    Notes: Es wird Über Versuche berichtet, die unternommen wurden, die Bedingungen zu studieren, die die Lichtempfindlichkeit des Zinksulfids beeinflussen.Es wird im einzelnen Folgendes festgestellt: 1 Die Belichtungstemperatur besitzt keinen wesentlichen Einfluß auf den Ablauf der Photolysereaktionen.2 Die Kristallstruktur ist entgegen der bisherigen Auffassung ohne wesentliche Bedeutung.3 Lichtempfindlichkeit und Korngrößenverteilung der Präparate gehen parallel, und es lassen sich aus einem Präparat durch Fraktionieren nach der Korngröße verschieden lichtempfindliche Fraktionen gewinnen. Es lassen sich weiterhin durch hydrothermale Synthese aus Zinksulfid reine Blenden herstellen, die dieselbe Lichtempfindlichkeit haben, wie hochgeglühte Präparate mit Wurtzitstruktur.Die Wirkung von Schwermetallzusätzen wird untersucht und auf Grund der von M. BODENSTEIN für die Silberhalogenide entwickelten Anschauungen diskutiert.Abschließend wird Über Versuche berichtet, die den Einfluß der Konzentrationsbedingungen bei der Fällung von reinem ZnS und von Lithopone zum Gegenstand haben.

    Notes: Abstract Background: Photodynamic therapy (PDT) might be of clinical value for patients with breast cancer with local recurrences or metastasis. However, there is a need for improved photosensitizers that are effective in combination with laser light and have few, if any, side-effects. We evaluated in vitro the effectiveness of a second generation photosensitizer by testing the influence of laser light on cell cultures of a human breast carcinoma cell line, incubated with meta-tetrahydroxy-phenylchlorin (m-THPC) (=Temoporfin®).Experimental design: Five thousand MCF-7 cells were plated in 96-well plates. Forty-eight hours before laser treatment, the cells were plated to achieve a monolayer configuration. Twenty-four hours after plating, they were incubated with m-THPC. On day 6 after treatment with m-THPC we lysed the cells to extract the intracellular ATP that correlates with the number of living cells. The ATP-CVA was used to assess the cytotoxicity of the tested photosensitizer m-THPC at various concentrations and the relevant laser light alone prior to their combination after six days of culture.Results: We found a dose-response for m-THPC alone ranging from 2 to 16 μg/ml. The calculated inhibition concentration to produce 50% cell kill (IC50) was 4.55 μg/ml. We also observed a very low cytotoxicity for laser irradiation alone but a very strong cell kill for the combination of m-THPC together with laser light.Conclusions: PDT gave almost total cell kill at m-THPC concentrations that are not toxic in vitro.

    Notes: Abstract Inactivation of tumor suppressor genes is thought to be a critical step in tumorigenesis. TheDCC (deleted in colorectal carcinoma) gene, located on the long arm of chromosome 18, has been shown to be frequently deleted in colorectal tumors. To investigate the involvement of allelic deletions on chromosome 18q in breast cancer tumorigenesis we analyzed 28 primary breast tumors and 28 colorectal, tumors (24 carcinomas, 4 adenomas) with four different polymorphic DNA markers detecting RFLPs on chromosome 18q. In breast cancer we found loss of heterozygosity (LOH) in 4 of 27 (15%) informative cases whereas 15 of 25 (60%) colorectal tumors showed allelic deletions. In all cases of allelic loss theDCC locus or its proximal vicinity (locus SSAV1) were involved. LOH on chromosome 18q occurs both in breast and colorectal cancer, yet the frequency of these deletions in breast tumors is lower than in colorectal tumors. Moreover, in breast cancer these mutations were only detected in large and undifferentiated tumors.

    Notes: A new enantioselective (80% ee) synthesis of (R)-sulforaphane and its epimer (S)-sulforaphane is described, which makes use of the pseudo-tetrahedral complex fragment [CpRu-(CHIRAPHOS)]+ as a chiral auxiliary. Reaction of the chloride complexes [CpRu(L-L)Cl] [L-L = 1,-bis(diphenylphosphino)ethane (dppe), (2S,3S)-and (2R,3R)-bis(diphenylphosphino)-butane ((S,S)- and (R, R)-CHIRAPHOS, respectively)] with phthalimidobutyl methyl sulfide gives the thioether complexes [CpRu(L-L)(MeSC4H8NPhth)]PF6. Oxygen transfer from dimethyldioxirane (DMD) produces the corresponding sulfoxide complexes in high yield and high diastereoselectivity. Cleavage of the phthaloyl group with aqueous hydrazine and subsequent reaction with thiophosgene yields the sulforaphane complexes [CpRu(L-L)(MeS(O)C4H8NCS)]PF6. Treatment of these with sodium iodide finally liberates the sulforaphane without noticeable racemization.

    Notes: To simulate production-scale conditions of gluconic acid fermentation by Gluconobacter oxydans, different experimental setups are presented in this study. From the determination of the time constants of a production-scale reactor, it can be concluded that mixing and oxygen transfer are the rate-limiting mechanisms. This results in oxygen concentration gradients which were simulated in a one-compartment reactor in which the oxygen concentration was fluctuated by a fluctuated gassing with air and nitrogen. It could be concluded that only very long periods of absence of oxygen (ca. 180 s) results in lower specific oxygen uptake rates by Gluconobacter oxydans. From scale-down studies carried out in a two-compartment system to simulate a production-scale reactor more accurately, it could be concluded that not only the residence time in the aerated part of the system is important, but the liquid flow in between the different parts of the reactor is also an essential parameter. It could also be concluded that the microorganisms are not influenced negatively by the fluctuated oxygen concentrations with respect to their maximal oxidation capacity. The two-compartment system can also be used for optimization experiments in which the “aerated” compartment was gassed with pure oxygen. From these experiments it was concluded that also a short residence of the cells at high oxygen concentrations diminished the growth and product formation rates. These experiments show the necessity of the scale-down experiments if optimization is carried out. The two-compartment system presented in this study is a very attractive tool for reliable scale-down experiments.

    Notes: -The stereoselective preparation of 1-hydroxycyclopentanecarboxylic acid derivatives is reported. The key reaction is either a radical cyclization or a radical annulation mediated by the transfer of a phenylseleno group. The radical precursors are easily synthesized by two consecutive enolate alkylations. Excellent stereochemical control for the quaternary C(1) stereogenic center has been achieved.
