    Notes: Abstract Little is known about the biologic behaviors of cultured melanocytes in relation to donor age. To investigate age-dependent differences, neonatal and adult melanocytes were isolated from the same anatomical site, the foreskin, and cultured in the same growth medium supplemented with cAMP inducers (choleratoxin and 3-isobutyl-methylxanthine). The morphology, melanin content, pattern of melanosome distribution, and growth rate were then compared. Neonatal melanocytes were bipolar in appearance, whereas adult melanocytes were highly dendritic in appearance. Image analysis showed that adult melanocytes were larger and longer, and had a greater number of dendrites than neonatal melanocytes. When the growth medium was replaced by a medium without cAMP inducers, adult melanocytes showed a change in their morphology from dendritic to spindle-shaped, while the morphology of neonatal melanocytes remained unchanged. Melanosomes of adult melanocytes were distributed singly along the dendrites, and extracellular secretion of melanosomes was also found. In contrast, melanosomes of neonatal melanocytes were aggregated near the nuclei. No age-dependent differences in melanin content and growth rate were noted in the donor site-matched cultured melanocytes. These results suggest that donor age is one of the factors involved in determining melanocyte dendricity and melanosome distribution, and that increased dendricity of adult melanocytes is due to increased sensitivity to cAMP inducers. In addition, the adult melanocytes established in our culture system, which resembled dendritic melanocytes in vivo, could be considered a desirable model for studying the mechanisms of adult-onset hyperpigmentary disorders and melanogenesis.

    Notes: Abstract Efforts to transfer wheat curl mite (Eriophyes tulipae Keifer) resistance from Lophopyrum ponticum 10X (Podb.) Love to bread wheat (Triticum aestivum L.) have resulted in the production of a number of cytogenetic stocks, including an addition line of 6Ag, a “ditelo” addition line, and a wheat-Lophopyrum translocation line. Characterization of these lines with C-banding, in situ hybridization with a Lophopyrum species-specific repetitive DNA probe (pLeUCD2), and Southern blotting with pLeUCD2 and a 5S ribosomal DNA probe (pScT7) confirmed that the distal portion of the short arm of 6Ag was translocated onto the distal portion of 5BS (5BL. 5BS-6AgS). It was also determined that the “ditelo” addition was an acrocentric chromosome of 6AgS.

    Notes: Summary In situ hybridisation and restriction fragment length polymorphism (RFLP) analysis were used to determine the relative location of the translocation breakpoint and the size of the integrated chromatin segment in hexaploid wheat-Lophopyrum translocation stocks. Three 7el2-7D recombinant stocks were Robertsonian translocations, 7DS.7el. The remaining recombinant stock (KS10-2) was 7elS.7el-7DL and contained only the distal one-half of the long arm of 7D. The recombinant stock with 7el1 (K11695) could be designated 7DS.7DL-7el where approximately the distal one-half of 7DL was replaced. RFLP analysis indicated that on the 7DL RFLP map the breakpoints for K11695 and KS10-2 are in different locations and that the two recombinants contain an overlapping region (a common region) of the Lophopyrum chromosome 7 in which Lr19, a leaf-rust resistant gene, is located. RFLP analysis also indicated that RFLP markers which mapped to within 1.5 cm of the centromere of chromosome 7D are located in the distal half of the long arm.

    Notes: Nitrogen-14 nuclear quadrupole resonance frequencies at 77 K are reported for a series of carbazole compounds including N-vinylcarbazole. Differences in charge distribution between pyrrole and carbazole are explained on the basis of delocalization of pi electrons. Effects of the aldehyde and vinyl groups on charge distribution are discussed. Sigma bond polarization between the pyrrole nitrogen and the nitroso nitrogen toward the pyrrole nitrogen exists in N-nitrosocarbazole.

    Notes: Nitrogen-14 nuclear quadrupole resonance (NQR) spectra of nicotinic acid derivatives, known as antitumor agents, are reported and analyzed in the framework of the Townes and Dailey theory. A strong correlation exists between the charge difference σNC - π at the pyridinic nitrogen and the inhibition of acetylcholinesterase by nicotinic acid derivatives, in which the inhibition potency (pI50) increases as the charge difference σNC - π increases. There are significant differences between 14N NQR spectra for the pyridinic nitrogen sites of nicotinic acid and nicotinamide; these can be explained in terms of the differences in hydrogen bond lengths.
