    Notes: Abstract Prompt and delayedγ-γ-coincidences were measured after slow neutron capture in185Re by use of Ge(Li)-detectors. New isomers were found at 99.4 keV (T1/2=27±7ns) and at ≈330keV (T1/2=17.4±0.7 ns). The evaluation of coincidence data established 10 rotational bands with band heads below 700 keV. The level scheme is discussed in detail: Nilsson assignments are proposed for the band configurations, and calculations of Coriolis mixing andγ-transition probabilities are presented which reproduce the experimental level energies and transition intensities very satisfactorily.

    Notes: Abstract Aims/hypothesis. The aim of this study was to examine the effect of weight loss on UCP2/UCP3 mRNA expression and UCP3 protein content in subjects with Type II (non-insulin-dependent) diabetes mellitus.¶Methods. We studied seven Type II diabetic subjects who followed a 10-week very low calorie diet. Expression of skeletal muscle UCP2 and UCP3 mRNA was measured using RT-competitive PCR and UCP3 protein content by western blotting, before and after the diet. Total and plasma fatty acid oxidation was measured using infusion of 13C labelled palmitate.¶Results. Body weight decreased from 105.5 ± 8.2 kg to 91.6 ± 7.2 kg (p 〈 0.001), after 10 weeks of diet intervention. Expression of UCP2 and UCP3 mRNA were significantly reduced after 10 weeks of diet (p 〈 0.05) but UCP3 protein contents were not significantly altered. Notably, the change in UCP3L mRNA expression and UCP3 protein content after the very low calorie diet were negatively associated with changes in body weight (r = – 0.97, p = 0.006 and r = – 0.83, p = 0.043, respectively) and BMI (r = – 0.99, p = 0.0007 and r = – 0.9, p = 0.016, respectively). Furthermore, changes in UCP3L mRNA expression and UCP3 protein content induced by the diet were positively correlated with changes in cytosolic fatty acid-binding protein content (r = 0.93, p = 0.023 and r = 0.84, p = 0.039, respectively). No correlation between diet-induced changes in UCP3 protein and resting energy expenditure or plasma non-esterified fatty acid concentrations were found.¶Conclusion/interpretation. The negative correlation between the change in UCP3 protein content after weight loss and the change in BMI, suggests that the decrease in UCP3 during weight loss could prevent further weight loss. The finding that the change in UCP3 protein content correlates with the change in skeletal muscle fatty acid-binding protein content, suggests a role for UCPs in the handling of lipids as a fuel.     Notes: Abstract The aim of this work was to study in the adult rat heart the effect of modifications of fatty acid (FA) supply on the content of cytoplasmic fatty acid-binding protein (H-FABPc). To modify the amount of circulating lipids, three different treatments were chosen: (i) an hypolipidemic treatment with Clofibrate, administered daily through a gastric tube at a dose of 250 mg/kg per day for one week, (ii) a continuous intravenous infusion of 20% Intralipid, a fat emulsion, for one week at a dose of 96 ml/kg per day, and (iii) a normobaric hypoxia exposure (pO2=10%) for three weeks. At the end of each treatment plasma lipids, myocardial H-FABPc content and the activities of three key enzymes (citrate synthase, CS, fructrose-6-phosphate kinase, FPK and hydroxy-acyl CoA-dehydrogenase, HAD) were assessed. With each of the three treatments a decrease of plasma cholesterol and phospholipid levels was observed. Plasma FA concentration increased with Intralipid infusion and decreased with chronic hypoxia. The heart H-FABPc content was increased by 20% with Clofibrate, decreased by 20% with chronic hypoxia and remained unaltered upon Intralipid treatment. The induced changes in H-FABPc content were not related directly to changes in plasma lipid levels. CS activity was slightly decreased in the hypoxia group, FPK activity decreased in the Clofibrate group, and HAD activity decreased in the Intralipid group. Among the various groups heart H-FABPc content was related to HAD activity. In conclusion, the H-FABPc content of adult rat heart appears responsive to changes in plasma lipid levels.

    Notes: Summary Heart tissue contains appreciable amounts of fatty acid-binding protein (FABP). FABP is thought to play a crucial role in the transport of fatty acids from the cellular membrane to the intracellular site of oxidation and also, in case of endothelial cells, in the transfer of fatty acids from the vascular to the interstitial compartment through the endothelial cytoplasm. The present study was designed to delineate a possible quantitative relationship between the capacity of different cell types in the heart to oxidize fatty acids and the presence of FABP. Palmitate oxidation capacity, measured in homogenates of cells isolated from adult rat hearts, was 2 nmol/min per mg tissue protein in freshly isolated cardiomyocytes (CMC), but only 0.09 and 0.31 nmol/min per mg tissue protein in cultivated endothelial (CEC) and fibroblast-like cells (CFLC), respectively. Palmitate oxidation rates were closely related to the cytochrome C oxidase activity and, hence, to the mitochondrial density in the cells under investigation. In CMC the content of cytosolic H-FABP (H-FABPc) was about 4.51 µg/mg tissue protein. However, in CEC and CFLC the FABP content was less than 0.01 and 0.004 µg/mg tissue protein, respectively, corresponding to at maximum 0.2% of the FABP content of CMC. These findings indicate a marked difference between CMC and non-myocytal cells in the heart regarding their capacity to oxidize fatty acids, and a marked disproportion between the fatty acid oxidation capacity and immunochemically determined FABP content in both CEC and CFLC. The functional implication of these observations remains to be elucidated.

