    Notes: The purpose of the present paper is to consider the von Neumann reflection (vNR) which takes place for a small incident shock Mach number and a small wedge angle. A series of experiments has been performed with ordinary smooth straight wedges and step-like wedges that simulate the former. The reflection configuration over the step-like wedge has suggested unsteady characteristics of the vNR. Contrary to the established notion of shock reflection phenomena over straight wedges, while the triple point trajectory was approximately a straight line through the apex of the wedge, the vNR showed unsteadiness in the relation between angles of incidence and reflection (ωi,ωr), and thus the flow-field proved to be non-self-similar near the triple point. Based on the measured values, the incident angle for the reflected wave and the Mach number of the flow ahead of the reflected wave were estimated. These values show that the reflected wave is not a Mach wave, but it moves on the (ωi,ωr)-plane almost along a trivial solution. In particular, the flow Mach number ahead of the reflected wave approaches unity, which leads to analytical formulas for angles of incidence and reflection as functions of the incident shock Mach number only. The reflected wave degenerates to a normal Mach wave as the incident shock proceeds. In the case of the vNR, the von Neumann paradox takes place only at an early stage of reflection, and the paradox is resolved later, since the flow properties such as angles of incidence and reflection are given by the trivial solution which is a particular solution for the three-shock theory. © 2000 American Institute of Physics.

    Notes: Histologically, the cassava pistil is trilocular with a single ovule in each locule producing a maximum of three seeds in nature. Heat stress (35°/45°C, night/day temperatures), as a physical treatment was attempted to induce useful reproductive and vegetative traits in 10 F1 hybrids derived from ‘OMR 36-41’and ‘OMR 36-42’crosses, and two Asian cassava cultivars,‘Rayong 60’and ‘Rayong 1′, under greenhouse and field conditions. Cytoembryological examination of the repro- ductive organs revealed morpho-structural features of normal pistil in all clones except ‘Rayong 1’which exhibited reproductive variations under greenhouse culture. Genetically, an important variation was ovule dichotomy per locule of the trilocular pistil so that each pistil contained six instead of the usual three ovules and lacked any outer integument. These ovules possessed functional embryo sacs with varying nuclear number. A pair of the dichotomous ovules possessed two embryo sacs of apparent different megaspore mother cells. Vegetatively, F1 hybrids were dwarf (0.50m) in their phenotype and flowered 1 month earlier than the 1.5-2.0m tall cultivars under greenhouse conditions.

    Notes: Microsporogenesis and megasporogenesis were cytogenetically and histologically analysed in three cassava clones:‘Rayong 1′,‘Rayong 60′,‘M. mga’ and two hybrid lines,‘OMR 3641-1’ and ‘OMR 3641-1’ to elucidate the evolution of sexual polyploids in cassava. At telophase II, formation of 17-21 micronuclei per pollen cell plate was observed in 16 out of 351 cell plates in ‘M. mga’. Micronuclei were observed at low (0.3-2.3%) frequencies, at the sporad stage in all clones. Monads, dyads, triads and tetrads, which are established sources of high ploidy levels were observed at low (2.6%) and high (22.2%) frequencies. Megasporogenesis in ‘Rayong 1’ and ‘Rayong 60’ showed a lack of second meiotic divisions after a successful first division that resulted in partly unreduced embryo sacs with 2n eggs, suggesting another unrecognized and, as yet, unreported source of sexual polyploid formation in cassava. Meiotic abnormalities during microsporogenesis and megasporogenesis are implicated as being responsible for the formation of mixoploids (triploids and tetraploids) in cassava breeding programmes. A cytogenetic mechanism resulting in bilateral sexual polyploids through different gametic fertilization pathways in cassava is suggested and its role in breeding is briefly discussed.

    Notes: To resolve the problem of low and unstable yield in buckwheat under unsuitable environmental conditions, five varieties were grown and compared in summer and autumn. Vegetative and reproductive growth characteristics were investigated, and embryo development was observed with Nomarski's differential interference contrast (DIG) microscopy. In summer, under high temperatures and longer day-length, autumn-type varieties showed faster vegetative growth and later reproductive development as compared to summer-types. In the autumn, with generally good climatic condition for buckwheat growth in southern part of Japan, the embryo sacs and zygotes of both types developed normally. At 3 days after pollination (DAP), the embryos reached the globular stage. In summer, however, mostly abnormal embryo sacs were observed 3 DAP, leading to incomplete development, failure of fertilization or abortion of embryo. The abnormal embryo sacs were classified into 6 types. Abnormality of either type occurred in 91—100 % of the tetraploids, which is higher than in the diploids (50—77 %). The present study, conducted to clarify the factors causing low seed set in buckwheat under summer conditions, provides detailed information for further breeding of environment-tolerant buckwheat. Di-ploid genotypes which are less sensitive to unfavourable conditions and, thus, show more efficient fertilization in summer, should be considered for use as breeding material in buckwheat.

    Notes: A method of sample assembly for four-probe resistivity measurements on fragile organic single crystals using a diamond anvil cell is presented. A procedure to keep insulation between the metal gasket and four leads of thin gold wires bonded to the sample crystal by gold paint is described in detail. The resistivity measurements performed on a single crystal of an organic semiconductor and that of neutral molecules up to 15 GPa and down to 4.2 K showed that this new procedure of four-probe diamond anvil resistivity measurements enables us to obtain sufficiently accurate resistivity data of organic crystals. © 2001 American Institute of Physics.

