed 〉3σ(F): and VALU-8, P21, a; = 10.596 (4) Å, b = 27.57 (6) Å, c = 17.745 (5) Å, β = 95.76 (3)°, Z = 4, C42H76N76O11·0.25 CH3OH, R = 6.63% for 4701 reflections observed 〉3σ(F).

    Description / Table of Contents: Frog Venoms.Isolation and Structure of Pumiliotoxin CPumiliotoxin C, a base of the composition C13H25N, is isolated as the crystalline hydrochloride from the skin extracts of Dendrobates pumilio, a strikingly colored Panamanian frog. Complete X-ray analysis of this new venom by the so-called symbolic addition procedure establishes the structure as 2-n-propyl-5-methyl-cis-decahydroquinoline, a type of base, though related to coniine, is here reported for the first time as a natural product. The relative configurations of the four asymmetric centers are elucidated. In addition to this venom there are the congeners pumiliotoxins A and B, as well as further homologs and analogs which have also been isolated from the related frog Dendrobates auratus, and characterized by mass spectra.
    Notes: Aus dem Hautextrakt des in Panama vorkommenden, auffallend gefärbten Froschs Dendrobates pumilio läβt sich das gut kristallisierende Hydrochlorid einer Base Pumiliotoxin C der Formel C13H25N isolieren. Durch Röntgen-Strukturanalyse dieser Base nach dem „symbolischen Additionsverfahren“ (symbolic addition procedure) läβt sich zeigen, daβ es sich um einen zum ersten Mal in der Natur beobachteten, dem Coniin weitläufig verwandten Giftstofftyp handelt, nämlich um ein 2-n-Propyl-5-methyl-cis-decahydrochinolin, dessen vier asymmetrische Zentren in ihrer relativen stereochemischen Bezeichnung aufgeklärt werden. Neben Pumiliotoxin C leiten sich auf Grund der Massenspektren die Pumiliotoxine A und B sowie noch weitere aus dem verwandten Frosch Dendrobates auratus isolierte Basen von einem ähnlichen cis-Deca- oder Octahydrochinolin-System ab.
