    Notes: Summary A pituitary adenoma was transsphenoidally removed from a 4.5-year-old girl suffering from gigantism. Prior to the operation both the growth hormone (GH) and the prolactin (PRL) levels in the serum were elevated. By light microscopy the tumor appeared to be an acidophilic adenoma. Two distinct cell types, the densely granulated and the sparsely granulated cells, could be distinguished by electron microscopy. Double immunolabeling revealed the presence of GH alone in some densely granulated cells and PRL alone in some sparsely granulated cells, as well as GH and PRL co-localized in both of the morphologically distinguished cell types. Both cell types were identified in the monolayer and the suspension cultures by electron microscopy. GH and PRL concentrations in the culture media were measured by radioimmunoassay. The basal secretion of growth hormone was almost uniform during the 3-week cell culture period. GH and PRL release was significantly inhibited by bromocriptine. Our studies revealed a bimorphous and bihormonal mixed adenoma in childhood.

    Notes: Summary E600 resistent non-specific esterase activity or acid phosphatase activity were localized in corticotrophic cells identified by postembedding immunocytochemistry (PAP or protein A-immunogold techniques). The lysosomal system of this cell type consists of dense bodies, of a population of small lysosomes mostly situated at the cell periphery in the vicinity of secretory granules as well as of tubular structures. These latter were located either in the central part of the cytoplasm and probably belonged to the Golgi apparatus or at the cell periphery, partly in the extensions. Small lysosomes occurred to be in continuity with enzyme-containing tubules. In a few structures lysosomal enzyme activity and ACTH immunoreactivity overlapped. Some autophagic vacuoles seemed to contain secretory granule matrix. It is suggested that the concept of crinophagy can be extended to the corticotrophs, though the lysosomal system may be involved in the specific function of this cell type by other mechanisms as well.

    Notes: Summary The involvement of lysosomes in ACTH and prolactin secretion was studied. Lysosomes were visualized in the anterior pituitary by their non-specific esterase (gold thioacetic acid technique) or acid phosphatase (Gomori technique) activity. Corticotrophs and mammotrophs were identified by postembedding immunocytochemistry for their respective hormones. Corticotrophs were rendered hypersecretory by bilateral adrenalectomy (7 or 12 days prior to examination), hyposecretory by dexamethasone administration. Prolactin secretion was enhanced by 17-beta-estradiol, prolactin release was inhibited by bromoergocriptine administration. Long-term hypersecretion of ACTH was accompanied by the presence of numerous autophagic vacuoles often containing secretory granules in the corticotrophs. Lysosomal enzyme-containing tubules and small lysosomes were abundant in the cytoplasm near the cell membrane, among the mature secretory granules. Feed-back inhibition of ACTH release by dexamethasone resulted in the extension of enzyme-containing tubules, continuous with cisternae and small lysosomes anywhere in the cytoplasm and in the appearance of numerous crinophagic vacuoles. A higher frequency of tubular lysosomes was described at the periphery of mammotrophs stimulated by 17-beta-estradiol. Bromoergocriptine caused a high incidence of characteristic crinophagic vacuoles in the prolactin cells. The concept of crinophagy has been extended to the corticotrophs. Morphological phenomena were attributed to the traffic and increased turnover of membranes, ligands and cytoplasmic organelles during stimulated secretion.

    Notes: Summary Cultured cells from adult rat anterior pituitaries or intermediate lobes were treated with the proteinase inhibitor tripeptide aldehydes BOC-DPhe-Pro-Arg-H (Boc-tPRH) and DPhe-Pro-Arg-H (fPRH), ovine corticotropin-releasing factor (oCRF), and bromocriptine. One millimolar fPRH stimulated basal, and slightly enhanced oCRF-induced ACTH release by melanotrophs in short-term experiments. The basal release of alpha-MSH was also stimulated by the drug. In long-term experiments, fPRH elevated markedly both the release and the intracellular level of ACTH; BOC-fPRH caused an increased alpha-MSH release. Tritiated fPRH had no preference for POMC-producing cells and BOC-fPRH or fPRH were harmless to the cell morphology. In anterior pituitary cell cultures, fPRH diminished slightly basal and oCRF-induced ACTH release. Bromocriptine was ineffective on corticotrophs, however, in melanotrophs it inhibited ACTH release markedly with or without fPRH in the medium. The dissimilar responsiveness of the corticotrophs and melanotrophs to the peptide aldehydes may be interpreted in terms of their differing membrane receptors or intracellular mechanism of stimulus-secretion coupling.

    Notes: Summary Ca-dependent ATPase activity in the rat anterior pituitary was demonstrated in 50-μm tissue slices of aldehyde-fixed tissue with the medium of Takano et al. (Cell Tissue Res. 243:91. 1986). — The outer surface of the plasma membrane of the parenchymal as well as the folliculo-stellate cells was lined with lead precipitate. The reaction deposit was particularly well localized in intercellular spaces both between two parenchymal cells, and between a parenchymal and a folliculo-stellate cell. A fine reaction deposit was also seen in the endoplasmic reticulum and Golgi apparatus of some parenchymal cells. Elimination of Ca2+ from the tissue and the substrate medium drastically reduced the amount of reaction product. If ATP was omitted or replaced by sodium β-glycerophosphate, no reaction product was seen. Changing the Ca2+ concentration or addition of Mg2+ to the standard medium caused a decrease in reaction intensity. Substitution of Mg2+ for Ca2+ resulted again in well-localized lead deposition which we attribute to the activity of another enzyme. We suggest that the activity we described in the membrane of glandular cells may correspond to the enzyme involved in the long-term regulation of intracellular Ca2+ level.

    Notes: Summary Cell suspensions were prepared from adult rat pituitary intermediate lobes and grown in medium TC 199 supplemented with foetal calf serum. 7 or 8 days old cultures were pulse labelled with tritiated thymidine and thereafter processed for immunostaining using an antiserum against a synthetic α-1-28-ACTH-analogue. Immunreactivity was mainly confined to epithelial cells, but some spindle-shaped cells were also immunpositive. Thymidine incorporation was observed in some immunopositive cells which shows that cells committed to hormone production may enter the mitotic cycle.
