Notes: Abstract 3H-Histamine binding, uptake and metabolism were investigated in intact isolated and enriched parietal cells from the dog and guinea pig. Histamine uptake was sodium dependent and followed by intracellular metabolism. The only metabolite that was detected and extracted from cytosol has been identified by TLC to beN r-methylhistamine. The histamineN-methyltransferase activity appeared to be sodium dependent and was inhibited by mepyramine and chlorpromazine, and also by higher concentrations (10−4–10−3 mol/l) of cimetidine. Two blockers of the sodium channel, amiloride and aminoguanidine, also reduced the enzyme activity by an as yet unknown mechanism.

Notes: Abstract 3H-atropine binding was studied in isolated intact guinea pig gastric mucosal cells and has been shown to be saturable. Scatchard plots of the binding system calculated under the assumption of only one binding site revealedK D values for the parietal cell enriched population of 1.25×10−6 mol/l and 1.38×10−6 mol/l for the nonparietal cells. The results show that in the gastric mucosa both parietal and nonparietal cells contain cholinergic receptors.

Notes: Summary 1. The absorption of tritiated water from rat colon in vivo and the influence of 5-HT on this process were investigated. 2. The rate of absorption from colon was only one-fifth of that from small intestine. 3. An i.v. infusion of 5-HT (5 μg/min) reduced the rate of labelled water absorption by about 50% whereas a single injection of 2.5 mg 5-HT into the lumen of the colon reduced the rate by only 25%. 4. The volume of distribution of tritiated water was reduced by an i.v. infusion of 5-HT but not by its injection into the colon.

Notes: Summary 1. The intestinal absorption and metabolism of 14C-5-HT have been investigated, in situ, in tied off sections of the cat jejunum. 2. Less than 2% 14C-5-HT appears in the venous effluent within 60 min. A large amount of the injected 14C-5-HT is metabolized to “free” or “conjugated” 14C-5-hydroxyindoleacetic acid (14C-5-HIAA). 3. Pretreatment with nialamide increases the amount of unchanged 14C-5-HT in the mucosa about 35 times, in the blood about 6 times; the formation of 14C-5-HIAA is diminished in the mucosa and its appearance in the blood to about one fifth. 4. The results after pretreatment with chlorpromazine in addition to nialamide lead to the conclusion that chlorpromazine slightly diminishes the uptake of 14C-5-HT by the mucosa.

Description / Table of Contents: Zusammenfassung 1.
Notes: Summary 1. The effect was studied of the gastrin-analogue tetrapeptide amide Try.Met.Asp.Phe-NH2 and electrical vagus stimulation on gastric secretion in anaesthetized gastric fistula cats. 2. The tetrapeptide in doses of 0.1–3.2 μg/min i.v. stimulated the secretion of gastric juice containing only small amounts of pepsin. 3. Electrical vagus stimulation provoked the secretion of large amounts of gastric juice and pepsin. 4. In pairs of cross circulated cats the gastric juice of the vagus stimulated animals contained much pepsin, the gastric juice of the not stimulated animals did not. 5. It was concluded from these results that gastrin was released by vagal excitation and stimulated as a circulating hormone acid secretion and that pepsin secretion was stimulated by a vagal release of acetylcholine.
