    Notes: Previous studies showed that the intermediate level of resistance in bread wheat line ‘VPM1’ to pathotype Ha12 of the cereal cyst nematode could be conferred by an Aegilops ventricosa-derived gene, CreX, in chromosome arm 2AS, which also carries the rust resistance genes Yrl7, Lr37 and Sr38. Near isogenic lines (NILs) differing for the presence and absence of the Ae. ventricosa-derived linked genes Yrl7/Lr37/Sr38 were tested with cereal cyst nematode. Lines carrying Yr17 produced significantly fewer nematode cysts than the controls. An infested soil experiment produced better differentiation among resistant and susceptible genotypes. Susceptibility of ‘Trident’ indicated that linkage between CreX and Yr17 is incomplete. Microsatellite markers did not differentiate between ‘Trident’ and CreX-carrying genotypes. However, Xgwm636 (104) was associated with the presence of Yr17 in all six genetic backgrounds. Since none of the reported cereal cyst nematode resistance genes is located in chromosome 2AS, CreX was designated as Cre5.

    Notes: Root-knot nematode is a significant root-parasite of common wheat. A dominant gene Rkn-mnl for resistance was transferred into wheat from Acgilops variabilis. This gene was shown to be on chromosome 3B. Further analysis indicated that it is on the long arm of this chromosome and independent of the centromere.

    Notes: Aegilops variabilis no. 1 is the only known source of resistance to the root-knot nematode Meloidogyne naasi in wheat. Previous studies showed that a dominant gene, Rkn-mn1, was transferred to a wheat translocation line from the donor Ae. variabilis. Random amplified polymorphic DNA (RAPD) analysis was performed on the wheat cultivar ‘Lutin’, on Ae. variabilis, on a resistant disomic addition line and on a resistant translocation line. For genetic and molecular studies, 114-117 BC3F2 plants and F3-derived families were tested. Five DNA and one isozyme marker were linked to Rkn-mn1. Three RAPD markers flanking the Rkn-mn1 locus were mapped at 0 cM (OpY16-1065), 0.8 cM (OpB12-1320) and 1.7 cM (OpN20-1235), respectively. Since the Rkn-mn1 gene remained effective, its introduction into different wheat cultivars by marker-assisted selection is suggested.

    Notes: A previous RFLP analysis showed that the Aegilops ventricosa chromosome 6MV which compensates for the absence of 6D in 6MV (6D) wheat substitution lines was a 2/6 translocated chromosome, either 2S–6S.6L or 2S–6L.6S. The distal part of its long arm consists of a translocated segment belonging to homoeologous group 2. Chromosome 6MV carries a gene(s) for resistance to cereal cyst nematode. In order to define the part of 6MV (2S or 6S or 6L) involved in this resistance, addition lines with a 6MV deleted either for its short arm or for the distal part of its long arm were evaluated. It was shown that the gene(s) is carried by the group 2 translocated segment. The hypothesis that the gene(s) could be allelic to Cre2, another gene conferring resistance to the nematode introduced into the wheat complement from Ae. ventricosa is discussed.

    Notes: Moderate resistance to eyespot was first incorporated in the variety Cappelle-Desprez (CD). Later the gene Pch1, which could confer a higher level of resistance, was introduced from Aegilops ventricosa. However Pch1-carrying varieties can sustain significant eyespot-induced yield losses in severe attack situations. A strategy to further enhance the resistance of wheat is by pyramiding Pch1 and the genes for resistance in CD. The first requirement to achieve this is a better understanding of the genetics of resistance in CD. The resistance of the 21 Cappelle-Desprez (Bezostaya) disomic substitution lines was evaluated. Chromosome 7A was confirmed as carrying a major gene for resistance to eyespot at the seedling stage. However, this study demonstrates that this chromosome has no effects at the adult stage. Chromosome 5A was shown to carry a major gene for resistance to eyespot at the adult stage, which was stably expressed each year of testing. Chromosomes 1A and 2B had significant effects for only two years among four.

    Notes: Hybrids of Aegilops variabilis (2n = 28, UUSvSv) with the low-pairing type lines of Ae. longissima, Ae. bicornis, Ae. searsii, Ae. speltoides and Ae. umbellulata were obtained. This is the first report of successful hybridization between Ae. variabilis and Ae. searsii. Meiotic analysis of these hybrids showed that the U genome of Ae. variabilis donated by Ae. umbelltilata remained nearly unchanged and Ae. longissima is the possible donor species of the S genome to Ae. variabilis. But the Sv genome and the S1 genome of Ae. longissima are not completely homologous and are structurally differentiated by at least one interchange. According to their levels of homology with Sv through the means of chromosome associations, the four genomes of Sitopsis section species can be classified as follow: Sv≃ S1 〉 Sb 〉 Ss 〉 S. The meiotic behaviour of the hybrids between the wheat ‘Chinese Spring’ and the four Sitopsis section species studied has not supported the view that one of the four species is the donor of the B genome of wheat.

