    Monospecific Polyclonal Anti-Anti-Idiotypic Antibodies to the Carboxyterminal Undecapeptide of the SV40 Large Tumour Antigen (1995)

    Notes: The murine monoclonal antibody PAb1605 defines an epitope, peptide Lys(698)-Thr(708) (KT), on the carboxyterminus of the tumour(T)antigen of SV40-transformed cells. In vivo and in vitro experiments had shown that this sequence represents an epitope for both humoral and cellular immune responses. When injected into rabbits PAb1605 induces anti-idiotypic antibodies (Ab-2). Ab-2β (internal image type) was purified by adsorption chromatography and characterized by the ability of KT to compete with the binding of ab-2 with ab-1. Murine anti-anti-idiotypic antibodies (ab-3) were obtained by immunization of mice with ab-2β. Both ab-1 and ab-3 JgG showed affinities to immunoprecipitated SV40 T antigen by immunoblot analysis and to nuclear SV40 T antigen by the immunofluorescence assay. The binding of ab-3 to SV40 T antigen was completely inhibited by competition with KT. We conclude that the polyclonal ab-3 is of the ab-3 subtype and specific for only one epitope which is represented by KT and defined by ab-1. The results demonstrate that the specificity for a defined peptide epitope of an antibody was conserved even after two consecutive steps of anti-idiotypic-antibody formation in two host species. Since this postulate of network theory could be verified for a sequence of a tumour-associated antigen which represents a B- and T cell epitope, this model is of great interest for further tumour immunological studies.

    Notes: Immune responsiveness to the streptococcal group polysaccharides at the clonal level can conveniently be monitored by analytical isoelectric focusing combined with autoradiography. The parameters investigated in this paper can be summarized as follows: [1] Within the first week after injection antistreptococcal polysaccharide responses are characterized by IgM antibodies, followed by subclass restriction in Balb/c mice to IgG2a antibodies. [2] The clonal pattern of specific IgG2a antibodies established in the second week after immunization does not change within the following 6 weeks. [3] Persistence of a specific response pattern established within a primary immunization course holds for Balb/c mice and rabbits for as long as 12 to 13 months. [4] Repeated immunization courses in mice and rabbits maintain established clonal antibody patterns in toto. Temporal variations of clonal expression are generally quantitative rather than qualitative. B-cell memory within this system is therefore stable and long-lived.

    Notes: Experimental primary, secondary and teritiary stimulation with streptococcal vaccine of isolated lymph nodes in vivo in the sheep model induces largely persistence of anti-group polysaccharide antibody clonotype patterns with the rure occurrence of additional clonotypes demonstrable after secondary stimulation persisting during the tertiary stimulus. It is not clear whether these additional clonotypes are the products of mutants or whether they pre-existed and were demonstrable only after a secondary stimulus, because of threshold concentrations required for identification. Further, isolated contralateral popliteal and prescapular lymph nodes of individual sheep share completely overlapping clonotype patterns during experimental primary anti-polysaccharide antibody responses indicating an identical repertoire of specific clonotypes under this condition of responsiveness.

    Notes: The level of antibodies specific for the two serologically distinct polysaccharides of group A and group A-variant streptococci was investigated in human sera by a modified Farr technique. Whereas there was an obvious relationship between the anti-streptolysin O titer and the level of group-A-specific antibodies, suggesting a recent streptococcal infection with group A streptococci, this correlation was not apparent for the level of group A-variant antibodies. There is evidence suggesting that children have significantly lower group A polysaccharide antibody levels than adults; this is not true of their group A-variant antibody levels. There was an apparent restriction of groups A- and A-variant-specific antibodies to the κ light-chain type and IgG2 heavy-chain subclass. Whereas only one serum contained group A-specific antibodies also associated with IgG3 heavy chains, nine sera contained anti-group A-variant-specific antibodies with both IgG2 and IgG3 heavy chains. No anti-polysaccharide activity was detected in the IgM and IgA class of serum antibodies. Anti-group A and anti-group A-variant antibodies of human sera are restricted with respect to the number of clonotypes that are shown by isoelectric focusing. The most heterogeneous sera contained roughly ten clonotypes.

    Notes: The variable-region subgroup determined by amino acid sequence analysis of heavy and light chains of two monoclonal cold agglutinins with the new anti-Gd specificity is reported. Both proteins belong to the VHI 11 subgroup of heavy chains; one light chain falls into the VKI subgroup, the other has a blocked N-terminus which so far has not been observed in human kappa chains. The comparison of anti-Gd with anti-l/-i or anti-Pr cold agglutinins indicates that anti-Gd differs from other cold agglutinins with respect to variable-region subgroup. The data extend previous findings on the restriction of certain antibodies to distinct variable-region subgroups.

    Notes: The early phase of antigen-dependent triggering of rabbit blood lymphocytes (PBL) is described. Specific activation of PBL by Streptococcal vaccines requires primed lymphocytes from high responder rabbits. B-cell memory is conferred by Ig receptors of both μ and γ chains. Antigen-induced incorporation of leucine and thymidine reach peak values al day 5, with stimulation indices of 10 to 〉 100, closely followed by increases in the number of living cells. A second and less intensive phase of cell proliferation is seen on days 8–11. A large fraction of cells (between 3 and 33%) participates in proliferation. A rough calculation of the average doubling lime of cells during the exponential growth phase gives values of 8–16 h.