Survival and death of Chara internodal cells were investigated in one of the alkali metal salts KCl, some of the alkali earth metal salts CaCl2, Ca(NO3)2, MgCl2, Mg(NO3)2, SrCl2, Sr(NO3)2, BaCl2 and Ba(NO3)2, potassium phosphate pH buffer solution (pH 7.0), Tris-maleate pH buffer solution (pH 7.0), HEPES (N-2-hydroxyethylpiperazine-N′-2-ethanesulphonic acid)-KOH (pH 7.0) pH buffer solution, calcium buffer solutions, and deionized water. Most of the internodal cells died within a day or a few days in KCl, MgCl2, Mg(NO3)2, BaCl2 and Ba(NO3)2 solutions of higher concentrations, calcium buffer solutions of pCa 6.0, 10.0 mol m-3 potassium phosphate pH buffer solution and 10.0 mol m-3 Trismaleate pH buffer solution. However, all of the internodal cells survived more than 10 d in deionized water, 80.0 mol m-3 CaCl2, 80.0 mol m-3 Ca(NO3)2, 80.0 mol m-3 SrCl2, 80.0 mol m-3 Sr(NO3)2 calcium buffer solutions of pCa 4.0 and pCa 5.0, and 10.0 mol m-3 HEPES-KOH (pH 7.0) pH buffer solution. Addition of Ca2+ or Sr2+ to K+, Mg2+ and Ba2+ salt solutions increased the survival rates of the internodal cells. Calcium release from the internodal cell wall was measured in deionized water, KCl, NaCl, MgCl2, CaCl2, SrCl2 and BaCl2 solutions. Except in deionized water and CaCl2 solution, most of the calcium binding to the cell wall was released within one or a few hours in respective electrolyte solutions. Thus, survival and death of the internodal cells in the electrolyte solutions tested were interpreted in terms of the calcium release from the cell wall and the cell membrane, and intrinsic ability of Sr2+ to maintain the cell membrane normal.

    Notes: Background Eosinophils play a critical role in the pathogenesis of allergic diseases. CC chemokines, such as regulated on activation, normal, T cell expressed, and secreted (RANTES), are key regulators of eosinophil locomotion. Although eosinophils migrate from the bloodstream into tissues, mechanisms that generate a chemogradient across the endothelium remain to be fully elucidated.Objective We first examined the polar secretion of RANTES by endothelial cells. We also studied the functional scavenging effect of red blood cells (RBCs) on RANTES secreted into the intravascular side.Methods and results Endothelial cells were cultured in a transwell chamber with a membrane pore size of 0.45, 3.0, and 8.0 μm and stimulated with TNF-α, IL-1β, or IFN-γ from the apical or basolateral side for 16 h. The measurement of RANTES in the supernatant was performed by ELISA. We did not see any difference in the amount of RANTES secreted from the cytokine-stimulated endothelium between inner (intravascular side) and outer (extravascular side) wells separated by the 8.0-μm membrane, although apical polarization was observed with the 0.45-μm membrane. The addition of RBCs (hemoglobin (Hb): 0.5–15 g/dL) to the apical supernatant of TNF-α-stimulated endothelial cells reduced the RANTES level in a concentration-dependent manner. The treatment of supernatant on the intravascular side with RBCs significantly enhanced the migration of eosinophils.Conclusion RBCs possess a scavenging effect on intravascular RANTES, and thereby regulate transendothelial migration of eosinophils. Our findings suggest a new role of RBCs in allergic inflammation.

    Notes: Background We have shown that interleukin-5 (IL-5) and granulocyte-macrophage colony-stimulating factor (GM-CSF) are present in sputum from patients experiencing acute asthma attacks, by eosinophil survival assay. The viability of guinea-pig eosinophils was significantly increased in the presence of such sputum extracts after 3 days' culture, and it was inhibited by the addition of anti-IL-5 and anti-GM-CSF antibodies. However, the contribution of IL-5 to the increase in eosinophil viabihty was less than expected from the values of IL-5 measured by enzyme-linked immunosorbent assay (ELISA). Therefore, we speculated that something in sputum inhibited the function of IL-5.Objective Tratnsforming growth factor-β (TGF-β) was the only cytokine we tested that inhibited the prolongation of survival of guinea-pig eosinophils induced by IL-5. The objective of this study is to detect TGF-β in the same sputum. Methods Guinea-pig eosinophils were cultured with or without anti-TGF-β antibody in the presence of sputum extracts, and the eosinophil viability was counted after 3 days. Measurement of TGF-βl in sputum was performed by ELISA. Results Eosinophil viabilities with and without anti-TGF-β antibody were 79.7 ± 2.9% and 69.0 ± 2.7%, respectively, and the difference between them was statistically significant (P 〈 0.05, n = 9). The concentration of TGF-β1 in the sputum was 21.7 ± 3.3 ng/mL (n = 9).Conclusion These observations suggest that TGF-β is present in sputum from patients with bronchial asthma.
