    Notes: Abstract A combination of automated solid-phase extraction (SPE) and subsequent two-step derivatisation has been developed for the simultaneous analysis of basic drugs of abuse and cocaine metabolites in serum samples. Substances included in this procedure are morphine, codeine, methadone, cocaine, benzoylecgonine, methylecgonine, amphetamine, methamphetamine, MDMA, MDEA and MDA. SPE with mixed-mode cartridges (RP-C8 and cation-exchange) was fully automated with a Zymark RapidTrace SPE robot. GC/MS analysis was performed after derivatisation with a new two-step reaction by trifluoroacetic anhydride and 2,2,3,3,3-pentafluoropropanol. High recoveries (〉 85%) with high reproducibility ¶(CV 1.1–3.8%) were found for all drugs. High correlation coefficients (r 〉 0.998) were obtained due to the addition of deuterated standards prior to extraction. Experience obtained over 2 years of applying this method to drug analysis in serum is discussed.

    Notes: Abstract Spectrophotometric measurements were performed on intra- and/or subcutaneous bruises occurring in direct temporal connection with peracute fatal trauma. The purpose of these measurements was to determine whether the visual colour impression of a fresh traumatic extravasation can give information on the localisation of the haemorrhage in a certain tissue layer. After visual assessment of the colour of the bruise, the spectral reflectance curves and the CIE-L*a*b* colour values were determined with the help of a diode array spectrophotometer. The localisation and size of the haemorrhages in the cutis and/or subcutis were evaluated morphologically after incision of the skin. It was confirmed that there is a relationship between the colour impression and the localisation of the bruise. Bruises localised near the surface have a more reddish appearance while bruises in deeper layers give a more bluish colour impression. An explanation may be found in the optical characteristics of skin. Blood localised in the subcutis appears blue on the surface due to scattering processes in the dermis (Rayleigh scattering), as the blue wavelengths of the light are scattered (and thus reflected) to a greater extent than the red wavelengths.

    Description / Table of Contents: The authors report a rare case of double suicide of an elderly married couple who had both been veterinary surgeons before retirement. The suicide pact was carried out by the husband who first killed his wife, who suffered from inoperable cancer, by firing a contact shot to the top of her head and afterwards committed suicide in the same way. On the basis of this case report and the pertinent literature several criminalistic aspects of double suicides and of shots with entrance sites in the parietal region are discussed.
    Notes: Anhand des gemeinsam begangenen Suizids zweier betagter Eheleute – beide waren vor dem Ruhestand als Veterinärmediziner tätig – wird auf das seltene Vorkommen von Scheitelschüssen hingewiesen. Im gegenseitigen Einvernehmen tötete der männliche Partner zuerst seine krebskranke Frau und dann sich selbst durch je einen aufgesetzten Scheitelschuss. Unter Bezugnahme auf den referierten Fall werden kriminalistische Aspekte gemeinschaftlicher Selbsttötungen und suizidaler Scheitelschüsse diskutiert.

    Description / Table of Contents: Zusammenfassung Es wird über eine Spätruptur der Leber nach vorausgegangener Reanimation berichtet. Bei einer 27jä hrigen Frau kam es im Gefolge einer Lungenembolie zu einer Asystolie, die durch extrathorakale Herzmassage behoben wurde. Die dabei entstandenen Leberrupturen führten primär zur Ausbildung eines subkapsulären Haematoms, und nach 5 1/2 Stunden infolge sekundärer Kapselzerreißung zur inneren Verblutung. Als Zeichen der mehrstündigen Überlebenszeit fanden sich degenerative Veränderungen der rupturnahen Parenchymzellen und Fibrinabscheidungen auf der abgehobenen Leberkapsel. Die histologische Untersuchung der Lungen ergab eine Leberzellembolie und — trotz des Fehlens von Rippenoder Sternumfrakturen — eine Fett- und Knochenmarkembolie. Die erhobenen Befunde werden unter Bezugnahme auf einschlägige Literaturberichte diskutiert.
    Notes: Summary A delayed rupture of the liver after resuscitation is reported. In the case of a 27-year-old woman, pulmonary embolism was followed by asystolia, which was remedied by external cardiac massage. This procedure caused ruptures of the liver, which first led to the formation of a subcapsular hematoma and 5 1/2 hours later, due to a secondary rupture of the capsule, led to internal bleeding and death. Degenerative changes of the parenchyma cells in the vicinity of the rupture were recognized, as well as fibrin separations on the detached Glisson's capsule, as signs of a survival period of several hours. Histological investigation of the lungs revealed a hepatocellular embolism and—in the absence of rib or sternum fractures—fat and bone-marrow embolisms. The morphological findings are discussed with reference to pertinent reports in the literature.