    Notes: Summary The chromosomes of the tetraploid wheats Triticum timopheevi (Genome AAGG) and T. araraticum (Genome AAGG) were C-banded at mitosis. The identity of the banded and unbanded chromosomes was then established by firstly making comparisons with the hexaploid species T. zhukovskyi which has the genome formula AAAAGG. Secondly, the meiotic pairing in F1 hybrids between T. timopheevi and diploid wheats was examined by means of C-banding. The results showed that the banded chromosomes belonged to the G genome, while the unbanded chromosomes belonged to the A genome. Only one of the two pairs of satellited chromosomes had strong heterochromatic bands. The relationship between the genomes of T. timopheevi and T. dicoccum (Genome AABB) was then assessed at meiosis in hybrids between these species, using the techniques of C-banding and in situ hybridisation of a cloned ribosomal RNA gene probe. It was concluded that there were differences both in the amount and distribution of heterochromatin and also translocation differences between the species.

    Notes: Abstract A non-radioactive digoxigenin-labelled DNA method was used successfully to identify RFLP markers in 54 Triticum aestivum cv ‘Chinese Spring’ — Agropyron cristatum (2n=28, genome PPPP) P-genome monosomic addition lines. Southern analysis using a set of 14 DNA probes identifying each homoeologous chromosome arm, combined with two restriction enzymes HindIII and EcoRI, indicated that six A. cristatum chromosomes (1P, 2P, 3P, 4P, 5P and 6P) and five A. cristatum chromosome arms (2PS, 2PL, 5PL, 6PS and 6PL) have been individually added to the wheat genome. The added chromosomes of three lines were Agropyron translocated chromosomes. It was also found that two addition plants possessed an Agropyron-wheat translocation. These results showed that RFLP analysis using the set of assigned wheat probes was a powerful tool in detecting and establishing homoeology of alien A. cristatum chromosomes, or arms, added to wheat, as well as in screening the alien addition material. The creation of the monosomic addition lines should be useful for the transfer of disease-resistance genes from A. cristatum to wheat.

    Notes: Abstract RFLP analysis has been used to characterise XMv, a chromosome of Aegilops ventricosa present in a disomic addition line of wheat. This chromosome is known to carry a major gene conferring resistance to leaf rust (Lr). The analysis demonstrated that XMv is translocated with respect to the standard wheat genome, and consists of a segment of the short arm of homoeologous group 2 attached to a group 6 chromosome lacking a distal part of the short arm. Lr was located to the region of XMv with homoeology to 2S by analysis of a leaf rust-susceptible deletion line that was found to lack the entire 2S segment. Confirmation and refinement of the location of Lr was obtained by analysis of a spontaneous resistant translocation in which a small part of XMv had been transferred to wheat chromosome 2A.

    Notes: Summary Intergeneric hybrids between Triticum aestivum cv ‘Chinese Spring’ and Agropyron cristatum 4x (2n= 5x=35, ABDPP genomes) with a high level of homoeologous meiotic pairing between the wheat chromosomes were backcrossed 3 times to wheat. Pollination of the F1 hybrid with ‘Chinese Spring’ resulted in 22 BC1 seeds with an average seed set of 1.52%. Five BC1 plants with 39–41 chromosomes were raised using embryo rescue techniques. Chromosome pairing in the BC1 was characterized by a high frequency of multivalent associations, but in spite of this there was no evidence of homoeologous pairing between chromosomes of wheat and those of Agropyron. All of the plants were self sterile. The embryo rescue technique was again essential to produce 39 BC2 plants with chromosome numbers ranging from 37 to 67. The phenomenon of meiotic non-reduction was also observed in the BC3 progenies. In this generation male and female fertility greatly increased, and meiotic pairing was fairly regular. Some monosomic (2n=43) and double monosomic (2n=44) lines were produced. Analysis of these progenies should permit the extraction of the seven possible wheat-Agropyron disomic addition lines including those with the added chromosomes carrying the genes involved in meiotic non-reduction and in suppression of Ph activity.
