    Description / Table of Contents: Summary The human fibrin in thrombi, in socalled placental infarcts and that of fibrinous inflammations was investigated for its enzymatic solubility. The morphologic progress of fibrinolysis was constantly observed in the phase contrast microscope. For comparison, the structure and composition of the fibrin material used, was investigated in slides by histochemical reactions. Fresh fibrin microscopically consisting of fine nets of fibrin, contains sufficient pro-activator and plasminogen, so that it can be dissolved by streptokinase. With increasing age the content of pro-activator and plasminogen becomes less. Therefore, this older fibrin that microscopically consists of thick fibres or homogenous plaques, can no longer be dissolved by streptokinase alone, but with activated plasmin, chymotrypsin and trypsin. The streptokinase that has been added therapeutically in vivo is most probably adsorbed to the fibrin fibres of thrombi.
    Notes: Zusammenfassung Menschliches Fibrin in Thromben, in sog. Placentarinfarkten und von fibrinösen Entzündungen wurde auf seine fermentative Löslichkeit untersucht und der morphologische Fibrinolyseablauf im Phasenkontrastmikroskop fortlaufend beobachtet. Vergleichend wurden an histologischen Schnitten und durch histochemische Reaktionen die Struktur und Zusammensetzung des verwendeten Fibrinmaterials untersucht. Frische Fibrinablagerungen, die mikroskopisch aus zarten Fibrinnetzen bestehen, enthalten in genügender Menge Proaktivator und aktivierbares Plasminogen, so daß ihre Auflösung mit Streptokinase gelingt. Mit der Alterung des Fibrins geht offensichtlich der Proaktivator und Plasminogengehalt verloren. Dieses ältere Fibrin, das mikroskopisch aus groben Balken oder homogenen Plaques besteht, ist daher mit Streptokinase allein nicht mehr, wohl aber durch aktiviertes Plasmin, Chymotrypsin und Trypsin aufzulösen. Therapeutisch zugeführte Streptokinase wird mit großer Wahrscheinlichkeit an die Fibrinfasern von Thromben adsorbiert.

    Description / Table of Contents: Zusammenfassung 21 Kaninchen erhielten 1 g Galaktosamin/kg Körpergewicht intravenös injiziert. Das Gerinnungs-system (Faktor II, X, V, VIII, Fibrinogen und Thrombocyten) wurde vor Galaktosamingabe und kurz vor dem Tod der Tiere untersucht. Nur 6 Kaninchen lebten 48 Std nach der Galaktosamininjektion. Während der Galaktosaminhepatitis, die mit den Zeichen einer Lebernekrose verlief, zeigten der Prothrombinkomplex, die Faktoren V und VIII einen signifikanten Abfall. Somit ist eine Bildungsstörung der Gerinnungsfaktoren nachgewiesen, eine Umsatzstörung läßt sich jedoch nicht ausschließen.
    Notes: Summary 21 rabbits were treated with 1 g galactosamine/kg. The coagulation system (factors II, X, V, VIII, fibrinogen, blood platelets) was investigated before injection and shortly before death. 6 rabbits still survived 48 hours after galactosamine injection. The results showed a significant decrease of the prothrombin complex, and of factors V and VIII during hepatitis with liver necrosis. There seems to be a disturbance in the production of coagulation factors; a consumption coagulopathy is not excluded.

    Description / Table of Contents: Zusammenfassung Acetylsalicyl-lysin wurde in den Dosierungen 0,9 g/48 h, 1,8 g/24 h und 3,6 g/24 h in drei nacheinander folgenden Studien zur Thromboseprophylaxe bei Hüftgelenksoperationen anhand des Radio-Fibrinogen-Tests untersucht. Mit den drei AS-lysin-Dosierungen wurde keine Verminderung von Thrombosen (TVT) gefunden. Die Kollagen-induzierte Thrombozytenaggregation war in allen drei Gruppen signifikant gesenkt. Unter den niedrigen AS-lysin-Dosen war die Aggregation beim Auftreten einer TVT weniger stark gehemmt als beim Ausbleiben einer TVT. Bei der 3,6 g-Dosis war die Aggregation maximal ohne signifikante Unterschiede vom Thrombosevorkommen gehemmt. Die Befunde ergeben keinen therapeutischen Bereich für die Acetylsalicylsäure zur Verhütung venöser Thrombosen.
    Notes: Summary In three consecutive studies, acetylsalicyliclysine using the dosages 0.9 g/48 h, 1.8 g/24 h and 3.6 g/24 h, was examined in order to see its effect in preventing deep vein thrombosis (DVT) after elective hip joint surgery. DVT was diagnosed by the 125-I-fibrinogen-test. No reduction of postoperative DVT was found under any of the administered AS-lysine dosages. The collagen-induced platelet aggregation was significantly decreased in all three groups. However, under the low AS-lysine dosages, aggregation was less inhibited with the incidence of DVT than without DVT. In the group with the 3.6 g-dosage, aggregation was maximally inhibited, without any significant difference related to the occurrence of DVT. The results reveal no therapeutic range of AS-lysine to prevent postoperative venous thrombosis.
