    Notes: Summary Of several murine autoimmune models, MRL/Mp-lpr/lpr (MRL/l) mice are the most attractive from a dermatopathological point of view, because their skin lesions resemble the erythematous lesions of systemic lupus erythematosus (SLE). In order to clarify the pathogenesis of lupus dermatoses, histamine and its metabolizing activities in the skin of MRL/l mice were investigated. The specific activities of histamine-N-methyltransferase (HMT) in the skin of MRL/l mice were significantly lower than those in the skin of control mice i.e., MRL/Mp-+/+ (MRL/n), C57BL/6J, and BALB/c mice. In the dorsal lesional skin of MRL/l mice, HMT activities were markedly lower than those in normal abdominal skin. In addition, age-related analysis of HMT levels in the dorsal skin of MRL/l mice revealed that HMT activities reached their maximum at the age of 2 or 3 months and then decreased at 4 or 5 months when skin manifestation appeared: however, HMT activities in the abdominal skin increased almost linearly with age. There were no significant differences in histamine content in these mice, and diamine-oxidase activities were not detected in any skin specimens. From these results, it is suggested that impaired histamine metabolism is a particular biochemical feature of the skin of MRL/l mice.

    Notes: Summary The activities of two histamine-metabolizing enzymes, histamine-N-methyltransferase (HMT) and diamine oxidase (DAO), were examined in various types of experimentally induced cutaneous inflammations in guinea pigs. In intact guinea-pig skin, the specific activities of HMT and DAO were 24.8±1.7 pmol/min per milligram of protein or 0.930±0.097 pmol/min per milligram of the wet weight of skin specimen, and 6.0±0.7 pmol/min per milligram of protein or 0.189±0.011 pmol/min per milligram of the wet weight, respectively. Both enzyme activities were markedly reduced in skin lesions of the Arthus reaction (P〈0.005), while those in dinitro-chlorobenzene allergic dermatitis, croton-oil dermatitis, and the intact areas in Arthus-reaction-induced aminals were almost within the normal limits. The activity of HMT decreased linearly with time from the onset of the Arthus reaction, reaching about 20% of the control activity at 48 h; the activity of DAO decreased even from the early stages of the reaction, and this decrease continued throughout first 48 h of the reaction. These results suggest that impaired histamine metabolism in the skin lesions of the reaction plays a distinct role in the formation and development of the Arthus reaction.

    Notes: Summary We examined the dynamic changes of histamine metabolism and infiltrating cell populations in lesional sites of representative cutaneous delayed type hypersensitivity (DTH), including dinitrochlorobenzene (DNCB) allergic dermatitis, purified protein derivative of tuberculin (PPD) reaction, and keyhole limpet homocyanin (KLH)-induced cutaneous basophil hypersensitivity. The concentration of histamine increased with time in all DTH reactions examined, though the time course varied among these reactions. In DNCB allergic dermatitis, the maximum content of the amine at 3 days after the initiation was about three times that of the control baseline. In PPD reaction, the maximum content and the time course were almost similar to that in DNCB allergic dermatitis. However, in KLH-induced cutaneous basophil hypersensitivity the maximum content was about ten times that in DNCB allergic dermatitis or PPD reaction, and was observed earlier, on the 2nd day. There was no remarkable change in the activities of histamine-degrading enzymes in these reactions. There was little infiltration of mast cells, while time-dependent changes of the basophil infiltration were almost parallel those of the histamine concentration in all these reactions. Basophils in DNCB allergic dermatitis showed a piecemeal degranulation, while those in either the PPD reaction or KLH-induced cutaneous basophil hypersensitivity remained intact. These results clearly suggest that the increase of histamine concentration in cutaneous DTH depends on the number of basophils infiltrating the lesional sites, even if the regulatory mechanisms of the activation of the cells differ among the DTH reactions.

    Notes: Abstract Uraemic pruritus is poorly understood despite the high incidence among chronic renal failure (CRF) patients undergoing haemodialysis. Serum histamine levels have been shown to be elevated in CRF patients with itching, and ultraviolet B (UVB) therapy, even if applied to only part of the body surface, has been reported to be beneficial for the generalized relief of the pruritus. A local mechanism of UVB action is suggested by evidence that UVB radiation is able to suppress histamine release from mast cells. However, detailed systemic mechanism(s) remain obscure. Sera from patients with or without uraemic pruritus were incubated with purified rat peritoneal mast cells and the resulting histamine release was compared. A higher histamine release was obtained with sera from uraemic pruritus patients (44.60 ± 6.32%, n=9, P〈0.005) than with sera from patients without itching (19.71 ± 3.14%, n=5, P〉0.25) and with normal control sera (23.62 ± 7.14%, n=6). This increased histamine release was dose-dependently restored to spontaneous release levels in five of seven patients by pre-exposure of the sera to UVB in vitro. From these results, sera of CRF patients with uraemic pruritus were considered to contain some histamine releasing factor(s) which was depleted or diminished by UVB irradiation, suggesting a possible systemic mechanism of UVB action.
