Notes: Summary The pharmacokinetics of two slow release theophylline preparations “Theo-Dur” (T) containing theophylline only and “Phyllocontin” (P) containing aminophylline have been compared in 12 patients with asthma. Each patient received both treatments in random order. The dose of treatment administered 12 hourly was increased or decreased to produce plasma theophylline concentrations of 10–20 mg/l at clinic visits normally 7 to 8 h after dosing. Pharmacokinetic studies were carried out after at least one week's treatment with this dose. After the first study day patients were crossed over to the second treatment at a dosage providing a similar amount of theophylline. They returned for a second study day after at least one week. Comparison of the dose corrected AUC, time to peak concentrations, within patient coefficients of variation (CV), number of concentration time points falling within 25% of Cmax and percentage fluctuations in plasma concentration showed no significant differences between the two preparations.

Notes: Summary Ten patients with moderate to severe Raynaud's syndrome were recruited into a four week randomised double blind crossover study to compare the efficacy of UK-38,485 50 mg, a new thromboxane synthetase inhibitor with that of placebo. With the doses used there was no significant difference between the two treatment periods in the number, severity and duration of ischaemic attacks, the mean hand temperatures, forearm and digital blood flow and red blood cell rheology.

Notes: Summary The effects of pentoxifylline on filterability of normal red blood cells (RBCs) and their adhesiveness to cultured endothelial cells were investigated. 1. In a balanced randomized and double blind trial, six healthy volunteers received 400 mg pentoxifylline or matching placebo 2 h before blood samples were taken. Filterability of RBCs of the subject while on pentoxifylline was significantly increased at 25 °C and 18 °C. 2. Lowering of the filteration temperature to 18 °C significantly decreased filterability of RBCs. 3. In vitro studies showed that 12 µg/ml pentoxifylline significantly increased RBC filterability and also partially prevented the effect of decreasing temperature on RBC filterability. 4. 12 µg/ml pentoxifylline significantly decreased the adherence of normal RBCs to cultured endothelial cells. Our results suggest that in addition to increasing filterability of RBCs, pentoxifylline also decreased the adherence of RBCs to endothelial cells and this may contribute to its therapeutic effect.

Notes: Summary Erythromycin kinetics were studied in 17 patients with end stage renal failure treated with maintenance haemodialysis and 9 normal volunteers to discover if dialysis patients needed a modified dose. The elimination half life in dialysis patients (on dialysis days) was similar to that reported in normal subjects. Only small amounts of drug appeared in the dialysate, no patient loosing more than 9 mg in one dialysis. Both patients and volunteers had similar plasma concentrations 8 h after the end of a 5-day course. Protein-binding did not change significantly during dialysis and was similar to that reported in normal subjects. We conclude that dialysis patients requiring 1.5 g of erythromycin stearate daily or less can be given normal doses.

Notes: Abstract In this paper, the Hilbert-space analogue of a result of Huang, that all the methods in the Huang class generate the same sequence of points when applied to a quadratic functional with exact linear searches, is established. The convergence of a class of direct prediction methods based on some work of Dixon is then proved, and these methods are then applied to some control problems. Their performance is found to be comparable with methods involving exact linear searches.

Notes: Abstract In this paper, the convergence of variable-metric methods without line searches (direct prediction methods) applied to quadratic functionals on a Hilbert space is established. The methods are then applied to certain control problems with both free endpoints and fixed endpoints. Computational results are reported and compared with earlier results. The methods discussed here are found to compare favorably with earlier methods involving line searches and with other direct prediction quasi-Newton methods.

Notes: Abstract This paper is concerned with some of the most powerful methods of minimizing functionals on Hilbert space. It is established that certain classes of these methods are equivalent and their convergence is proved for certain nonquadratic functionals on a Hilbert space. A computational study of these methods applied to a control problem is also included with particular reference to the equivalence of methods mentioned above.
