    Notes: Optical properties of poly(phenylene pyridine) derivatives, poly(2,5-dialkoxy-1,4phenylenealt-2,5-pyridine) (PHOnPY25), such as optical absorption, photoluminescence (PL), and electroluminescence (EL), have been studied. The electronic energy structures of PHOnPY25 have been determined by optical and electrochemical measurements. Strong PL with high quantum efficiency has been clarified in the films, and spectral narrowing has been observed by pulse excitation of a nitrogen laser. Intense blue EL has also been demonstrated. © 2000 American Institute of Physics.

    Notes: Periodic multilayer structures of poly(p-phenylene vinylene)s have been fabricated by a self-assembly method on flat surfaces and round surfaces of optical fibers. Alternating multilayers consisting of poly{1,4-[2-(5-carboxypentyloxy)-5-methoxyphenylene]vinylene} (CPMOPPV) and poly(p-phenylene vinylene) (PPV) were adsorbed onto the positively charged substrates. The optical properties of the periodic multilayer structures of CPMOPPV/PPV have been studied. Periodic multilayers with microring geometry have also been fabricated around the quartz fibers. Their optical properties have been studied, and yellow electroluminescence from a light-emitting device with microring geometry has been observed. © 2000 American Institute of Physics.

    Notes: Background Prominent infiltration of eosinophils in airway mucosa is the pathognomonic sign of asthma. The role of airway epithelial cells in eosinophil infiltration, however, has not been fully elucidated.Objective The aim of this study is to develop a new in vitro transmigration system composed of airway epithelial cells and extracellular matrix, and to investigate the role of airway epithelial cells in eosinophil infiltration.Methods A layer of type I collagen gel was formed in Netwell™, and BEAS-2B bronchial epithelial cells were cultured on the gel. Then the wells covered with epithelial monolayer were filled with medium, inverted, and new upper chambers were constructed on the gel side by applying a ring cap. After further incubation with or without exogenous cytokines for 48 h, eosinophils or neutrophils were loaded in upper chambers (the gel side) and cells transmigrated to lower chambers (the epithelial cell side) were counted. Immunohistochemical analyses were also performed.Results While a simple collagen gel hardly promoted eosinophil migration even in the presence of eotaxin or RANTES, significant numbers of eosinophils migrated to lower chambers in the presence of the epithelial cells. Replacement of medium in the lower chamber (the epithelial cell side) with fresh medium, addition of exogenous eotaxin or RANTES in the upper chamber (the gel side), or pre-treatment of eosinophils with anti-CCR3 all inhibited transmigration. We found that the epithelial cells produced and deposited extracellular matrix proteins such as type IV collagen onto the type I collagen gel. Separately, we found that type IV collagen itself was capable of enhancing eotaxin-induced eosinophil migration in a standard chemotaxis assay. Neutrophils also efficiently migrated in the present transmigration system. Pre-treatment of epithelial cells with TNF-α and IL-4 enhanced eosinophil transmigration, while that of neutrophils was enhanced by TNF-α but suppressed by IL-4.Conclusion By utilizing a new in vitro transmigration system mimicking the airway mucosa, we have demonstrated that airway epithelial cells play an essential role in transmigration of eosinophils and that multiple factors such as chemokines, extracellular matrix proteins and exogenous inflammatory cytokines are involved in efficient transmigration.

    Notes: Solid cell nests of the thyroid gland were studied in 44 patients with medullary thyroid carcinoma. In 10 (22.7%) patients, solid cell nests were revealed in the vicinity of tumour foci (five cases) or in the contralateral thyroid lobe and isthmus (four cases); in one case the location was indeterminate. In all seven cases in which immunohistochemical studies were carried out, solid cell nests showed negative staining for thyroglobulin, calcitonin and chromogranin A, findings which were distinct from those in medullary thyroid carcinoma. It is therefore suggested that solid cell nests of the thyroid are not precursors of this tumour.

    Notes: Background : The attenuated anti-secretory activity of H2 receptor antagonists (H2RA) during continuous administration is referred to as the tolerance phenomenon. However, it is not clarified whether Helicobacter pylori infection affects the occurrence of tolerance to H2RA. It is also not clarified whether the tolerance phenomenon occurs to a new H2RA, lafutidine.Aim : To investigate the occurrence of the tolerance phenomenon in subjects with and without H. pylori infection during the continuous administration of lafutidine and famotidine.Subjects and Methods : Subjects were 20 healthy male volunteers (seven H. pylori positive and 13 H. pylori negative cases). All subjects were examined by ambulatory intragastric pH monitoring five times without medication, on the first and 15th day of the administration of 20 mg b.d. famotidine and 10 mg b.d. lafutidine in a cross-over fashion.Results : The tolerance phenomenon was not observed in H. pylori-positive subjects during the 15-day-long administration of both H2RAs. In contrast, the tolerance phenomenon was observed in H. pylori negative subjects, which has been previously reported.Conclusions : This study demonstrated that H. pylori infection affects the tolerance phenomenon during continuous administration of H2RAs.

    Notes: Small-to-moderate-angle X-ray scattering studies were performed on a troponin C solution prepared from rabbit skeletal muscle. The main feature of the scattering profile is the existence of two regions in the Guinier plot, in which data points lie on a straight line. The scattering profile was analyzed by way of a structural model with 'dumbbell' shape, based on the structure given by X-ray crystallography on troponin C. The dumbbell model shows a `quasi-' straight-line region at higher scattering angles in the Guinier plot, in addition to the intrinsic straight-line region at very small scattering angles. A ln[I(S)/S] versus S2 plot of the scattering profile at higher scattering angles gives information as to the average radius of gyration of the N-terminal and C-terminal domains of troponin C. The molecular parameters determined from small-angle X-ray scattering data of troponin C (10 mg ml−1), in the absence of Ca2+ ions, are as follows: the radius of gyration of the whole troponin C molecule is 20.4 Å and the average radius of gyration of the N- and C-terminal domains is 15.6 Å. From these radii of gyration, the center-to-center distance between the two domains is estimated to be about 26 Å.

    Notes: RIKEN beamline I (BL45XU) is an undulator beamline with two branches. One is for protein crystallography (PX) and the other is for small-angle x-ray scattering (SAXS). The beam is split into the two branches by a diamond monochromator so that two experiments can be done simultaneously [Yamamoto et al. (1995) Rev. Sci. Instrum. 66, 1833-1835]. The SAXS branch was designed for studying the weak interaction of proteins or subunits of fibrous or protein solutions especially using hydrostatic pressure. The optics makes use of the good parallelism of the undulator beam in order to reduce parasitic scattering. The beamline consists of a double crystal monochromator and a K-B type focusing mirror system. In order to cope with the high flux of the beam, an x-ray image intensifier (Hamamatsu Photonics, V5445P) with a cooled CCD camera (C4880-82) was used. As a result, decreases in both collection time and sample amount were realized in standard static experiments. These improvements will greatly facilitate SAXS experiments under high pressure.
