    Notizen: Summary Frozen sections of eight primary malignant lymphomas of the brain were examined by the immunohistochemical methods using a panel of monoclonal and heterologous antibodies to B lymphocyte (immunoglobulins, BA-1, Leu-12 and HLA-DR), T lymphocyte (OKT-11 and Leu-1) and monocyte (OKM-1) surface markers. Paraffin sections were also used in the examination of cytoplasmic immunoglobulins. Surface and/or cytoplasmic immunoglobulins (Ig) were observed in seven cases,four of which were shown to be distinctly monoclonal and the other three less so. The remaining 1 case showed no distinct staining for Ig. BA-1, Leu-12 and HLA-DR stainings were positive in four, four and five cases, respectively. The marker phenotypes of (BA-1, Leu-12, HLA-DR) were shown to be (+,+,+) in one lymphoma, (+,-,-) in three, (-,+,+,)in three, and (-,-,+) in one. Thus, it was demonstrated that the present lymphoma cases showed a marked immunological heterogeneity, and it was shown that all of them including the Ig-negative case revealed one or more of these three additional B cell markers, indicating B cell lineage of these cases. Examination of T cell and monocyte markers revealed positive staining in normal or reactive lymphoid cells distributed around blood vessels or sporadically in tumor tissues, but not in lymphoma cells. Epstein-Barr virus (EBV)-associated nuclear antigen was not demonstrated in the seven cases examined, making it unlikely that these lymphomas were related with EBV infection.

    Notizen: Summary Using the Southern blot hybridization technique, four cases of the primary malignant lymphomas of the brain, histologically diffuse large cell lymphoma, were examined for the immunoglobulin gene rearrangements. In three lymphomas, the rearrangements were observed in both heavy and light chain genes, providing strong evidence for a B cell lineage of these tumors. On the other hand, in the remaining lymphoma, the rearrangement was observed only in the heavy chain gene. Despite this, immunohistochemical examination revealed positive stainings for heavy and light chain immunoglobulins in tumor cells, suggesting the occurrence of light chain gene rearrangement at the undetectable level. Thus, B lymphocytic differentiation at the gene level was demonstrated in three, or possibly all, of the primary intracerebral malignant lymphomas examined. Since no more than two rearrangements were detected in each immunoglobulin gene, these lymphomas were considered to be monoclonal in nature.

    Notizen: Summary An immunohistochemical study was performed on small lymphoid cells present in frozen tissue sections of seven cases of primary B cell malignant lymphomas of the brain by using monoclonal antibodies to T cell (Leu-1, OKT-11, Leu-3a, and Leu-2a) and B cell (BA-1 and Leu-12) surface markers. In all the seven cases, positive reaction for Leu-1 and OKT-11 was seen in the majority of the small lymphoid cells which were dispersed among the lymphoma cells or clustered around blood vessles. The large neoplastic cells were unstained by these antibodies. Staining for T cell subsets with antibodies to Leu-3a and Leu-2a showed heterogeneous staining in each case. The ratio of Leu-3a+ to Leu-2a+ cells was less than one in six cases, demonstrating a suppressor/cytotoxic phenotype predominance. Most of these small lymphoid cells were negatively stained by antibodies to BA-1 and Leu-12. From these findings, it was shown that the small lymphoid cells observed in primary B cell lymphomas of the brain were of T cell lineage, distinct from the neoplastic cells, and probably reactive in nature. The implications of these findings are discussed.

    Notizen: The effect of LiCl, NaCl, and CsCl as univalent salts, and of CaCl2, ZnCl2, and MgCl2 as divalent salts, on the α and antiparallel β-sheet, and random conformations of poly(L-lysine) (PLL), in water at room temperature were examined by means of CD and compared quantitatively on the basis of elliptical strength at the maximal peak. Changes in the α-helical and antiparallel β-sheet helical conformations of PLL were markedly dependent on the salt concentrations of LiCl, NaCl, and CsCl, which induced decreases in negative intensity in that order. The CD spectrum of the random conformation, the most disordered form, displayed positive cotton effect in concentrations of these salts up to 3.0M and a negative peak in concentrations of 6.0M. The effect of these salts on the random conformation of PLL was stronger than that on the α- and β-conformations in higher concentrations. The CD spectrum of the random conformation in the presence of CaCl2, ZnCl2, and MgCl2, on the other hand, showed negative cotton effect in salt concentrations as low as 3.0M. It was impossible, however, to measure the effect on α- and β-conformations of ZnCl2 and MgCl2 above concentrations of 10 mM because of a solubility problem with salts in alkaline solution.

    Notizen: Studies on combined supercritical-fluid chromatography (SFC) using a packed column, with fast-atom bombardment mass spectrometry (FAB-MS) are described. The interfacing was successfully carried out by simply coupling a frit interface between the liquid chromatograph (LC) and the mass spectrometer. The interface was equipped with a pneumatic splitter and post-column addition device delivering FAB matrix material to a release valve which maintained a constant back-pressure at the column outlet. Using a 1% glycerol solution as a matrix compound in the release valve, the SFC/FAB-MS combination operates as well as a normal LC/MS instrument. The performance was demonstrated with a mixture of α-, β- and γ-tocopherol and Triton X-100 (poly(ethylene glycol) p-isooctylphenyl ether).

    Notizen: In contrast to many previous investigations, reaction conditions were found under which the nitration of durene (nitrating ratio of 1:1) gives predominantly, and in high yield, mononitrodurene rather than the usually formed dinitrodurene (plus unreacted durene). The method consists in nitrating by nitronium phosphorohexafluoride in nitromethane as solvent in presence of two equivalents of water.With mixed acid (HNO3 + H2SO4) in nitromethane and in acetonitrile durene forms also monoand no dinitrodurene; but under most conditions by-products are formed. Some of the by-products were identified.Some preliminary mechanistic results are reported: An addition complex of unknown structure is formed rapidly; 3, 6-dideuterodurene + D2O do not show a hydrogen isotope effect; the preferential formation of dinitrodurene under conventional conditions is due to the fact that the reaction occurs at encounter-controlled rate.

    Notizen: The ammonium perchlorate (AP)-oxidized composite propellants, each of which contains separately copper chromite (CC) as a burning rate adjuster and carbon black (CB) as an opacifier, have been ignited at subatmospheric pressures of argon gas by means of a carbon dioxide laser, and the effects of the additives on the ignition behavior have been studied. It has been found that copper chromite shortens the ignition time especially below 100 torr and that at the same time it enhances the ignitability, i.e., self-sustaining ignition.Carbon black, being an opacifier decreasing reflectivity and increasing radiative absorption at propellant surface, can not be recognized to be an active catalyst in ignition at subatmospheric pressures.The data of differential thermal analysis (DTA) for above specimens have indicated that the maximum exothermic peak temperature is shifted toward a lower one with the increase in CC concentration, the exothermic peak structure becoming sharper. However, CB addition to the basic propellant makes exothermic peaks less distinct. The results of DTA support those obtained from the ignition experiments above.
