    Notes: Summary Proteins of membranes and cytosols were extracted from the livers and brains of mice (inbred strain DBA/6J) and rats (inbred strain DA/Han) and separated by two-dimensional electrophoresis (2-DE). The 2-DE patterns were compared with regard to qualitative (spot position) and quantitative (spot intensity) characteristics of the proteins of these two species. The following results were obtained: (1) Brain had more (higher percentage) conservative proteins (proteins found in both mice and rats) than liver; (2) plasma membranes had more conservative proteins than the cytosols; (3) organ-unspecific proteins contained more conservative proteins than relatively organ-specific proteins; (4) the pattern of distribution of genetic variability among different classes of proteins represented by findings 1–3 was the same for the qualitative and quantative characteristics of the proteins; and (5) some observations indicated that quantitative variability occurred more frequently among proteins than did qualitative variability. Our conclusion is that regulatory sequences in the DNA (regulatory genes) are subjected to functional constraints that differ in strength among different classes of proteins by the same ratios as the constraints acting on the structural genes. The overall effect of the selective pressure is, however, less stringent for regulatory genes than for structural genes. The results obtained here by comparing two different species are very similar to previous results we obtained by studying different subspecies (inbred strains of the mouse). From this finding arises a new concept: the study of molecular evolution on the basis of different classes of proteins. Our results were compared with data from the literature that were obtained in part from studies on cultured cells. The comparison suggested that cultured cells have lost their tissue-specific proteins, and so generate predominantly extremely conservative proteins.

    Description / Table of Contents: Summary We report a rare case of pulmonary nodular amyloidosis featuring all typical morphologic alterations associated with this disease; multiple amyloidomas, calcification and cavernous transformation were present. Definite diagnosis was established histologically with CT-guided biopsy. The different types of pulmonary amyloidosis and their radiological appearances are described and discussed.
    Notes: Zusammenfassung Wir berichten über einen seltenen Fall einer pulmonalen nodulären Amyloidose mit allen typischen Merkmalen dieser Erkrankung bei gleichzeitigem Vorliegen von multiplen Amyloidomen mit Verkalkungen und zentralen Einschmelzungen. Die definitive Diagnose wurde histologisch mit einer CT-gesteuerten Biopsie gesichert. Die verschiedenen Formen der pulmonalen Amyloidose und deren radiologische Erscheinungsbilder werden beschrieben und diskutiert.

    Notes: Abstract Mouse brain proteins were separated by two-dimensional electrophoresis (2-DE). The proteins of a section of the 2-DE pattern were blotted onto hydrophobic membranes and 43 of them were excised and hydrolyzed by liquid-phase hydrolysis. The amino acid composition of these proteins was determined by orthophthaldialdehyde precolumn derivatization and compared with the compositions of known proteins stored in the NBRF sequence database. An identification program named ASA was developed for this purpose. The ASA program includes correction and weighting factors, data reduction by molecular weight windows, and exclusion or inclusion of certain organisms as desired. As a control, eight test proteins and five well-known proteins from mouse brain, all separated by 2-DE, were correctly identified by the program. Out of the 43 brain proteins selected, 19 were identified with high confidence.

    Description / Table of Contents: Zusammenfassung Die „stadiengerechte Therapie” der akuten Pankreatitis, die jedem Schweregrad der Erkrankung angemessen ist, besteht neben der konservativen Basistherapie und Intensivmedizin im richtigen Einsatz operativer Maßnahmen. Voraussetzung dafür ist die frühe Diagnose und Beurteilung des Schweregrades der akuten Pankreatitis. Neben klinischen und klinisch-chemischen Befunden und dem Verlauf unter konservativer Therapie ermöglichen die Sonographie und die Computer-Tomographie eine bessere Beurteilung der jeweiligen Verlaufsform. Die für die Klinik der Erkrankung so wichtigen morphologischen Veränderungen der Bauchspeicheldrüse und ihrer Umgebung werden durch diese neuartigen Untersuchungsmethoden transparenter. Wäh rend bei der leichten Pankreatitis die Sonographie als erstes morphologisches Untersuchungsverfahren eingesetzt wird, besteht bei den schweren Verlaufsformen eine klassische Indikation zur Computer-Tomographie. Durch Einsatz von Sonographie und Computer-Tomographie läßt sich die Indikation zur frühzeitigen oder verzögerten Operation der hämorrhagisch-nekrotisierenden Pankreatitis, die bisher nach klinischen Kriterien erfolgte, in den meisten Fällen exakter stellen.
    Notes: Summary Adequate stage-depending therapy of acute pancreatitis includes basic conservative treatment, intensive care measurements, and operative interventions depending on the grade of severity. Prerequisites are early diagnosis and accurate clinical assessment of the stage of severity. Beside clinical and laboratory findings, as well as the development of acute pancreatitis under conservative treatment, sonography and computed tomography allow a better prediction of the underlying morphological changes, thus leading to an exact staging of the patient's individual situation. Sonography is regarded a screening procedure of high accuracy in mild forms of acute pancreatitis. Computed tomography is the method of choice in all severe forms of this disease. The indication for immediate or delayed operative treatment of hemorrhagic necrotising pancreatitis, heretofore depending on clinical findings solely, is supported by these new-invasive diagnostic modalities.

