    Notes: The effect of prolonged water deficit on mRNA translation was quantitatively analysed in apical and basal leaves of whole tobacco (Nicotiana tabacum) plants. The level of large polysomes (six or more ribosomes per mRNA) was significantly higher in apical than basal leaves under well-watered conditions. In both young and old leaves, water deficit caused a progressive reduction in levels of polysomes with concomitant increases in 80S monosomes, indicative of reduced initiation of translation. Despite the global reduction in polysome formation over 144 h of water deficit, the water-deficit-induced putative lipid transfer protein (ltp) mRNA was associated with large polysomes and LTP content increased. Osmotin (osm) mRNA, another water-deficit-induced transcript, also remained associated with large polysomes during prolonged water deficit. In contrast, mRNAs encoding the non-stress proteins ribulose bisphosphate carboxylase/oxygenase small subunit (rbcS) and eukaryotic initiation factor 4A (eIF4A) decreased in abundance and shifted to small polysomes and non-polysomal complexes, indicating that initiation of translation of these mRNAs was impaired by water deficit. These results show that translational regulation is a component of the water-deficit response and the differential translation of individual mRNAs is distinct in leaves of different age.

    Notes: Helicobacter pylori infection is involved in the formation of chronic peptic ulcer. However, a previously reported hypothesis concerning the involvement of central autonomic nervous disorder in this condition cannot be ruled out.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To use spectrum analysis of heart rate viability to examine autonomic nervous activity before and after H. pylori eradication.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:Twenty patients with chronic duodenal ulcer (duodenal ulcer group) and 20 age-matched normal adults (N group). In both groups, 24-h Holter electrocardiograms (ECGs) were recorded and spectrum analysis of heartrate variability was performed. In the duodenal ulcer group, Holter ECG was recorded before and after H. pylori eradication.〈section xml:id="abs1-4"〉〈title type="main"〉Results:In the N group, analysis of heart rate variability showed that high frequency (HF) power, an index of parasympathetic activity, was high at night, while the low frequency (LF)/HF ratio, an index of sympathetic function, was high during the daytime. In the duodenal ulcer group, HF power was higher at night than during the daytime, showing a similar pattern to the N group, but the power value was higher than in the N group (P 〈 0.05). In the duodenal ulcer group, LF/HF at night was significantly higher than that of the N group. In addition, in the duodenal ulcer group, autonomic activity after H. pylori eradication did not differ significantly from that before H. pylori eradication.〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:In patients with chronic peptic ulcer, both sympatheticotonia and parasympatheticotonia may occur at night, and this abnormality in autonomic nervous activity may cause increased gastric acid secretion and gastric mucosal vasoconstriction. Abnormalities in autonomic activity persist even after H. pylori eradication, suggesting that they may be an independent risk factor in the formation of chronic peptic ulcer in addition to H. pylori infection.

    Notes: Summary To investigate the influence of the mitochondrial aldehyde dehydrogenase 2 (ALDH2) genotype on the clinical features of diabetes, 212 Japanese patients with non-insulin-dependent diabetes mellitus (NIDDM) (154 males and 58 females aged 17–83 years; mean age 58.2 years) were investigated. Genotyping of ALDH2 was performed by the polymerase chain reaction – restriction fragment length polymorphism (PCR-RFLP) method. The pattern of inheritance of diabetes and various clinical parameters was compared between active and inactive ALDH2 groups. Of the 212 subjects, 120 had active ALDH2 and 92 had inactive ALDH2. The percentage of patients with a diabetic mother was higher in the inactive ALDH2 group (32.6 %) than in the active ALDH2 group (19.2 %) (p 〈 0.05). The prevalence of proliferative retinopathy was lower in the inactive ALDH2 group than in the active ALDH2 group (p 〈 0.05). However, other clinical parameters showed no difference. We conclude that maternal inheritance of diabetes was common in the inactive ALDH2 group. The finding is suggestive of a relationship between alcohol intolerance and inheritance of diabetes. We speculate that the interaction between mitochondrial DNA and ALDH2 inactivity causes an increase of mitochondrial DNA mutations or deletions, thereby inducing the maternal inheritance of diabetes. The relationship of the ALDH2 genotype with proliferative retinopathy is interesting, because it resembles that of chlorpropamide alcohol flushing with severe diabetic retinopathy. The interaction of aldehyde dehydrogenase isoenzymes might have an aetiological role, since aldehyde dehydrogenase 1 plays an important part in oxidation of retinal to retinoic acid. However, the number of affected patients with proliferative retinopathy was small, hence, our result should be considered as a preliminary finding. [Diabetologia (1996) 39: 1115–1118]
