    Notizen: Summary The immunogold technique was applied to detect cardiodilatin-immunoreactivity (CDD-IR) in human heart. Using antibodies directed against C-terminal regions of CDD, we could identify colloidal gold label in specific atrial granules of human heart. The granules in all areas of the myoendocrine cells were homogeneously labelled and all right atrial biopsy specimen of patients with different heart diseases reacted positively and had the same distribution of label. The Golgi-apparatus and ergastoplasma exhibited and extremely weak CDD-IR. The results indicate a predominant storage of CDD in specific granules.

    Notizen: Summary The placental labyrinth of the chorioallantoic placenta of the rat was studied with transmission and scanning electron microscopy. Full-term placentas were investigated after perfusion fixation from the maternal and fetal circulation, including thin-sectioning and freeze-fracturing. The labyrinthal three-dimensional structure was found not to be trabecular but spongious lamellar. We propose a division of the lamellae into three groups—the first, second, and third order. The trophoblastic layers are described in detail, showing a cellular layer I and two syncytial layers II and III. Layer I is found to be fenestrated and highly permeable. Layers II and III are connected by extended gap junction areas. It is suggested that the gap junctions function as a molecular sieve and represent the limiting barrier in diaplacental transport of the choriollantoic placenta. Fetal capillaries are fenestrated and endothelial cells are connected by tight junctions. The functional significance of the morphologic findings is discussed.

    Notizen: Summary The carotid sinus of the guinea pig was analysed immunohistochemically for the occurrence of neuropeptides. Immunoreactivity (IR) for neurotensin (NT) and substance P (SP) is distributed in two different populations of nerve endings and varicosities. NT-IR fibers penetrate deeply into the tunica media of the elastic segment of the carotid sinus and form the large, branched lanceolate nerve terminals. Electron-microscopic investigations have revealed that the NT-IR varicosities correspond to the large afferent baroreceptor endings containing abundant mitochondria. SP-IR fibers are located mainly at the mediaadventitial border. They seem to be correlated to dense-core, vesiclecontaining varicosities identified in the electron microscope. Therefore, these fibers may constitute afferent and efferent perivascular plexus regulating the vascular tone of the carotid sinus wall.

    Notizen: Summary The myoendocrine cells of the heart of Hypostomus cordovae (Günther 1880), a teleost fish from South America, were investigated by electron microscopy and immunohistochemistry. By applying antibodies raised against synthetic cardiodilatin 99–126 (CDD/ ANP 99–126), a specific labeling of this hormone was found in the heart of this fish, mainly in myoendocrine cells of atrial trabeculae, where specific secretory granules are stored. The distribution of secretory granules exhibited striking seasonal variations. In winter there were fewer differentiated myoendocrine cells, which were easily recognized by the presence of specific secretory granules, most of which occur clustered in perinuclear areas of the cells. By contrast, in summer the majority of the myocardic cells of the atrium are active endocrine cells. They contain abundant secretory granules widely scattered in the cytoplasm, many of them polarized toward the subendocardial aspect of the cell. The secretory granules can be easily differentiated from the Weibel-Palade granules of endothelial cells, the shape, size and content of which were typical at electron-microscopic level. In addition, these endothelial granules did not display CDD immunoreactivity. The presence of cardiodilatin in a fish such as Hypostomus cordovae further supports the view that cardiac hormones are present in many Vertebrates and may preserve analogous roles such as those reported in other species throughout the group.

    Notizen: Summary Using various region specific antibodies raised against partial sequences of synthetic cardiodilatin (CDD) we detected immunoreactive neurons with their perikarya in the nucleus periventricularis of Tupaia belangeri. The fibers could be traced laterally directed towards the amygdaloid complex and some varicosities were also observed in the lateral parts of the nucleus periventricularis. It is postulated that brain CDD represents a new neuropeptide and that these CDD-IR neurons are involved in specific functions related to the modulation of the cardiovascular centers.

    Notizen: Summary Immunohistochemical investigations with different antisera against cardiodilatin 99–126 or alpha atrial natriuretic polypeptide revealed the presence of cardiac hormones not only in the atria of rats but also in strands of myoendocrine cells located in subendocardial regions of the ventricular septum. The localization of CDD-IR (cardiodilatin immunoreactivity) in the ventricle is associated with the location of the conduction system in the rat. The significance of the morphological relationship between cardiodilatin and the conduction system of the rat heart is discussed.

Bei bestimmten Tieren finden sich in den Intermediärzellen „matrixreiche“ Riesenmitochondrien. Diese Riesenmitochondrien scheinen aus normalen Mitochondrien hervorzugehen und enthalten verschieden geformte Einschlüsse. Andere Organe und Zellen bei Maulwürfen mit Riesenmitochondrien in den Intermediärzellen des Harnblasenepithels weisen normale Mitochondrien auf. Es handelt sich demnach hier um eine sog. „Mitochondriose der Intermediärzellen“. Bedeutung, Bildung und Abbau der Riesenmitochondrien werden diskutiert.
    Notizen: Summary Giant mitochondria containing an enormous matrix volume were found in the intermediary cells of the urinary bladder transitional epithelium of the mole. Regularly shaped mitochondria develop club-like protrusions forming giant mitochondria that contain several types of inclusions. Various other tissues were examined, but we could state that giant mitochondria exist only in the intermediary cells of the urinary bladder epithelium. Thus a peculiar “intermediary cell mitochondriosis” is described in this paper, not regularly occurring in the mole. The functional significance of the giant mitochondria is discussed.

