    Notizen: X-ray absorption near edge structure (XANES) spectra of hard amorphous a-Si–C–N thin films with various compositions were measured at the C and N K-edge using sample drain current and fluorescent modes. The C K-edge XANES spectra of a-Si–C–N contain a relatively large 1s→π* peak, indicating that a substantial percentage of carbon atoms in the a-Si–C–N films have sp2 or graphite-like bonding. Both the observed sp2 intensity and the Young's modulus decrease with an increase in the carbon content. For N K-edge XANES spectra of the a-Si–C–N films we find the emergence of a sharp peak near the threshold when the carbon content is larger than between 9% and 36%, which indicates that carbon and nitrogen atoms tend to form local graphitic carbon nitride. © 2001 American Institute of Physics.

    Notizen: Three types of oxygen precipitate morphologies at 1050 °C have been identified to be plate-type, pyramidal, and polyhedral. This study was carried out using a set of slices with the same initial oxygen concentration but varying carbon concentrations. The morphology is shown to depend on carbon concentration and heat treatment. It is demonstrated that increasing carbon concentration tends to change the morphology from the plate to polyhedra and enhance the nucleation rate at low temperatures.

    Notizen: The generation and propagation of threading dislocations in GaAs grown on Si was studied by transmission electron microscopy. This study was conducted by analyzing dislocation structures in GaAs at various growth stages. The dislocation structure changes as the GaAs grows from initial islands to a thick film. These changes are explained by the following physical processes: (1) nucleation of dislocations in GaAs islands, (2) generation and propagation of threading dislocations during coalescence of initial GaAs islands and their subsequent growth, and (3) bending of threading dislocations under misfit-strain force and their interactions. The second process results in threading dislocations extending from the interface to the surface in thin films. The third process causes an improvement in near-surface quality in thick films. The iplication of this study in confining threading dislocations to near the interface is discussed.

    Notizen: Oestradiol exerts negative- and positive-feedback actions on luteinizing hormone (LH) secretion by modulating gonadotropin-releasing hormone (GnRH) release. Furthermore, a chronic increase in circulating oestradiol in either young ovariectomized (OVX) rats, or in middle-aged persistent oestrous (PE) rats, causes a gradual attenuation of LH surges until the positive-feedback action of oestradiol disappears. Based on these findings, and on the equivocal evidence regarding a direct action of oestradiol on GnRH neurones, we tested the hypothesis that chronic oestradiol abolishes LH surges by decreasing the proportion of GnRH neurones containing oestrogen receptor (ER)α or β. Regularly cycling rats were ovariectomized, and half immediately received oestradiol. Three days, or 2 or 4 weeks later, rats were perfused at 18.00 h, and GnRH was colocalized with ERα or ERβ by immunocytochemistry. ERβ was expressed in 76% of GnRH neurones, whereas virtually no GnRH cells were immunopositive for ERα. The proportion of GnRH cells expressing ERα or β in OVX rats was not altered by oestradiol or time after OVX, and this was the case regardless of their medial to lateral, or rostral to caudal location. The results indicate that the mechanisms for the positive-feedback action of oestradiol, and the loss of LH surges in OVX rats after chronic oestradiol, are not mediated by changes in the proportion of oestrogen-receptor containing GnRH neurones.

    Notizen: : The quality of phytoestrogenic licorice was studied by using both chemical and biological fingerprints. A recombinant yeast strain that consists of an estrogen responsive element linked with a reporter gene (ADE2) and a transformed human estrogen receptor–containing plasmid was used for screening and evaluation of estrogenic activity in licorice. Several estrogen-like components in licorice were screened, and licoisoflavone B and formononetin were identified. Licorice extracted with 70% ethanol showed 5 different patterns of chemical fingerprints (LR-A, LR-E, LR-F, LR-H, LR-K), as identified by chromatographic analysis. Among these, LR-E exhibited the strongest estrogenic activity, whereas LR-A, LR-F, and LR-H were in the middle, and LR-K had the weakest activity.

    Notizen: A cDNA fragment of 302 bp coding for the mature region of the gonadotropin α-subunit of the yellowfin porgy (Acanthopagrus latus Houttuyn 1782) was isolated by polymerase chain reaction (PCR). This PCR product was engineered into the Escherichia coli expression vector pRE1 which is based on the lambda PL/cII transcription-translation system. The resultant recombinant construct pREYP1 was introduced into E. coli MZ-1 and resulted in the expression of the recombinant α-polypeptide of gonadotropin of 14 kilodaltons (kDa) in the cell extracts. The intensity of the 14 kDa protein band on the SDS-polyacrylamide gel increased in relation to the length of the induction time periods. This E. coli-produced 14 kDa protein showed a positive immunoreaction with the rabbit polyclonal antiserum against the natural carp α-polypeptide.

    Notizen: Recombinant growth hormone (rGH) of fish was highly efficiently synthesized by Escherichia coli cells harbouring an expression plasmid (pKLYP) containing a 600 base pairs segment of cDNA which encoded the mature region of yellowfin porgy (Acanthopagrus latus) pre-growth hormone. After dena-turation and renaturation, rGH purified from inclusion bodies became the refolding form. rGH were then processed into the fishmeal pellet at various concentrations. Juvenile black sea-bream (Acanthopagrus schlegeli) fed twice daily with meal containing 0.5% rGH exhibited significant increases (P 〈 0.01) in percentage weight gain (60%) and feed efficiency (41%) relative to fish in the control group at week 12. Growth enhancement was maintained for at least 4 weeks (at week 16) after the termination of treatment.
