    Notes: The activity of tyrosine hydroxylase (EC 1.10.3.1) when assayed under ideal conditions in young human brains, was comparable to that in brains of other species in level of activity and distribution. The highest levels of activity were in the putamen, caudate nucleus and substantia nigra, in keeping with data on other species. The caudate activity in human brain appeared to decrease substantially with increasing age. In both humans and baboons, the enzyme in the neostriatum was particle-bound and inhibited by the 2-amino-4-hydroxy-6,7-dimethyltetrahydropteridine cofactor system. In the substantia nigra it was soluble and stimulated by the 2-amino-4-hydroxy-6,7-dimethyltetrahydropteridine cofactor system. The data suggest that tyrosine hydroxylase may be produced in a soluble form in the cell bodies of the substantia nigra but become bound as it moves toward the nerve endings in the putamen and caudate nucleus. The bound form of the enzyme was unstable but the soluble form exhibited considerable stability.

    Notes: The levels of certain amines (catecholamines, 5-HT, and ‘histamine’) and of certain enzymes (tyrosine hydroxylase, tryptophan hydroxylase, ChAc, or AChE) in whole brain or selected brain areas of rats and mice susceptible to audiogenic seizure have been compared with the levels in matched groups of non-sensitive animals. Sensitive groups included both those where susceptibility is inborn and those where it is induced by administration of methionine sulphoximine or thiosemicarbazide. No significant difference was found which could be correlated with susceptibility to audiogenic seizure.

    Notes: Abstract— An analysis of proteins undergoing axonal transport in nigro-striatal neurons, after the stereotaxic injection of [3H]leucine into the substantia nigra of rat brain was performed. As early as 6 h after the injection [3H]proteins appeared in the caudate-putamen. The maximum accumulation was at 5 days and there was still residual protein radioactivity present at 30 days. About 70 per cent of the total radioactive protein in the caudate-putamen was solubilized by homogenization in 0–5%, (v/v) Triton X-100 and remained in the supernatant on centrifuging for 1 h at 100,000 g. The supernatant fraction, when chroma-tographed on a DEAE-cellulose column, was resolved into four protein peaks (A, B. C and D) which were found to be labelled differently as a function of time after the injection of [3H]leucine. Peak A was substantially labelled in a first phase (6–24 h) and reached its maximum in a second phase (5 days). The proteins comprising this peak appeared to undergo both fast and slow axonal transport. Although some labelling in peak B was evident at 6 h, maximal activity did not occur until 5 days. No radioactivity could be detected in peaks C and D at 6 h. Maximal labelling of these two peaks also occurred at 5 days. These data suggest that the proteins of peaks B, C and D were transported primarily by slow axoplasmic flow. The radioactive protein peaks A and B from the second phase of the transport were excluded from a Sephadex G-200 column, pointing to their high molecular weights (13,000–200,000). Peak B. which had the highest specific radioactivity (c.p.m./mg protein) at 5 days, contained a significant level of tyrosine hydroxylase, an important component of dopaminergic neurons.

    Notes: Abstract: A radiometric assay for ornithine aminotransferase was developed using [1-14C]α-ketoglutarate as the labeled substrate and glutamate decarboxylation as a linking step. This assay gives near total measurement of ornithine aminotransferase activities that are, respectively, about 1.5 and 10 times larger than those obtained by the spectrophotometric assay and the radiometric assay using [1-14C]ornithine. It is also the most sensitive of the three assay procedures.Consistent with previous reports, brain ornithine aminotransferase was found to be present predominantly in synaptosomes. Regional distribution of the enzyme correlated with that of the high-affinity uptake of glutamate, but not with the distribution of glutamate decarboxylase. Ornithine aminotransferase may be responsible for the synthesis of glutamate in glutamatergic neurons but it is clearly not localized exclusively in such neurons.

    Notes: Abstract: Regional differences in peroxide levels in rat brain are relatively small but the highest levels were found in the substantia nigra. Lipid peroxides of rat whole brain were significantly increased (22%) immediately after exposure to oxygen at high pressure (OHP). However, this increase was transient. No significant difference from control was observed 3 h after exposure to OHP. The greatest increases were in the cerebral cortex and spinal cord. The olfactory bulb, caudate-putamen, globus pallidus, corpus callosum, septum, hypothalamus, substantia nigra, and inferior colliculus all showed moderate but significant increases, whereas the accumbens, thalamus, hippocampus, cerebellum, and amygdala showed no significant change. This regional difference in lipid peroxidation may indicate a regional susceptibility to the damaging effects of free radical reactions.
