    Notes: Zusammenfassung Vor dem Hintergrund der vielfältigen Endotoxoidwirkung wurde diese Substanz in chronischen Toxizitätsstudien beiMacaca mulatta geprüft. Endotoxoid wurdeMacaca mulatta in einer Dosierung von jeweils 100γ/kg in insgesamt 51 Injektionen innerhalb von 12 Wochen injiziert. Dabei zeigte sich, dass diese Substanz keinen Einfluss auf die geprüften klinischchemischen Parameter hatte und bei makroskopischer und histologischer Untersuchung keine Veränderungen auftraten.

    Notes: Summary The findings reported here indicate that toxic reactions to endotoxin do not have to be part of the mechanisms leading to clinically improved status of patients or of experimental animals. It seems to be possible to mobilize the host defenses without the unwanted or dangerous side effects which were encountered during fever therapy. For the demonstration of activated host defenses, several experimental models are available. Non-specific resistance to bacterial infections, to lethal irradiation or to experimental challenge with viable tumors are clear manifestations of enhanced capacity of the endotoxin-treated host to compensate for the induced damage, or to cope with an otherwise overwhelming number of pathogenic cells. Immunopotentiation is obviously one of the contributing factors in some of the above parameters. How these results are applicable to human therapy remains to be seen. Such use of endotoxin derivatives, or better, the mediators of beneficial endotoxic effects, must wait until we learn more about the nature of the alternating positive and negative effects of endotoxin application. The importance of the proper timing of such a therapeutic modality cannot be overemphasized.

    Notes: Summary Five tumor chemotherapeutic drugs: adriamycin (ADM), formyl leurosine (FLE), 1,2–5,6-dianhydrogalactitol (DAD), 5-fluorouracyl (5-FU), and vincristine (VCR) were tested for their effect on the immune response to sheep red blood cells (SRBC) in mice in the Jerne plaque assay. It was found that four of these can act as both immunostimulators and immunosuppressants, depending upon the time elapsing between their application and immunization. VCR was found to be slightly immunopotentiating. Proper combination and timing of injections of DAD and FLE enhanced the immune response over 23fold. Administration of the same drugs in reversed sequence led to an almost complete immune suppression. Repeated injections of some drugs alone or in combination also led to marked or complete immunosuppression. The combined use of drugs and endotoxic lipopolysaccharide (LPS) or its nontoxic, polysaccharide-rich hydrolytic breakdown product (PS) could compensate for some of the immunosuppressive effects. The most striking adjuvant effect was elicited by the combined use of VCR and LPS or VCR and the nontoxic PS preparation.

    Notes: Summary In this study a variety of immunostimulatory agents was tested alone or in combination with other agents to measure their ability to enhance nonspecific resistance to challenge with the TA3-Ha transplantable murine ascites tumor. Lipopolysaccharides (LPS) were effective alone in protecting mice from tumor. Their lipid-free, nontoxic, polysaccharide-rich hydrolytic products (PS) showed a much reduced, but still significant effect in anti-TA3-Ha resistance induction. Other agents that stimulated tumor resistance included nontoxic ‘native hapten’ and trehalose-6,6′-dimycolate (P3) of Ribi, synthetic glycolipids, cord factor, and killed BCG preparations. Simultaneous pretreatment with a combination of any of these agents and either LPS or PS resulted in a significantly higher level of tumor resistance. Administration of LPS, PS, or native hapten to mice that had been previously infected with viable BCG resulted in the strongest antitumor effect. These studies demonstrate that nonspecific resistance can be enhanced in an additive or synergistic manner by using combinations of agents, which presumably stimulate different cell populations of the host's immune system.

    Notes: Summary The transplantable line-10 hepatocellular carcinoma of guinea-pigs has been used as a model for the study of immunotherapy of malignant tumors. Cure rates of up to 100% have been obtained with ReGl-CM from 0 antigen-deficient (Re) mutant strains of Enterobacteriaceae, provided they were combined with mycobacterial trehalose dimycolate (cord factor, P3). Whereas highly endotoxic LPS extracts from all wild-type strains so far tested have failed to cause tumor regression, acid hydrolysis of such LPS samples led to residual fractions (RESI) that cross-reacted serologically with ReGl-CM samples (Chang and Nowotny, 1975) and provided cure rates up to 100%. RESI from Serratia marcescens was essentially nonpyrogenic and 100 times less lethal for chick embryos than potent endotoxins. Antigenic material associated with endotoxic extracts appears to be cryptic or sterically hindered from being effective in wild-type LPS but is exposed in ReGl and RESI samples. Reduction of the aminoacid content of ReGl-CM by microparticulate silica gel chromatography or by treatment with Triton X-100 significantly lowered the ability to bring about tumor regression without affecting endotoxicity. Antitumor activity could be restored by the addition of synthetic N-acetyl-muramyl-l-seryl-d-isoglutamine (MDP) or a nontoxic lipoid side fraction recovered during the isolation of ReGl-CM, which contained a small amount of peptidic substances. It is concluded that the addition of peptidic material, which may act as an adjuvant, to endotoxins is required to make them useful for immunotherapy of the weakly immunogenic line-10 tumor. Chemical procedures known to ‘detoxify’ endotoxins while retaining adjuvanticity, such as succinylation and phthalylation, resulted in complete loss of endotoxicity and tumor-regressive potency of ReGl-CM. Transesterification with sodium methoxide led to a water-soluble phase, which cured 50% of tumor-bearing animals even though lethality and pyrogenicity were reduced by 100 times and 50 times, respectively. Thus there was no direct correlation between endotoxic potency and tumor-regressive activity. In addition, our findings indicate that a low level of toxicity may be required to obtain optimal levels of tumoricidal action.