    Description / Table of Contents: Zusammenfassung Seit 1981 wurden 109 Patienten mit Morbus Crohn und 31 Patienten mit Colitis ulcerosa prospektiv computertomographisch untersucht. Beim Nachweis colitis- und crohntypischer, extramucöser Befunde wie Fisteln, Abscesse, Darmwandverdickung und Mesenterialveränderungen war die CT der herkömmlichen radiologischen Diagnostik überlegen. Der hochfloriden Colitis ulcerosa und dem Colon-Crohn konnten computertomographisch typische Wandschichtungen zugeordnet werden, die in 4 von 6 unklaren Fällen mit reinem Colonbefall zur richtigen Diagnose führten. Der Stellenwert der CT in der präoperativen Abklärung und der Differentialdiagnose unklarer Colitiden wird unterstrichen.
    Notes: Summary Since 1981, 109 patients with Crohn's disease and 31 patients with ulcerative colitis have been investigated prospectively by computed tomography (CT). In the diagnosis of fistulas, abscesses, thickening of the bowel wall, and mesenteric pathology, CT was more sensitive than the usual radiological procedure. Pathognomonic layering of the bowel wall was demonstrated by CT in higly active ulcerative colitis and Crohn's disease of the colon. In 4 of 6 obscure cases of colitis, a clear distinction between Crohn's disease and ulcerative colitis was possible. The value of CT in preoperative assessment and differential diagnosis is to be emphasized.

    Notes: The two-dimensional electrophoresis (2-DE) technique developed by Klose in 1975 (Humangenetik 1975, 26, 211-234), independently of the technique developed by O'Farrell (J. Biol. Chem. 1975, 250, 4007-4021), has been revised in our laboratory and an updated protocol is presented. This protocol is the result of our experience in using this method since its introduction. Many modifications and suggestions found in the literature were also tested and then integrated into our original method if advantageous. Gel and buffer composition, size of gels, use of stacking gels or not, necessity of isoelectric focusing (IEF) gel incubation, freezing of IEF gels or immediate use, carrier ampholytes versus Immobilines, regulation of electric current, conditions for staining and drying the gels - these and other problems were the subject of our concern. Among the technical details and special equipment which constitute our 2-DE method presented here, a few features are of particular significance: (i) sample loading onto the acid side of the IEF gel with the result that both acidic and basic proteins are well resolved in the same gel; (ii) use of large (46 × 30 cm) gels to achieve high resolution, but without the need of unusually large, flat gel equipment; (iii) preparation of ready-made gel solutions which can be stored frozen, a prerequisite, among others, for high reproducibility. Using the 2-DE method described we demonstrate that protein patterns revealing more than 10 000 polypeptide spots can be obtained from mouse tissues. This is by far the highest resolution so far reported in the literature for 2-DE of complex protein mixtures. The 2-DE patterns were of high quality with regard to spot shape and background. The reproducibility of the protein patterns is demonstrated and shown to be thoroughly satisfactory. An example is given to show how effectively 2-DE of high resolution and reproducibility can be used to study the genetic variability of proteins in an interspecific mouse backcross (Mus musculus × Mus spretus) established by the European Backcross Collaborative Group for mapping the mouse genome. We outline our opinion that the structural analysis of the human genome, currently pursued most intensively on a worldwide scale, should be accompanied by a functional analysis of the genome that starts from the proteins of the organism.
