    Notes: We investigated if IRFI 042, an analog of vitamin E, protects the brain against oxidative stress induced by intraperitoneal administration of Kainic acid (KA) (10 mg/kg); sham brain injury rats were used as controls. Animals received either IRFI 042 (20 mg/kg) or its vehicle 30 min before KA injection and after 6 h were sacrificed to measure malonildyaldheide (MDA) and glutathione levels (GSH) in the diencephalon. Behavioral changes were also monitored. Intraperitoneal administration of IRFI decreased MDA (micromol/g wet tissue: KA + vehicle = 22.5 ± 4.2; KA + IRFI = 17.1 ± 1; P 〈 0.005) and prevented GSH loss (nmol/g wet tissue: KA + vehicle = 0.41 ± 0.1; KA + IRFI = 1.86 ± 0.2; P 〈 0.005) in the diencephalon. The latency of occurrence of behavioral signs increased from 39 ± 1 to 62 ± 6 min in IRFI 042 group. The data suggest that IRFI 042 might protect against KA-induced oxidative stress.

    Notes: SDS-sedimentation volume (SV) is a biochemical index widely used to evaluate flour quality in durum and bread wheats. Significant association between SV and endosperm proteins (gliadin, high-molecular-weight- and low-molecular-weight-glutenin subunits) have been reported. Protein loci, however, account for only a portion of the total genetic variability. The objective of this study was to identify and locate quantitative trait loci (QTLs) associated with SV in a set of recombinant inbred (RI) lines, derived from a cross between the cv.‘Messapia’ of durum wheat and the accession MG4343 of the var. dicoccoides, and characterized for 259 genetic and molecular (RFLP) markers. Significant differences were detected for the quality index in the six environments examined, while the pattern of variability was that of a quantitative trait. Regression analysis of marker loci and sedimentation volume indicated, as expected, that chromosome 1B, on which are located the Gli-B 1/Glu-B 3 loci for some gliadin and glutenin subunits, is important for wheat quality. Two additional regions located on chromosomes 6AL and 7BS, and four regions on 1AL, 3AS, 3BL and 5AL, were shown to have single-factor effects on sedimentation volume at P 〈 0.001 and P 〈 0.01, respectively. Positive effects were contributed by both parents. A multiple linear regression model consisting of seven significant loci on different chromosomes explained 62–91% of the genotypic variation of the trait. The availability of linked markers to QTLs may facilitate the genetic dissection of quantitative traits and the early selection in wheat breeding programmes.

    Notes: Summary Toluene exposure was studied in 20 workers employed in painting and hand-finishing in an art furniture factory. Toluene was determined in the environmental air of places of work and in the alveolar air and blood of the workers. Hippuric acid and cresols were also tested in the workers' urine. Blood and urine tests were carried out before the work shift on Monday and Friday morning and at the end of the work shift on Friday afternoon. The other tests were performed on Friday afternoon only. Alveolar toluene concentrations, which were significantly correlated with environmental toluene concentrations (r= 0.6230; P 〈 0.01), corresponded to 19.4% of the toluene concentration in the atmosphere. Blood toluene was also found in painters on Monday morning and was significantly correlated with the other parameters. On Friday afternoon it was three times higher than the environmental toluene concentration. Urinary o-Cresol was highly correlated with toluene in the atmosphere, in blood and with hippuric acid in urine. On the basis of the slope of the regression line the ratio between urinary o-Cresol and blood toluene concentration was 0.99. At the end of the work shift urinary hippuric acid concentration was highly correlated with o-Cresoluria and with toluene in blood and in the atmosphere.

    Notes: Summary Exposure to toluene was studied in a group of 14 subjects working in a printing industry, who were exposed to this solvent only. Environmental monitoring was carried out using personal samplers for the whole workshift over three consecutive days. Toluene TWA concentrations ranged from 37 to 229 mg/m3. At the end of the workshift on each day of investigation, urine samples were collected for the determination of hippuric acid and ortho-cresol. Hippuric acid was also determined for urine before the workshift and on the Saturday and Monday mornings after the end of exposure; hippuric acid was also determined in 16 controls over the same five-day period. At the end of the workshift, hippuricuria levels in exposed workers always turned out to be statistically different from pre-workshift levels and those of the controls. The end-of-workshift hippuricuria levels of exposed workers were significantly correlated with the mean daily environmental concentration (TWA): in the three days of comparative study, we found r = 0.63 (P 〈 0.05) on Day 1, r = 0.90 (P 〈 0.001) on Day 2, and r = 0.87 (P 〈 0.001) on Day 3. Orthocresol turned out to be correlated with daily exposure less significantly than hippuric acid: r = 0.49 (n.s.) on Day 1; r = 0.78 (P 〈 0.001) on Day 2, and r = 0.65 (P 〈 0.05) on Day 3. Using all available data (41 observations), a very significant correlation (P 〈 0.001) was found between the TWA and both metabolites (r = 0.80 for hippuric acid; r = 0.68 for o-cresol). The values of the two metabolites in the end-of-workshift urine samples (41 observations) also turned out to be well correlated (r = 0.70; P 〈 0.001). The authors conclude that hippuric acid is a valid test for evaluating even low exposures to toluene.

    Notes: Abstract  A genetic linkage map of tetraploid wheat [Triticum turgidum (L.) Thell.] was constructed using segregation data from a population of 65 recombinant inbred lines (RILs) derived from a cross between the durum wheat cultivar Messapia and accession MG4343 of T. turgidum (L.) Thell. ssp dicoccoides (Korn.) Thell. A total of 259 loci were analysed, including 244 restriction fragment length polymorphisms (RFLPs), one PCR (polymerase chain reaction) marker (a sequence coding for a LMW (low-molecular-weight) glutenin subunit gene located at the Glu-B3 locus), seven biochemical (six seed-storage protein loci and one isozyme locus) and seven morphological markers. A total of 213 loci were mapped at a LOD≥3 on all 14 chromosomes of the A and B genomes. The total length of the map is 1352 cM and the average distance between adjacent markers is 6.3 cM. Forty six loci could not be mapped at a LOD≥3. A fraction (18.6%) of the markers deviated significantly from the expected Mendelian ratios; clusters of loci showing distorted segregation were found on chromosomes 1B, 3AL, 4AL, 6AL and 7AL. The durum wheat map was compared with the published maps of bread wheat using several common RFLP markers and general features are discussed. The markers detected the known structural rearrangements involving chromosomes 4A, 5A and 7B as well as the translocation between 2B-6B, but not the deletion on 2BS. This map provides a useful tool for analysing and breeding economically important quantitative traits and for marker-assisted selection, as well as for studies of genome organisation in small grain cereal species.
