Zusammenfassung Künstliche Fibringerinnsel, die Ratten und Kaninchen in die Bauchwand implantiert wurden, werden unter Mitwirkung von Zellen (Granulocyten, Histiocyten) abgebaut und durch Bindegewebe ersetzt. Mit zunehmendem Alter der Fibrinablagerung nimmt die Menge der Granulocyten ab. Gleichzeitig vermindert sich die Menge des in das Gerinnsel eingeströmten Kaninchen- bzw. Rattenplasminogens. Der Gehalt an im Implantat von vornherein enthaltenem Plasminogen, in unserem Ansatz Rinderplasminogen, nahm innerhalb 21 Tagen nicht meßbar ab. Die Struktur des implantierten Fibringerinnsels hat weder auf die celluläre Reaktion des Organismus noch auf die fermentative Löslichkeit des Fibrins einen Einfluß. Die celluläre Reaktion läßt sich durch Serotonin und EACA beeinflussen. Serotonin verstärkt gering die granulocytäre Reaktion, stimuliert aber besonders die Vermehrung von Fibroblasten. Dies deutet neben den Blutgefäßwirkungen auf eine direkte Einwirkung des Serotonins auf Fibroblasten hin. EACA vermindert im wesentlichen die Menge der am Organisationsprozeß beteiligten Granulocyten. Dieser Mechanismus kann mit einer Hemmung der latenten Fibrinolyse in der Blutgefäßbahn erklärt werden, wodurch der Austritt von Granulocyten erschwert werden dürfte. Die Tatsache, daß das Fibrin auch 21 Tage nach Implantation noch reichlich aktivierbares Plasminogen enthält, erklärt zwanglos die Erfolge bei der Behandlung chronisch arterieller Verschlüsse durch Streptokinase.

Summary Artificially produced fibrin clots implanted into rats and rabbits are digested with the aid of granulocytes and histiocytes and later on replaced by connective tissue. As the fibrin clots age the number of granulocytes is diminuished. At the same time the quantity of rabbit plasminogen or rat plasminogen flowed in the clots is decreased. The amount of plasminogen as contaminant in the implanted fibrin did not decrease to a measurable degree within 21 days. The structure of the implanted clot neither influence the cellular reactions of the organism nor the clot lysis by means of enzymes. The cellular reaction can be influenced by serotonin and EACA. Serotonin increases granulocytic reaction only to a minimal extent, for the most part it stimulates the generation of fibroblasts. Besides the vasoactive effect of serotonin its direct influence on fibroblasts is evident. EACA diminuishes the number of granulocytes involved in the organisation of the clots. This mechanism can be explained by inhibition of latent fibrinolysis in the microcirculation, wich thus aggravates the extravasation of granulocytes. The fact, that fibrin is still rich in plasminogen after 21 days post implantation, may account for the successful treatment of chronic arterial occlusion by streptokinase therapy.
