    Description / Table of Contents: Summary First experience with continuous pH measurement is reported. It was possible to apply well calibrated electrodes to the fetal scalp in 22 cases. Of these 22 cases eight must be regarded as failures because of unstable values. Of the remaining 14 measurements, seven were of good (or acceptable) quality and seven of poor quality. The size of the electrodes and the excessive depth of penetration are probable reasons for these failures. An important condition for obtaining reliable pH values is a perpendicular position of the electrode in the fetal scalp. An indicator for correct position of the electrode appears to be the irregularity in the recording of the pH curve. Experience so far indicates that the pH electrode in its present for is not suitable for routine use.
    Notes: Zusammenfassung Es wird über die ersten Erfahrungen mit der kontinuierlich messenden Gewebe-pH-Elektrode berichtet. 22mal gelang es, gut geeichte Elektroden an den fetalen Kopf anzulegen. Von diesen müssen acht Fälle wegen nicht stabiler Meßwerte als Versager betrachtet werden. Von den verbleibenden 14 Messungen waren sieben von guter bzw. akzeptabler Qualität, die übrigen sieben von schlechter Qualität. Als Ursache für die Versager wird einmal die Größe der Elektrode und zum anderen die zu große Eindringtiefe angesehen. Eine wichtige Voraussetzung, glaubhafte pH-Werte zu erhalten, ist die senkrechte Position der Elektrode in der fetalen Kopfschwarte. Ein Zeichen für die korrekte Lage der Elektrodenspitze scheint die Irregularität in der Aufzeichnung der pH-Kurve zu sein. Aufgrund der bisherigen Erfahrungen scheint uns die Gewebe-pH-Elektrode in ihrer augenblicklichen Ausführung noch nicht zum routinemäßigen Einsatz für jedermann geeignet zu sein.

    Notes: Summary A new quantitative analytical method for the determination of phospholipids in amniotic fluid by high performance liquid chromatography (HPLC) is described. In addition to the main compounds, phosphatidylcholine (lecithin) and sphingomyelin, the so-called minor phospholipids, phosphatidylglycerol, phosphatidylinositol and phosphatidylethanolamine can also be determined. Separation is achieved using a guard-column of Lichrosorb Si 60 and an analytical column of Lichrosorb DIOL. Acetonitrile/water is used as mobile phase at an elevated temperature. By determining the recovery rates, the within-run and the between-run precision, it was shown that sufficient accuracy and precision could be achieved for all the parameters examined. The method is highly sensitive, the detection limit for sphingomyelin is 0.2 μg and 0.1 μg for all the other components. A single determination of 5 phospholipids in an amniotic fluid sample takes about two hours. By performing simultaneous extractions it is possible to analyse 5 samples per day.
