    Notes: Abstract Basic fibroblast growth factor (bFGF) is a classical mitogen in fibroblasts and endothelial cells. Our previous studies have demonstrated that bFGF inhibits the growth of MCF-7 human breast cancer cells. The aim of the present study was to examine the effect of bFGF on cis-diamminedichloroplatinum(cisplatin)-induced cytotoxicity in MCF-7 breast cancer cells as compared to normal endothelial cells. MCF-7/NCF cells transduced with a vector expressing the bFGF gene and overexpressing its product, and MCF-7/N2 cells transduced with the backbone vector were incubated with a combination of bFGF and cisplatin for 5 days; results were compared with those obtained with bovine aortic endothelial cells. Cell proliferation was assessed with the sulforhodamine B colorimetric cytotoxicity assay. Apoptosis was quantitatively determined by flow-cytometric analysis for DNA damage and the apoptotic death assay for DNA fragmentation, and qualitatively by electron microscopy. Reverse transcriptase/polymerase chain reaction analysis and an enzyme immunoassay were used to determine the mRNA and protein level, respectively, of the anti-apoptotic bcl-2 gene product. We found that bFGF enhanced cisplatin-induced cytotoxicity in MCF-7 breast cancer sublines. bFGF enhanced proliferation of normal endothelial cells and did not increase cisplatin-induced cytotoxicity. This effect was accompanied by down-regulation of the anti-apoptotic protooncogene bcl-2 and the enhancement of cisplatin-induced apoptosis. We suggest that the improved understanding of the role of bFGF in the differential modulation of the response of breast cancer and normal endothelial cells to chemotherapy may enable active intervention to alter the therapeutic ratio favorably in breast cancer patients.

    Description / Table of Contents: Zusammenfassung Die Konzentration des schwangerschaftsspezifischen Proteins SP-1 im Fruchtwasser wurde bei normalen und pathologischen Graviditäten geprüft. Die SP-1 Konzentration im Fruchtwasser betrug bei normalen Schwangerschaften etwa ein Hundertstel der SP-1 Konzentration im maternellen Serum. Bei Fällen mit Diabetes mellitus, intrauterinem Fruchttod und Rhesusinkompatibilität fanden sich signifikant erhöhte SP-1 Konzentrationen im Fruchtwasser.
    Notes: Summary The concentration of the pregnancy specific protein SP-1 in amniotic fluid was determined in normal and pathologic pregnancies. SP-1 concentrations in amniotic fluid of normal pregnancies amounted to one percent of the maternal serum concentrations. In cases with diabetes mellitus intrauterine death and rhesus incompatibility SP-1 levels were increased significantly.

    Notes: Abstract Pentacaine was found to prevent the development of acute haemorrhagic lesions induced by ethanol in rats in a dose-dependent way. Electron microscopy in the untreated group showed extensive disruption of the surface epithelium and deep necrosis of the mucosa after ethanol exposure. Degranulation or even complete destruction of mast cells was observed. The microvasculature exhibited several signs of derangement. After pentacaine treatment, these signs were absent and no degranulation of mucosal mast cells was observed. The mast cell-mediated effect of pentacaine appears to be only one component of its gastroprotective action.

    Notes: Abstract The photodissociation iodine laser system PERUN I is described. The laser generates and amplifies the subnanosecond pulse with the energy about 80 J of the fundamental wavelength 1·315 μm and beam diameter 10 cm. The physical principles of this type of the high power laser are briefly explained and the laser system and its characteristics are described from the physical and technical point of view. Special attention is paid to the realization of such system with the slow optical pumping (≈300 μs).

    Description / Table of Contents: Summary Serum pregnancy specific protein (SP-1) levels were measured preoperatively in 26 women with suspected ectopic pregnancy. We used an enzyme immunoassay with a sensitivity of 2 ng/ml. SP-1 was detected in all 23 patients with a tubal pregnancy, but not in three nonpregnant women with ovarian cysts. Tests for urinary HCG with a sensitivity of 1,000 IU/l were negative in seven of 19 patients with an ectopic pregnancy.
    Notes: Zusammenfassung Mit Hilfe des Enzym-Immunoassays Enzygnost-SP-1 wurde die Serumkonzentration von SP-1 bei 26 Frauen, die unter der Verdachtsdiagnose Tubargravidität operiert wurden, praeoperativ bestimmt. Bei allen 23 Patientinnen mit verifizierter Tubargravidität war SP-1 im Serum nachweisbar, während bei drei nicht graviden Frauen mit Ovarialcysten die Werte von SP-1 unter 2 ng/ml lagen. Routinemäßig durchgeführte Schwangerschaftsteste im Harn hingegen waren in sieben von 19 Fällen mit Tubargravidität negativ. Offenbar kann durch die Messung der SP-1 Serumkonzentration bei Fällen mit Verdacht auf Extrauteringravidität die Diagnostik entscheidend verbessert werden.

