    Notes: A combination of genomic in situ hybridization (GISH) and meiotic pairing analysis of crosses between a series of 2n= 56 partial amphiploids confirmed that the alien genome of the BYDV-immune Agro-tricum line OK7211542 is derived from Thinopyrum ponticum and not from Thinopyrum intermedium. The evidence from meiotic pairing analysis indicated that the chromosome constitution of OK7211542 is similar to another Agrotricum line, ORRPX, which was derived from a cross of wheat and Th. ponticum, but different from other Agrotricum lines, Zhong 5 and TAF 46 which were derived from the crosses between wheat and Th. intermedium. The GISH analysis confirmed that OK7211542 contained one complete set of 14 Th. ponticum chromosomes, in which no S chromosome was present in the alien genome. GISH also detected a small alien translocation attached to one of the wheat chromosomes, a result that was consistent with the pairing data.

An average meiotic pairing of 24.44I+ 5.07II+ 0.14IIIin the ‘Chinese Spring’×T. scirpeum hybrid (ABDE1E2) is attributed to two similar genomes from T. scirpeum (E1E2E3E4). An average meiotic chromosome pairing in the other hybrid (ABDJ1J2E3) was 31.70I+ 3.80II+ 0.90III and is attributed to autosyndetic pairing between the three genomes of T. junceum.

    Notes: Basal leaf sections of Triticum aestivum L., cv. ‘Chinese Spring’, and XTriticosecale Wittmack cv.‘Welsh’were cultured on Kao's medium with 2 mg/l of 2,4-D. Callus was induced on the leaf sections at frequencies of 64 and 63 %, respectively, and plant-lets regenerated at 18 and 31 %, respectively. Compared to the controls, the regenerated plants showed higher frequencies of univalent chromosomes at meiosis and low frequencies of trivalents, and quadrivalents; indicating that the callus phase had induced low frequencies of chromosome irregularity.

    Notes: The diploid hybrid Hordeum californicum×Secale vavilovii and the tetraploid hybrid H. parodii× S. anatolicum, were obtained through embryo rescue techniques at relative frequencies of 0.70 and 5.3 % of pollinated florets, respectively. Means of 11.741+ 1.1311 per cell at metaphase I of meiosis were obtained in the diploid hybrid and 14.741+ 6.3l11+ 0.22111+ 0.02IV in the tetraploid hybrid with a maximum of 6.01+ 4.011 and 9.011+2.0111+ 1.0IV respectively. In the diploid hybrid there was preponderance of intragenomic pairing (86.7 %). Increased pairing in the tetraploid hybrid was attributed to autosyndesis among Hordeum chromosomes.

    Notes: Summary In vitro culture of inflorescence tissue of a Triticum crassum (6x) x Hordeum vulgare cv. ‘Bomi’ (2x) intergeneric hybrid resulted in the proliferation of totipotent callus from which plants were regenerated. Regeneration was also achieved from immature inflorescence callus of T. crassum but not from H. vulgare. T. crassum x H. vulgare regenerates had a somatic chromosome number of 28, identical to that of the original hybrid. Four chimeric plants with a partially doubled chromosome number were obtained by in vitro colchicine treatment of hybrid callus prior to induction of plant regeneration. All T. crassum regenerates had 35 chromosomes rather than the expected number of 42. Meiotic analysis of a 35-chromosome plant revealed an extremely abnormal meiosis which might be attributable to a complete disturbance in meiotic control system(s) including that of meiotic pairing.

    Notes: Summary Chromosome pairing was studied in hybrids of (Hordeum vulgare ×Triticum aestivum) ×Secale cereale. Chiasma frequency per cell varied from 1.94 to 3.16 between the different hybrids. This variation was attributed to genetic variability in rye parents which affected homoeologous pairing. The pairing of rye chromosomes as revealed by Giemsa C-banding was a combination of nonhomologous association between rye chromosomes and associations with chromosomes of wheat and barley.

