    Notes: Abstract This paper gives an overview of our recent research activities on the lateral supramolecular order of a variety of native and man-made cellulosics considering respective results from the literature. Wide-angle X-ray scattering (WAXS) was the main investigation technique used. Lateral root mean squared lattice strains between 2 and 3% were determined for the materials investigated. Crystallite sizes obtained without considering lattice distortions usually do not deviate by much more than −10% from the real, i.e. fully corrected values. This means that it is sufficient to use the simple Scherrer equation for determining lateral crystallite sizes for most routine investigations of cellulosic materials. The possible superposition of WAXS peaks of the triclinic Iα and monoclinic Iβ lattice types, however, has to be considered in crystallite size determinations for Valonia cellulose. It could be shown that neglecting this fact can lead to crystallite sizes being about 20% below the true ones. Lateral crystallite dimaensions for native celluloses vary between 4nm (dissolving pulps) and 10-15 nm (Valonia). Except for bacterial cellulose, the WAXS crystallite sizes are distinctly smaller than the microfibril dimensions obtained from electron microscopy. The man-made fibres investigated showed lateral crystallite dimensions between 3 and 5nm. The importance of lateral crystallite dimensions for the properties of man-made fibres and for the alkalization process of native cellulose id demonstrated.

    Notes: Sorption of Penicillium cellulase onto cotton linters samples differing in physical structure and onto KLASON- lignin from spruce has been investigated by determining cellobiase, CMCL- and ‘linters-activity’ as well as protein content of a cellulase culture filtrate before and after contact with the sorbent at different temperature for different time intervals. CMCL- and cellobiase-activity were found to be reduced much less by the sorption process onto the cellulose samples than ‘linters-activity’ and protein content. Sorption increased in the order: untreated linters 〈 mercerized linters 〈 wet fibrillated (‘colloidal milled’) linters. With the KLASON-lignin also a considerable sorption of the cellulase system was observed. Results are discussed with respect to preferential sorption of enzyme components.

    Notes: By comparing different activity data of the buffered cellulase solution before and after contact with the substrate the interaction between Penicillium janthinellum cellulase and wheat straw, resp. its components (holocellulose and isolated lignin) has been investigated. The loss of activity due to sorption or denaturation has been found to differ widely between the different activity data and between the various substrates. A remarkable loss of enzyme activity was observed after contact with isolated straw lignin. The differences in activity decrease between the cellulose and the lignin moiety were found to be largent with the cellobiase activity.

    Notes: Using as substrates various water-soluble cellulose derivatives differing in DS and type of substituent, the efficiency of enzyme action in dependence of pH and the “CMC ase activity” of a Gliocladium spec. cellulase was investigated, stressing the significant role of effective charge density in connection with the results obtained with ionic derivatives of cellulose.     Notes: Starting from cellulose samples prepared from cotton lintes and differing in lattice type, crystallinity and fibrillar morphology, enzymatic hydrolysis of fibre cellulose has been studied employing complete enzyme systems from Trichoderma, Sporotrichum, Gliocladium and Penicillium as well as isolated endo- and exo-1,4-β-glucanases from Trichoderma reesei and Sporotorichum pulverulentum. The effect of hydrolysis was characterized by content of reducing sugars (RS) and of glucose in the hydrolyzate as well as by DP and X-ray diffraction pattern of the residues. With all the complete enzyme systems investigated about the same order of degradability was found with a series of substrates differing in physical structure. The hydrolysis effect of cellulase from S. pulverulentum proved to be sensitive to the gas atmosphere above the system (N2 or O2), probably due to the interaction of an O2-atmosphere with the activity of the cellubiose-oxydase existent in the system. Isolated endoglucanase from S. pulverulentum and T.reesei still led to a considerable formation of RS and glucose, a corrosion of the fibre surface and a significant descrease in DP. Influence of substrate physical structure was rather small with regard to RS, but still considerable with regard to residue-DP.The effect of isolated exoglucanases depends largely on the chemical structure of the cellobiohydrolase in question, as demonstrated with the two samples “CBH I” and “CBH II” from T. reesei. With CBH I, rather resembling endo-glucanase behaviour, a considerable formation of RS and a significant corrosion of the fibre surface has been observed. On the other hand, only negligibly small amounts of RS were formed by CBH II.Results are discussed with regard to the complex mechanism of cellulase action on fibrous cellulose and with regard to the relevance of different parameters of physical structure of cellulose in connection with enzymatic hydrolysis. A remarkable acceleration of the Cellulose III → Cellulose I lattice transition due to chain fragmentations in the presence of cellulase can be concluded the experiments.

    Notes: Starting from cotton linters cellulose modified in physical structure type of lattice, degree of order, state of swelling by different pretreatments, and from culture filtrates of Gliocladium spec., the initial stage of enzymatic hydrolysis of cellulose has been investigated. Especially with substrates of high degradability a considerable effect of stirring on rate of formation of soluble products was found. For linerization of yield-vs-time-curves (% residue resp. 7percnt; solubles as a criterion for yield), a 2-parameatric first order rate law was found to be suitable within a limited time interval, values of k1 were higher and for the accessible part of the substrate were lower in the initial stage of hydrolysis than in the later one. The MICHAELIS-MENTEN-constant kM has been determined for substrates of different physical structure after different times of reaction. Data found for kM indicated a stronger dependence of kM on reaction time than on physical structure of the substrate under conditions applied.

    Notes: To enhance the digestibility of wheat straw as an animal feedstock a partial delignification by action of 1percnt; aqueous alkali in combination with mechanical pre- and after-treatment steps is proposed. On the basis of in vitro digestibility by cellulase the influence of various process parameters on the effect obtained is summarized. Sheep feeding studies comparing original and chemomechanically pretreated straw confirm the suitability of the process-principle proposed for upgrading cereal straw. The practical feasibility of the process is considered with regard to animal nutrition and technology.

    Notes: Employing anionic and non-ionic cellulose ethers, differing in type of substituent and degree of substitution, as substrates, the pH-profile of enzyme activity and the parameter Km and Vmax of the MICHAELIS-MENTEN kinetics have been determined with Penicillium citrioviride cellulase in an homogeneous system. Within rather wide limits, a linear correlation was found between the DS of the substrate and Km or Vmax, respectively. Also the pH-profile was found to depend on DS mainly, being bimodal at higher DS. On the other hand, no significant difference was observed between anionic and non-ionic cellulose ethers of about the same DS with regard to the parameters determined. It is assumed, that substrate-enzyme interaction is governed mainly by the length of non-derivatized chain sequences mainly and not by Coulomb interactions.