    Description / Table of Contents: Zusammenfassung In mehreren Fällen von stumpfer Traumatisierung oder hochgradiger Brandeinwirkung fanden sich im venösen Blut des rechten Ventrikels großtropfige, mit freiem Auge erkennbare Fettsubstanzen. Durch dünnschichtchromatographische Auftrennung der Lipidextrakte wurde festgestellt, daß diese sichtbare Fettakkumulierung auf einer Zunahme der Triglyceride beruhte. Die Analyse der Fettsäuren mittels kombinierter GC/MS ergab eine Verschiebung zugunsten der Ölsäure. Zur näheren Charakterisierung der Triglyceride wurde deren Molekulargewichtsverteilung mit Hilfe der DCI-Massenspektrometrie bestimmt. Die Gesamtheit der Untersuchungsergebnisse sprach dafür, daß die im lipämischen Blut des rechten Ventrikels vorgefundenen Lipide überwiegend aus dem Depotfett stammen. Bei ausschließlich postmortaler Verbrennung (ohne vitale Traumatisierung) ist es — trotz massiver Lipämie des venösen Blutes — nicht zur Ausbildung einer kapillären Lungenfettembolie gekommen.
    Notes: Summary In several cases of fatal blunt-trauma injuries or after exposure to severe burning, large-drop fatty substances, discernible to the naked eye, were found in the venous blood of the right ventricle. Thin-layer chromatographic separation of the extracted lipids demonstrated an increase in the triglyceride fraction. Analysis of the fatty acids by means of combined gas chromatography/mass spectrometry showed a shift towards oleic acid. The distribution pattern of the molecular weights was determined by means of DCI-mass spectrometry in order to characterize the triglycerides in detail. The total chemical findings indicated that the visible lipids present in the right ventricle had predominantly derived from the adipose tissues. In cases of post mortem burning (without vital traumata) no typical pulmonary fat embolism had developed despite massive lipemia in the venous blood.

    Notes: Abstract In order to identify polymorphic positions and to determine their frequency in the human mitochondrial D-loop containing region, the mitochondrial DNA (mtDNA) control region of 200 unrelated individuals from Germany were amplified and directly sequenced. Sequence comparison led to the identification of 190 mitochondrial lineages as defined by 202 variable positions. The most frequently occurring lineage comprised 5 individuals, whereas 186 types of D-loop sequences were observed in only one individual. Of the sequences studied 7% are not unique but show at least one counterpart with an identical haplotype. The majority (61%) of the control regions investigated showed between four and eight nucleotide positions deviating from the reference sequence. The maximum number of deviations observed in a single control region was 18. The majority of the variable positions in the D-loop region (88%) are located within three hypervariable regions. Sequence variations are caused by nucleotide substitutions, insertions or deletions. As compared to insertions and deletions, nucleotide substitutions make up the vast majority of the mutations (90%). We have predominantly found transitions (75%) and a significantly lower frequency of transversions (15%) whereas insertions (6%) as well as deletions (4%) are rather rare. Upon sequencing the mitochondrial control region from 200 German Caucasians the genetic diversity was estimated at 0.99. The probability of two randomly selected individuals from a population having identical mtDNA types is 0.6%.

    Notes: Abstract Two cases are presented in order to emphasize the importance of mitochondrial DNA in forensic medicine. The first case involved a charred body which could not be identified by morphological means because of severe destruction of all tissues. The parallel use of PCR methods using genomic DNA and sequencing of the mitochondrial d-loop region produced unequivocal and reproducible results. In the second case, various parts of a highly decomposed body were investigated. The application of standard PCR methods for genomic DNA proved unsuitable to answer the question whether the body parts belonged to the same body. However, when sequencing of mitochondrial DNA segments amplified from tissue and bone samples was performed, clearly interpretable results were obtained.