    Notes: Abstract The cellular component of the fluid of female urethral expulsions was analysed in two healthy women (37-year-old multipara and 27-year-old nullipara) on days 1, 10, 12, 14, 26 and 27 of their menstrual cycle. Transmission and scanning electron microscopy were used to determine standard values of uroepithelial squamous cells of the fluid of female urethral expulsions on different days of the menstrual cycle. In agreement with the known changes of these cells found in cytological smears during the cycle, electron microscopy confirmed characteristic changes in the appearance of the fine cell structure and the space configuration of the squamous cells. Changes in the desmosomal junctions among squamous cells during the cycle were found. Striking adherence of bacteria and mucosubstances to the squamous cell surfaces were observed in the first half of the menstrual cycle and at the time of ovulation.

    Description / Table of Contents: Summary Measurement of baroreflex sensitivity is a new method to identify patients after myocardial infarction with a high risk for sudden cardiac death, ventricular tachycardia or ventricular fibrillation. In this retrospective study the baroreflex sensitivity was obtained noninvasively by measuring the systolic blood pressure with a FINAPRES-device and correlating this with the R-R intervals of the ECG after raising blood pressure with an intravenous dose of Norfenefrinhydrochlorid (Novadral). According to other investigators a correlation of 〉0.7 with a significance of p〈0.05 was recommended for evaluable results with a baroreflex sensitivity 〈3 ms/mm Hg being judged as decreased. We investigated 302 patients (mean age 59±17 years, 224 males, 78 females). 75% of the investigations showed acceptable results. In 77 cases (25%) reasonable results could not be achieved. We found premature ventricular beats to be responsible in 18 investigations (6% of all investigations). 41 (13.1%) of all investigations were not evaluable because of bad correlation for unknown reason. When we looked closely at these nonevaluable results, we found a significantly higher number of patients with impaired left ventricular ejection fraction (〈40%), diabetes or inducible sustained ventricular tachyarrhythmia in the electrophysiologic study in this group. During all investigations no severe side effects were observed. We conclude that the noninvasive measuring of the baroreflex sensitivity is a save method and leads to reasonable results in 75% of the investigations. In 13.1% it is not possible for unknown reason to achieve sufficiently correlating values. These measurements cannot be evaluated from nowadays' standards and have to be further investigated as they may indicate a population at high risk.
    Notes: Zusammenfassung Die Baroreflexsensitivitätsmessung ist eine neue Methode zur Indentifikation von Postinfarktpatienten mit einem hohen Mortalitätsrisiko bzw. mit hohem Risiko für den plötzlichen Herztod, Kammerflimmern oder Kammertachykardien. Für die hier geschilderte Untersuchung wurde die Baroreflexsensitivität nichtinvasiv gemessen. Intravenöse Gabe von Norfenefrinhydrochlorid (Novadral) erzielte einen Blutdruckanstieg von 15–40 mm Hg, und die Steigung der Korrelationsgeraden zum Herzfrequenzabfall, also zur Verlängerung der Sinuszykluslänge, wurde errechnet. Hierbei wurden Messungen mit einer Korrelation 〉0,7 (bei einem Signifikanzniveau p〈0,05) als auswertbar angesehen. Bei 302 Patienten (mittleres Alter 59±17 Jahre, 224 Männer, 78 Frauen) wurden Baroreflexuntersuchungen durchgeführt. Bei insgesamt 69 Patienten (29%) war die Baroreflexsensitivität (BRS) 〈3 ms/mm Hg und wurde als pathologisch gewertet. Bei 236/313 Untersuchungen (75%) war das Ergebnis verwertbar, wobei 14% der Untersuchungen Mehrfachmessungen benötigten. Bei 17 nicht auswertbaren Untersuchungen (25%) konnten in 18 Fällen (6% aller Untersuchungen) ventrikuläre Extrasystolen verantwortlich gemacht werden. 41 Untersuchungen (13,1% aller Untersuchungen) waren jedoch aus unklarer Ursache wegen mangelnder Korrelation nicht auswertbar. Eine Erklärung für die mangelnde Korrelation der Messung konnte bei diesen Patienten nicht gefunden werden. Diese 41 Patienten hatten signifikant häufiger eine linksventrikuläre Auswurffraktion (EF) 〈40%, einen Diabetes mellitus und eine induzierbare anhaltende Kammertachykardie oder Kammerflimmern bei der elektrophysiologischen Untersuchung als das Kollektiv der auswertbaren Patienten. Die nichtinvasive Baroreflexsensitivitätsmessung führt bei 75% der Untersuchungen zu verwertbaren Ergebnissen. 6% der Untersuchungen sind wegen ventrikulärer Extrasystolie nicht auszuwerten, 13,1% der Untersuchungen erzielen aus unklarer Ursache keine ausreichende Korrelation der Wertepaare und können somit nach bisherigen Standards nicht ausgewertet werden.

    Notes: In this paper mathematical and computational aspects of device modelling are treated. Four main subjects are discussed; the analytical model, the discretizations, the non-linear and the linear systems of equations. On the one hand the most commonly used numerical techniques are described; on the other hand several new techniques are presented. Noteworthy new techniques are the generalized upwind box scheme and the mixed variable approach. Aspects such as the choice of variables and the singularly perturbed nature of the problem are treated. Properties of the matrices involved are presented in a systematic way. Throughout the paper a standard one-dimensional diode is used to illustrate the concepts involved. Finally several realistic two-dimensional examples are given.