    Notes: Summary The C-banding pattern of the satellited chromosomes in Thinopyrum distichum and Triticum durum was established. Both T. durum and Th. distichum contained two pairs of satellited chromosomes, which could be distinguished from one another. In the amphiploids [T. durum×Th. distichum (2x=56)] and in the backcross T. durum/(T. durum×Th. distichum)2, BC1F3, and BC1F5 (2n = 42) the satellite was visible on only 1B and 6B of T. durum. The vector pTa 71 containing the rRNA gene from wheat hybridized to two pairs of chromosomes (four hybridization sites) in T. durum and Th. distichum, to eight sites in the amphiploid hybrid (2n=56), and to six sites in the backcross populations BC1F1. BC1F3, and BC1F5 (2n=42). The two satellite pairs in Th. distichum could be distinguished by the chromosomal location of the rRNA site (median or subterminal) and by the centromere position. One copy of each pair was present in the BC1F1, but in the BC1F3 and BC1F5 populations the pair with the subterminal location of rRNA genes was absent. Silver nitrate staining indicated that the rRNA genes of T. durum did not completely suppress those of Th. distichum. The octoploid amphiploid (2n = 56) contained a maximum of four large and four small nucleoli and the hexaploid BC1s (2n=42), four large and two small nucleoli.

    Notes: Abstract Twenty-five partial amphiploids (2n=8x=56), which were derived from hybrids of wheat (Triticum aestivum L.) with either Thinopyrum ponticum (Podpera) Liu & Wang, Th. intermedium (Host) Barkworth & D. Dewey, or Th. junceum (L.) A. Löve, were assayed for resistance to BYDV serotype PAV by slot-blot hybridization with viral cDNA of a partial coat protein gene. Three immune lines were found among seven partial amphiploids involving Th. ponticum. Seven highly resistant lines were found in ten partial amphiploids involving Th. intermedium. None of eight partial amphiploids or 13 addition lines of Chinese Spring –Th. junceum were resistant to BYDV. Genomic in situ hybridization demonstrated that all of the resistant partial amphiploids, except TAF46, carried an alien genome most closely related to St, whether it was derived from Th. ponticum or Th. intermedium. The two partial amphiploids carrying an intact E genome of Th. ponticum are very susceptible to BYDV-PAV. In TAF46, which contains three pairs of St- and four pairs of E-genome chromosomes, the gene for BYDV resistance has been located to a modified 7St chromosome in the addition line L1. This indicates that BYDV resistance in perennial polyploid parents, i.e., Th. ponticum and Th. intermedium, of these partial amphiploids is probably controlled by a gene(s) located on the St-genome chromosome(s).

    Notes: Abstract  Fusarium head blight (FHB or scab) caused by Fusarium spp. is a widespread disease of cereals causing yield and quality losses and contaminating cereal products with mycotoxins. The breeding of resistant varieties is the method of choice for controlling the disease. Unfortunately, the genetic basis of scab resistance is still poorly understood. We present the results of a back-cross reciprocal monosomic analysis of FHB resistance using the highly resistant Hungarian winter wheat line ‘U-136.1’ and the highly susceptible cultivar ‘Hobbit-sib’. Resistance testing was performed in a field trial artificially inoculated with a Fusarium culmorum conidial suspension. Five hemizygous families containing ‘U-136.1’ chromosomes 6B, 5A, 6D, 1B, and 4B had a visually reduced spread of infection compared to lines having the ‘Hobbit-sib’ chromosome. Chromosome 2B from ‘U-136.1’ had an increased spread of infection. The critical chromosomes controlling seed weight were 6D, 3B, 5A, and 6B while those controlling deoxynivalenol (DON) content were homoeologous groups 2 and 6, although the latter effects were not significant due to a high coefficient of variation. Results from this and other studies show that chromosomes 6D, 6B, 5A, 4D, and 7A have frequently been associated with scab resistance in a number of wheat cultivars. Research groups now attempting to map scab resistance in wheat using markers should pay special attention to the above-mentioned chromosomes.
