    Notizen: Direct anaerobic bioconversion of cellulosic substances into ethanol by Clostridium thermocellum ATCC 27405 has been carried out at 60°C and pH 7.0 (initial for 100 L) under continuous sparging of oxygen free nitrogen in a culture vessel. Raw bagasse, mild alkali-treated bagasse, and solka floc were used as substrates. The extent of conversion of raw bagasse (cellulose, 50%; hemicellulose, 25%; lignin, 19%) was observed as 52% (w/w) and 79% (w/w) in the case of mild alkali and steam-treated bagasse (cellulose, 72%; hemicellulose, 11%; lignin, 12%), respectively. Use of bagasse concentration above 10 g/L showed a decreased rate in ethanol production. An inoculum age between 28-30 h and cell mass content of 0.027-0.036 g/L (dry basis) were used. The results obtained with raw and pretreated bagasse have been compared with those of highly pure Solka Floc (hemicellulose, 10%). Studies on the product inhibition indicated a linear fall of the percent of survivors with time. An Arrhenius type correlation between the cell decay rate constant and the product concentration was predicted. Even at low levels, the inhibitory effects of products on cell viability, the specific growth rate, and extracellular cellulase enzyme were observed.

    Notizen: Synthesis of the β-subunit of human chorionic gonado-tropin (βhCG) in Vero cells by the recombinant vaccinia virus has been studied. The yield of βhCG was a function of the multiplicity of infection (MOI), and was highest at 25 MOI. The kinetics of synthesis and initial secretion of βhCG, deduced from the pulse-chase experiments were “zero order.” At 30 h postinfection, the relative values of net synthesis and secretion rates were 4.0 AU. mm2 βhCG/106 cells. h and 1.55 AU. mm2 βhCG/106 cells. h, respectively. The time required to secrete 50% of intracellular βhCG was 210 min. Pulse-chase data also showed that 24% of βhCG was degraded intracelluiarly within 10 h, of which 17% was detected in the autoradiogram. Along with 30 kD βhCG, a satellite band of 28 kD was evident among the peptide synthesized in Vero cells. The molecular weight of vaccinia-derived βhCG was 13 kD more that its nonglycosylated form, indicating extensive glycosylation in Vero cells. The mRNA levels in infected Vero cells at different postinfection times were quantified by excess DNA dot-blot hybridization. It appears that the Vero cell possesses some host cell-associated factor(s), which prevented the transcription of early βhCG-mRNA promoted by the early signal of the vaccinia P 7.5 promoter. The half-life of βhCG-mRNA, as determined by follow-up of decay after blocking transcription initiation, was found to be 6.4 h. The synthesized βhCG was immunoreactive as it reacted with monoclonal and polyclonal monospecific antibodies. The subunit was also biologically active, as it combined with native βhCG to form heterodimer βhCG, which competed with 125I-hCG for radioreceptors and stimulated testosterone synthesis in Leydig cells. © 1995 John Wiley & Sons, Inc.

    Notizen: Cultivation of Trichoderma reesei QM 9414 on 3% (w/v) cellulose medium (C/N ratio = 8.5) produced 4.5 IU/ml celulase 180 hr at a cell growth of 8.0 g/liter (0.266 g cell/g cellulose). It corresponded to an average cellulase productivity 25.0 IU/liter/hr (3.5 IU/g cell/hr). In the same medium 9.5 g/liter cell mass (0.316 g cell/g cellulose), 6.2 IU/ml cellulase, and 38.75 IU/liter/hr (4.0 IU/g cell/hr) cellulase productivity could be obtained using pH cycling condition during cultivation. Cell mass, cellulase yield, and productivity were further increased to 10.0 g/liter, 7.2 IU/ml, and 44.0 IU/liter/hr (4.5 IU/g cell/hr), respectively, by simultaneous pH cycling and temperature profiling strategy. Results are described.

    Notizen: Bioconversion of acetic acid to methane by a crude culture of methanogens in a continuous multireactor system was investigated. Culture of methanogens was drawn from an active cow-dung digester (12 days) and was grown in a semisynthetic medium (pH 6.3, 37°C) with acetic acid as the sole carbon source. The solubilities of CO2, HCO3-, and CO32- increased with the rise in pH and exercised considerable influence on the gas composition. Various mechanisms for methanogenesis of acetic acid based on the available pathways were considered. Experimental data were compared with these mechanisms, the best fit was determined, and the corresponding rate expression was identified. This mechanism predicted that, of the total methane produced, 72%;comes from acetic acid directly and 28%;via the CO2 reduction route.

    Notizen: Bacillus amyloliquefaciens H produces a restriction endonuclease enzyme BamHl which is heat labile even at low temperatures. Studies were conducted to enhance thermal stability of BamHl using cross-linking reagents, namely, glutaraldehyde, dimethyl adipimidate (DMA), dimethyl suberimidate (DMS), and dimethyl 3,3′-dithiobispropionimidate (DTBP). Reaction with glutaraldehyde did not result in a preparation with enhanced thermal stability. However, the DMA-, DMS-, and DTBP-cross-linked preparations of BamHI exhibited significant improvement in thermal stability. Studies on thermal denaturation of the cross-linked enzyme preparations revealed that these do not follow a true first-order kinetics A possible deactivation scheme has been proposed in which the enzyme has been envisaged to go through a fully active but more susceptible transient state which, on prolonged heat exposure, exhibits a first-order decay kinetics. At 35°C, which is close to the optimum reaction temperature of 37°C for BamHl activity, the half-line of DMA-, DMS-, and DTBP-cross-linked preparations were 4.0, 5.25, and 5.5 h, respectively, whereas the native enzyme exhibited a half-line of 1.2 h only. The apparent values of deactivation rate constants for native, DMA-, DMS-, and DTBP-cross-linked BamHl were 1.13, 0.39, 0.29, and 0.26 h-1, respectively, at the same temperature, and the apparent values of activation energies for denaturation of native, DMA-, DMS-, and DTBP-cross-linked BamHl were 2.63, 5.24, 6.55, and 9.2 kcal/mol, respectively. The DTBP-cross-linked Bam HI was, therefore, the best heat-stable preparation among those tested. The unusually low values of activation energies for denaturation of Bam Hl represent their highly thermolabile nature compared to other commonly encountered enzymes such as trypsin, having activation energies of more than 40 kcal/mol for their denaturation.