Im Stadium der Nebenniereninsuffizienz, sechs Tage nach der Operation, wurde quantitativ morphometrisch eine Abnahme der Volumendichte der Sekretgranula (von 32% auf 16%) festgestellt. Golgi-Apparat und Ergastoplasma proliferieren stark, wobei insbesondere die erweiterten Cisternen des Ergastoplasmas auffallen.
    Notizen: Summary The response of the Juxtaglomerular apparatus to bilateral adrenalectomy was studied in rats. After the sixth postoperative day, symptoms of severe adrenal insufficiency had developed. In the Juxtaglomerular cells, the volume density of the secretory granules decreased from 32% to 16%. The Golgi apparatus and rough endoplasmic reticulum underwent considerable proliferation, the latter showing strikingly enlarged cisternae.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Die perinatale Entwicklung des juxtaglomerulären Apparates, insbesondere das Verhalten der epitheloiden Zellen wurde an 18 trächtigen Ratten and 65 Embryonen bzw. Jungtieren elektronenmikroskopisch untersucht. Parallel dazu wurde die Reninkonzentration bzw. der Reningehalt bestimmt. Spezifische Sekretgranula finden sich erstmals am 18. Tag der Fetalzeit, und zwar in den epitheloiden Zellen des juxtaglomerulären Apparates juxtamedullärer Nephrone. Die Granula machen einen Struktur- und wahrscheinlich auch Funktionswandel durch, der eine Einteilung in drei verschiedene Granulatypen nahelegt. Diesen drei Typen spezifischer (reninhaltiger) Sekretgranula werden die Lysosomen als unspezifische Einschlüsse der epitheloiden Zellen gegenübergestellt. Im einzelnen kommt es während der perinatalen Phase in den fetalen Epitheloidzellen zuerst, d.h. pränatal, zu einer Hypertrophie des rauhwandigen endoplasmatischen Retikulums und des Golgi-Apparates. Hierin sehen die Autoren einen Hinweis auf eine erhöhte Proteinsynthese mit entsprechender Zunahme der Transport- und Konzentrationsvorgänge. Nach der Geburt findet sich dann auch eine deutliche Zunahme der Sekretgranula vom Typ III, die als reife Reningranula (mature granules) angesehen werden. Die bisher noch nicht beschriebenen kleinen Sekretgranula vom Typ I dagegen kommen in weniger entwickelten Epitheloidzellen häufiger vor. Bestimmungen der Reninverteilung zwischen Zytoplasma und den durch Differential-zentrifugation gewonnenen Granula-Fraktionen ergeben auffallende Parallelen zu den morphologischen Befunden. Diese sprechen im allgemeinen dafür, daß “freies Renin” im Ergasto-plasma und “gebundenes Renin” in den Sekretgranula vorliegt. Mit zunehmendem Alter scheint es außerdem — parallel zur relativen Abnahme des Ergastoplasmas und Zunahme der reifen Reningranula — zu einer Verminderung des extragranulären zugunsten des granulär gebundenen Renins zu kommen.
    Notizen: Summary The perinatal development of the Juxtaglomerular apparatus (JGA), with particular reference to the epithelioid cells, was studied. Electron microscopic studies were carried out on 18 gravid rats and 65 embryos with corresponding determinations of renin concentration and the renin contents by bio-assay. Specific secretory granules of the epithelioid cells of the JGA of juxtamedullary nephrons are first observed on the 18th day of gestation. The classification into three types of secretory granules suggests that they undergo a structural and probably also a functional transformation. All three types of secretory granule may be regarded as specific, renin-containing, cytoplasmic organelles. During the last days of gestation, the rough-surfaced endoplasmic reticulum and the Golgi apparatus become hypertrophied in epithelioid cells, indicating an increased protein synthesis as well as transport and concentration processes. After birth, there is a definite increase in the number of Type III secretory granules-which may be regarded as mature granules. The small Type I secretory granulesnot previously described-are found more frequently in the immature epithelioid cells. Determinations of the renin distribution in the plasma and in fractions of cytoplasmic constituents (total homogenate, secretory granules) prepared by differential centrifugation showed striking congruence with the morphological findings. It is generally assumed that “soluble renin” occurs in the endoplasmic reticulum and “bound renin” in secretory granules. A shift from extragranular to granular (bound) renin, occurs with increasing age, having its morphological equivalent in the relative decrease of the endoplasmic reticulum and increase in mature secretory granules.