    Notes: Abstract The effect was investigated of treadmill training of moderate intensity on the fatty acid-binding protein (FABP) content in relation to parameters of oxidative and glycolytic metabolism. To this end, the cytoplasmic FABP content and the activity of β-hydroxyacyl-coenzyme A dehydrogenase (HAD), citrate synthase (CS), and 6-phosphofructokinase (PFK) were measured in heart, fast-twitch extensor digitorum longus (EDL) and slow-twitch soleus muscles (SOL) of male Wistar rats. To investigate the influence of the amount of training (defined as the product of exercise duration, intensity and frequency), two training groups were created that differed in training frequency (HF, high frequency 5 days · week−1, n=9; LF, low frequency 2 days · week−1, n = 9; the exercise being 20 m · min−1 for 2 h with no gradient, over 6 weeks) and compared with SC, sedentary controls (n = 7). In heart muscle, the cytoplasmic FABP content was 34% higher in HF than in SC but was the same as in LF. The CS and HAD activities were no different in the three groups, suggesting that the capacity to oxidize fatty acids (FA) was not affected by training. The PFK activity was higher (43%) in HF, suggesting a shift towards carbohydrate utilization. The FABP content and HAD activity did not change in SOL and EDL after training whereas the CS activity increased (27%) in SOL and decreased (21%) in EDL in both training groups. In addition, PFK activity in EDL was much higher (113%) in the HF than in SC group. The HF training was associated with a fine-tuning of FA availability and use in heart muscle, and with a more efficient energy production. It is suggested therefore that cytoplasmic FABP could be an early marker of muscle adaptation to training in heart but not in skeletal muscle. The training reinforced the metabolic profile of the skeletal muscles, in particular that of the fast-twitch glycolytic muscle. We concluded that a large amount of training is needed when the effect on both oxidative and glycolytic parameters is to be studied.

    Description / Table of Contents: Zusammenfassung Ziel dieser offenen, randomisierten Multizenterstudie ist es, Nutzen und Wirtschaftlichkeit des Selbstmanagements der oralen Antikoagulation bei Patienten mit Vorhofflimmern (SMAAF-Studie) im Vergleich zur herkömmlichen beim Haus- oder Facharzt betreuten Patientengruppe zu untersuchen. Liegt eine Eignung zur Selbstkontrolle vor, werden 2000 Patienten randomisiert entweder dem Selbstmanagement oder der Kontrollgruppe zugeteilt. Als primäre Endpunkte während des zweijährigen Beobachtungszeitraums werden die Anzahl behandlungsbedürftiger thromboembolischer bzw. hämorrhagischer Komplikationen ermittelt. Sekundäre Zielparameter sind die Einhaltung der INR-Werte im individuellen Zielbereich, die INR-Varianz, der Zeitverlauf der Komplikation sowie die Kosteneffizienz des Selbstmanagements im Vergleich zum routinemäßigen Verfahren. Letztere schließt die durchgeführten diagnostischen und/oder therapeutischen Maßnahmen, die Krankenhausliegedauer sowie auch die sozialen Folgen mit ein (Anschlußheilbehandlung, Berufsunfähigkeit, Berentung). Die Fallzahlschätzung ergibt sich aus der Annahme, daß während dauerhafter Antikoagulation bei Patienten mit chronischem nichtvalvulärem Vorhofflimern im Rahmen der Primär- und Sekundärprävention vier schwere thromboembolische bzw. hämorrhagische Komplikationen pro 100 Patientenjahre auftreten. Da sich diese Rate durch das Selbstmanagement halbieren läßt, ist bei einer Power von 80% und einem Signifikanzniveau von 5% bei einem einseitigen χ2-Test zur Absicherung eine Stichprobengröße von n = 997 Patienten pro Gruppe erforderlich. Die Ergebnisse der SMAAF-Studie werden den sozio-ökonomischen Nutzen des Selbstmanagements bei Patienten mit chronischem nichtvalvulärem Vorhofflimmern ermitteln.
    Notes: Summary The objective of this open, randomized, multicenter study is to investigate the benefits and economic efficiency of self-management of oral anticoagulation in patients with atrial fibrillation (SMAAF study) in comparison with a group of patients given conventional care by a general practitioner or specialist. Two thousand patients suitable for self-management will be assigned at random to either the self-management group or the control group. The numbers of thromboembolic and hemorrhagic complications requiring treatment during the 2-year follow-up period will be recorded as the primary end point. The secondary endpoint variables will be maintenance of the INR value in the individual target range, INR variance, the course of complications over time, and the cost efficiency of self-management compared with the routine procedures. The last of these parameters will include the diagnostic and/or therapeutic measures carried out, the duration of inpatient hospital treatment, and the social consequences (subsequent rehabilitation treatment, inability to work, forced rentirement). The estimate of the required number of patients was based on the assumption that during long-term anticoagulant therapy within the framework of primary and secondary prevention 4% of patients with chronic non-valvular atrial fibrillation would have severe thromboembolic of hemorrhagic complications each year. Since this rate can be halved by self-management, a one-tailed χ2-test of 80% power and a 5% significance threshold would require n = 997 patients per group. The results of the SMAAF study will establish the socioeconomic benefits of selfmanagement in patients with non-valvular atrial fibrillation.

    Notes: Abstract The half-lives of the intrinsic states at 1,285.8 and 1,502.2 keV in184W have been measured in the decay of184Re m to be 8.33±0.18Μs and 2.35±0.10 ns, respectively. These half-lives determine absolute gamma-ray transition probabilities of eight transitions and yield upper limits for three transitions. The decay scheme of184Re m →184W is improved through measurements of delayed gamma-ray spectra with a Ge(Li) detector. Spin and Nilsson model interpretations are discussed.

The dissolution behavior was investigated and the residues analyzed by scanning electron microscopy and by ICP-MS and IDMS after dissolution.
