Notizen: Previous studies showed that the intermediate level of resistance in bread wheat line ‘VPM1’ to pathotype Ha12 of the cereal cyst nematode could be conferred by an Aegilops ventricosa-derived gene, CreX, in chromosome arm 2AS, which also carries the rust resistance genes Yrl7, Lr37 and Sr38. Near isogenic lines (NILs) differing for the presence and absence of the Ae. ventricosa-derived linked genes Yrl7/Lr37/Sr38 were tested with cereal cyst nematode. Lines carrying Yr17 produced significantly fewer nematode cysts than the controls. An infested soil experiment produced better differentiation among resistant and susceptible genotypes. Susceptibility of ‘Trident’ indicated that linkage between CreX and Yr17 is incomplete. Microsatellite markers did not differentiate between ‘Trident’ and CreX-carrying genotypes. However, Xgwm636 (104) was associated with the presence of Yr17 in all six genetic backgrounds. Since none of the reported cereal cyst nematode resistance genes is located in chromosome 2AS, CreX was designated as Cre5.

Notizen: Monosomic analysis indicated that a seedling leaf rust resistance gene present in the Australian wheat cultivar ‘Harrier’(tentatively designated LrH) is located on chromosome 2A. LrH segregated independently of the stripe rust resistance gene Yr1 located in the long arm of that chromosome, but failed to recombine with Lr17 located in the short arm. LrH was therefore designated Lr17b and the allele formerly known as Lr17 was redesignated as Lr17a. The genes Lr17b and Lr37 showed close repulsion linkage. Tests of allelism indicated that Lr1 7b is also present in the English wheats ‘Dwarf A’(‘Hobbit Sib’), ‘Maris Fundin’ and ‘Norman’. Virulence for Lr17b occurs in Australia, and pathogenicity studies have also demonstrated virulence in many western European isolates of the leaf rust pathogen. Despite this, it is possible that the gene may be of value in some regions if used in combination with other leaf rust resistance genes.

Notizen: Long-term resistance to rust diseases depends on the identification and use of durable resistance sources or on the continuing use of new resistances and combinations of genes for specific resistance. These studies include four Australian wheats with intermediate, but inadequate levels of resistance and a French wheat ‘Hybride-de-Bersée’ (‘Bersee’), with reputed durable resistance to stripe rust. Studies of F2 and F3 populations from crosses with the susceptible ‘Avocet’ indicated that intermediate levels of adult plant stripe rust resistance in cultivars ‘Harrier’, ‘Flinders’ and ‘M2435’ were inherited monogenically, whereas King possessed two genes for resistance. Cultivars Harrier and M2435 possessed the same gene. Similarly, cvs. King and Flinders carried a gene in common. Like ‘Harrier’ and ‘M2435’, ‘King’ and ‘Flinders’ share common parents. The higher level of resistance in ‘Bersee’ was controlled by four genes. This conclusion was based on conventional genetic analysis, tests on F2-derived F7 single-seed descent lines and testcross progenies.

Notizen: Abstract  A detailed RFLP map was constructed of the distal end of the short arm of chromosome 1D of Aegilops tauschii and wheat. At least two unrelated resistance-gene analogs (RGAs) mapped close to known leaf rust resistance genes (Lr21 and Lr40) located distal to seed storage protein genes on chromosome 1DS. One of the two RGA clones, which was previously shown to be part of a candidate gene for stripe rust resistance (Yr10) located within the homoeologous region on 1BS, identified at least three gene family members on chromosome 1DS of Ae. tauschii. One of the gene members co-segregated with the leaf rust resistance genes, Lr21 and Lr40, in Ae. tauschii and wheat segregating families. Hence, a RGA clone derived from a candidate gene for stripe rust resistance located on chromosome 1BS detected candidate genes for leaf rust resistance located in the corresponding region on 1DS of wheat.

Notizen: Summary The expression of rust resistances conferred by closely linked genes derived from VPM1 varied with environmental conditions and with genetic backgrounds. Under low light and low temperature conditions seedlings carrying Yr17 showed susceptible responses. Stem rust and leaf rust resistance genes Sr38 and Lr37 tended to confer more resistance at 17±2° C than at normal temperatures above 〉 20° C. These studies supported the hypothesis that Yr17, Lr37 and Sr38 were derived from Aegilops ventricosa, whereas Pm4b was probably derived from T. persicum. Studies on certain addition lines and parental stocks indicated that wheat cytoplasm may enhance the expression of Sr38.
