    Notes: Five patients with Behçet's disease were satisfactorily treated with colchicine. Oral aphthosis, erythema nodosum-like lesions and genital erosions improved greatly within a month as did laboratory findings. We believe colchicine to be the first choice in the management of the cutaneous and ocular lesions of Behçet's disease.

    Notes: An effect of colchicine on the active Arthus reaction in rabbits was investigated in vivo with simultaneous evaluation of in vitro chemotacdc activity of polymorphonuclear leukocytes. Arthus reactions were induced by intracutancous injection of ovalbumin into rabbits prcimmunized with ovalbumin. Colchicine, 1 mg/kg or 2 mg/kg, injected intraperitoneally one hour prior to the induction of the Arthus reaction, suppressed erythema and induration at 12 h, when histology showed decreased numbers of infiltrated polymorphonuclear leukocytes and no apparent vascular damage. Perivascular deposits of C3, suggestive of the presence of immune complexes, persisted to 24 h. In vitro chcmotaxis of polymorphonuclear leukocytes using Boyden chamber techniques revealed suppression of the activity in the colchicinc–treatcd animals at 6 and 12 h.These findings suggest that colchicine inhibits vascular injury by interfering with directional chemotaxis of polymorphonuclear leukocytes to the lesional sites.

    Notes: Background Patients with systemic sclerosis (SSC) show a capillary abnormality of nailfolds with controversial correlation with organ involvement. Our purpose was to study the correlation between this nailfold capillary abnormality and pulmonary hypertension in patients with SSC. Methods We studied the nailfold capillaries, using capillary microscopy, and the pulmonary arterial pressure, using right-heart catheterlzation, in 44 patients with SSC. Canonical discriminant analysis was used to define the capillary abnormality in patients with SSC, which was then compared with that of 40 normal controls. The correlations between the patterns of nailfold capillaries and the cardiopulmonary findings. Including the pulmonary arterial pressure, were examined using Fisher's test. Results Thirty-two of 44 patients with SSC could be differentiated from normal controls by our definition of the SSC pattern. The SSC pattern correlated significantly with elevated pulmonary vascular resistance, as well as with pulmonary fibrosis, eiectrocardiographic abnormalities, decreased vital capacity, and decreased diffusing capacity for carbon monoxide. All SSC patients with pulmonary hypertension showed this SSC pattern. In patients with elevated pulmonary arterial pressure, capillary microscopy and diffusion capacity for carbon monoxide (DCCM) showed the highest rate of abnormalities. A limited-type SSC significantly correlated with DCCM and with anticentromere antibody, and the diffuse-type SSC with pulmonary fibrosis and anti-sci-70 antibody. Conclusion Our data suggest that in patients with SSC, nailfold capillary abnormalities correlate with pulmonary arterial hypertension as well as with clinical and laboratory findings indicating pulmonary hypertension.

    Notes: Abstract The effect of the supernatants obtained from the liquid culture medium ofSporothrix schenkii andCandida albicans on the generation of superoxide anion (O 2 − and hydroxyl radicals OH., the elements of reactive oxygen species (ROS), and chemilimunescence (CL), a measure of several ROS, by polymorphonuclear leukocytes (PMNs) was examined. In our study, it was shown that the supernatant ofS. schenkii increased all types of ROS generation examined and CL, while that ofC. albicans increased OH. generation and CL. The effect of the supernatants ofS. schenkii on OH. generation and CL and that ofC. albicans on CL were most remarkable when the supernatant obtained 8 weeks after the inoculation was used. The supernatant ofS. schenkii was shown to be a much more potent stimulant than the supernatant ofC. albicans. This ROS-stimulating effect of the supernatant ofS. schenkii was heat stable but not dialyzable. These findings suggest the possible role of ROS produced by infiltrated PMNs in the inflammatory skin lesions induced byS. schenkii.

    Notes: Summary The effects of psoriatic sera were investigated on the generation of oxygen intermediates (OIs) by normal polymorphonuclear leucocytes (PMNs). Although increased superoxide generation was noted, a further respiratory burst of the PMNs was significantly suppressed. Since superoxide dismutase (SOD) and catalase activities of the sera assayed in the xanthine-xanthine oxidase system were comparable to the controls, it still remains obscure why this dissociation occurs. It is suggested that increased generation of superoxide anion from the PMNs may be another facet of PMN activation which plays an important role in the psoriatic process.

    Description / Table of Contents: Zusammenfassung Die celluläre Lokalisation von cAMP und cGMP in der Meerschweinchenlippe wurde mit einer indirekten Immunofluorescenztechnik untersucht. Es konnte gezeigt werden, daß cAMP in Zellmembranen und in cytoplasmatischen Elementen mit grober und granulärer intranukleärer Fluorescenz verteilt ist, während cGMP in nukleären Lagen einschließlich nukleärer Membranen sowie Zellmembranen und Cytoplasmata vorkommt. Im basalen Anteil war die Fluorescenz von cAMP und cGMP in Cytoplasma und Zellmembranen schwächer als im oberen Anteil.
    Notes: Summary The cellular localization of cAMP and cGMP in the guinea pig lip was investigated with an indirect immunofluorescence technique. cAMP was distributed both in cell membranes and cytoplasmic elements with coarse and granular intranuclear fluorescence, while cGMP was localized in nuclear sites including nuclear membranes as well as cell membranes and cytoplasm. In the basal portion, both cAMP and cGMP fluorescence was weaker in cytoplasm and cell membranes than seen in the upper portion.

    Notes: Summary The effect of sera from patients with untreated leukocytoclastic vasculitis was investigated on the generation of oxygen intermediates by normal polymorphonuclear leukocytes. Sera from untreated patients induced increased hydroxyl radical generation, which is one of the most potent oxidants capable of causing tissue damage. It is suggested that vascular injury may be mediated in part by enhanced production of hydroxyl radical by polymorphonuclear leukocytes. Circulating immune complexes in the sera of the patients are considered to be one of the factors responsible for enhanced generation of hydroxyl radical.
