    Notes: Summary The absorption of iron [59Fe-(FeSO4)] from jejunal loops was studied in rats after acute and subacute exposure to hexachlorobenzene (HCB). Female rats were given orally 100 mg HCB/kg body weight or fed a diet with 0, 500, 1,000, or 2,000 parts/106 HCB for 1 and 4 weeks. Male rats were fed on diets with 0 and 2,000 parts/106 HCB only. An increase in total urinary porphyrins and a decrease in the ratio of coproporphyrin to uroporphyrin in individual 24-h urine samples indicated different stages of porphyria in the rats at the time of determination of iron absorption. After acute oral administration of HCB, iron absorption decreased to about 70% of control values. After feeding the animals on HCB-containing diets, relative liver weight as well as total plasma protein increased depending on time and dose. The absorption of iron was not altered after 1 week of HCB exposure. After 4 weeks, the uptake of iron into the carcass decreased to less than 40% of control values, independently of dose and sex. The uptake of iron into the liver, expressed as percentage of the amount absorbed by the intestine, decreased significantly after 1 and 4 weeks of HCB feeding. No correlation could be observed between iron metabolism and urinary porphyrin excretion. The development of HCB-induced porphyria in rats is apparently not accelerated by an increased iron absorption.

    Description / Table of Contents: Zusammenfassung Bei Wistar-Ratten (FW 49) wurde durch 1 g/kg KG D-Galaktosamin eine toxische Leberschädigung (Galaktosamin-Hepatitis) ausgelöst und das Verhalten von Hämatokrit, Erythrocyten, Hb und aktiver Blutmenge (131J-Albumin-Verdünnung) gemessen. Dabei zeigte sich bei einem Anstieg des Hämatokrits auf 73,2% nach 48 Std (Kontrolle: 49,3%) eine Abnahme der „aktiven Blutmenge“ um 20% des Kontrollwertes. Diese Veränderungen haben sich nach 72 Std normalisiert. Die Hämokonzentration ist teilweise auf eine Anorexie der Tiere, im wesentlichen aber auf eine Plasmaextravasation, möglicherweise infolge eines toxischen Gefäßschadens, zurückzuführen. Bei der Interpretation von Blutspiegelmessung plasmatischer Substanzen muß diese Verkleinerung des Verteilungsraumes in Rechnung gestellt werden.
    Notes: Summary In Wistar-rats (FW 49) toxic liver damage was induced by application of 1.0 g/kg body-weight galactosamine (“galactosamine-hepatitis”). Hematocrit, erythrocytes pro mm3 blood and hemoglobin were determined as well as the circulating blood volume (131J-labelled albumin-dilution technique). 48 hours after application of galactosamine the hematocrit had increased from 49.3% to 73.2% while circulating blood volume had decreased from 5.61 ml/100 g to 4.49 ml/100 g (=20%). 72 hours after galactosamine application these parameters were normalized. Although the hemoconcentration was partially induced by an anorexia an extravasation of the plasma—probably due to toxic damage of the blood vessels—was the main cause of the decrease of blood volume. From these results it is concluded that the plasma levels of various substances should always be interpreted in connection with changes in blood volume.

An Hand dieses Falles wird die Bedeutung androgener Hormone für die Genese von Epithelveränderungen am weiblichen Kehlkopf unterstrichen und die Anwendung antiandrogener Substanzen als neuartige therapeutische Möglichkeit diskutiert.
    Notes: Summary Case-report about a 56 year old white female patient with pachydermia laryngis and hirsutism, successfully treated with cyproterone-acetate, a testosterone antagonist. The significance of androgenic hormones for the development of epithelial changes in the female larynx is emphasized. The administration of antiandrogenic substances as a new therapeutic measure is discussed.

    Description / Table of Contents: Summary First clinical experiences with the anti-androgenic drug cyproteroneacetate in treatment of pachydermia laryngis are reported in eleven male and female patients. This endocrine therapy led to complete healing in four and to marked improvement in six patients. One did not respond. Evaluation of the endocrine state of these patients showed imbalance in the androgen/estrogen equilibrium. As the larynx constitutes a secondary sex characteristic and an endocrine target organ, the significance of endocrine factors for pathogenesis and therapy of epithelial changes on the vocal cords is discussed.
    Notes: Zusammenfassung Es wird über erste klinische Erfahrungen in der Behandlung der Pachydermia laryngis mit dem Testosteronantagonisten Cyproteronazetat an elf Patienten beiderlei Geschlechts berichtet. Diese neuartige Therapie führte in vier Fällen zu vollständiger Heilung, bei sechs Patienten zu wesentlicher Besserung und blieb in einem Fall erfolglos! Hormonbestimmungen deckten Störungen im Androgen/Östrogen-Gleichgewicht auf. Die Bedeutung endokriner Faktoren für die Pathogenese und die Therapie von Epithelveränderungen im Larynx als einem sekundären Geschlechtsmerkmal wird diskutiert.
