Purpose: Lipofuscin accumulates with age and in association with various retinal diseases. To investigate cellular effects of lipofuscin components in an in vitro RPE cell model, specific loading of the lysosomal compartment is required. Herein a major lipofuscin fluorophor was complexed to LDL and the subsequent subcellular localization of the retinoid was examined. Methods: The lipofuscin component N-retinylidene-N-retinylethanolamine (A2-E) was synthesized and coupled to LDL. Human RPE cell cultures were loaded with the A2-E/LDL complex over 4 weeks. Thereafter, RPE cells were harvested by trypsinization and disrupted by nitrogen cavitation. After ultracentrifugation, the postnuclear supernatant was fractionated on a self-generating gradient and fractions were analyzed by measuring marker enzyme activities of various cellular compartments. Results: A2-E accumulated almost exclusively in the lysosomal compartment, as indicated by the identical peaks of the marker enzyme ß-hexosaminidase and the relative fluorescence of A2-E. Only a small amount of A2-E appeared to associate with the cell membrane, as shown by a minor peak of A2-E corresponding to the distribution of phosphodiesterase activity. The lysosomal marker enzyme was not present in the cytosolic fraction. Conclusions: The feeding of A2-E/LDL complexes to cultured RPE-cells proved to be highly effective in specific loading of the lysosomal compartment, providing a suitable in vitro cell culture model for RPE aging and the investigation of A2-E-effects on lysosomal functions in RPE cells. Such a model may contribute to the understanding of the pathogenesis of degenerative diseases of the outer retina associated with excessive lipofuscin accumulation, including age-related macular degeneration, Stargardt's disease and Best's disease.

Hintergrund: Lipofuszin akkumuliert in postmitotischen retinalen Pigmentepithelzellen altersabhängig und im Rahmen zahlreicher Netzhauterkrankungen. Um einen möglichen Effekt von Lipofuszinbestandteilen auf die Zellfunktion und damit die pathophysiologische Bedeutung in vitro zu untersuchen, ist eine spezifische Beladung des lysosomalen Kompartiments erforderlich. Wir untersuchten, inwieweit dies mit einer Ankopplung des Lipofuszin-Fluorophors N-Retinyliden-N-Retinyläthanolamin (A2-E) an LDL-Partikel erreicht werden kann. Material und Methode: A2-E wurde aus Retinal und Äthanolamin synthetisiert und in einen Komplex mit LDL-Partikeln gebracht. Primäre humane RPE-Zellkulturen wurden mit dem A2-E/LDL-Komplex über 4 Wochen inkubiert. Anschließend wurden die Zellen mittels Stickstoff-Kavitation eröffnet und durch Ultrazentrifugation fraktioniert. Der postnukleäre Überstand wurde hinsichtlich der Markerenzymaktivitäten der verschiedenen zellulären Kompartimente analysiert. Ergebnisse: Im Percoll-Gradienten zeigten sich übereinstimmende Peaks des lysosomalen Markerenzyms ß-Hexosaminidase und der relativen A2-E-Fluoreszenz. Ein Anteil von A2-E korrespondierte mit der Verteilung der Phosphodiesteraseaktivität als Hinweis für eine in der Plasmamembran verbliebene geringe Menge von A2-E. Das lysosomale Markerenzym ß-Hexosaminidase trat nicht in das zytosolische Kompartiment aus. Schlußfolgerung: Durch die Kopplung von A2-E an LDL-Partikel ist eine effektive Einschleusung eines Retinoid-Bestandteils von Lipofuszin in das lysosomale Kompartiment humaner RPE-Zellen möglich. Das Verfahren kann somit für Untersuchungen zur Wirkung von A2-E auf pigmentepitheliale Zellfunktionen und damit für die Klärung der Frage hilfreich sein, inwieweit der Lipofuszinakkumulation im retinalen Pigmentepithel eine pathophysiologische Bedeutung bei Netzhauterkrankungen, die mit einer exzessiven Lipofuszinakkumulation einhergehen, einschließlich der altersabhängigen Makuladegeneration, zukommt. Die Befunde sprechen gegen eine Detergentienwirkung von A2-E auf die lysosomale Membran.
