    Notes: The occurrence of neuropeptides was studied in neurofibromas of von Recklinghausen's disease by indirect immunofluorescence. All non-plexiform cutaneous neurofibromas contained abundant vasoactive intestinal polypeptide, peptide histidine–isoleucine and calcitonin gene-related peptide immunoreactive nerves. The nerves were small and unmyelinated. Neuropeptides might be responsible for itch that occurs especially in small cutaneous neurofibromas. Neuropeptides are also suggested to act as modulators and/or trophic factors for neurofibroma growth.

    Notes: Summary Neuropeptide Y (NPY)-like immunoreactive nerves were demonstrated in human cardiac muscle. The atrial specimens were obtained from open-heart surgery. The PAP method was applied for immunocytochemistry for light and electron microscopy. A dense, extensive network of NPY-like immunoreactive nerve fibres was seen between cardiac muscle cells and around blood vessels. In electron microscope PAP precipitates were localized in large dense-cored vesicles of 80–120 nm in size in separate nerve terminals or in the terminals situated in the nerve bundles. Close contacts were observed between NPY nerves and muscle cells and blood vessels. The possible functional role of NPY innervation in the human heart is discussed.

    Notes: Summary In a clinical series of patients undergoing open-heart surgery, capillary lesions in the myocardium were common when hypothermic chemical cardioplegia was used for myocardial protection during aortic cross-clamping. It seems obvious that the resulting oedematous swelling of the capillary wall, especially that of the endothelial cells, may constitute an anatomic intraluminal obstacle to myocardial reperfusion, thereby further delaying the recovery. After (mere) topical cooling, only minor capillary changes were observed. In contrast to the oedematous transformation described above for the former group, capillary endothelial cells and pericytes of the latter were diminished in cellular volume.

    Notes: Summary Norepinephrine-stimulated adenylate cyclase enzyme (EC 4.6.1.1) was histochemically demonstrated by electron microscope in the plasmalemmas of the newborn rat adipocytes. The precipitating agent in the histochemical reaction was cobalt salt. Metoprolol, a selective β 1-blocker given in vivo abolished almost totally the reaction products from the outer surfaces of the plasmalemmas. In vitro treatment did not yield constantly reproducible results. Prenalterol (H 133/22 —CGP 7760/B Hässle) a selective β-agonist given in vivo stimulated enzyme activity in the plasmalemmas. The reaction, however, showed different localization patterns and enhanced amounts of reaction products in the membranes. It is proposed that the histochemical demonstration of amine-stimulated adenylate cyclase activity in the adipocyte plasmalemmas corresponds to the hormonal receptor sites. The question of the sensitivity of the receptors is discussed.

    Notes: Summary Calcitonin gene-related peptide (CGRP)-like immunoreactivity was localized immunocytochemically in the large motoneurons in the ventral horn of rat spinal cord. Using fluorescence double-labelling substance P (SP)-immunoreactive nerve fibres were found to surround both the CGRP-positive and negative motoneurons, whereas enkephalin (ENK)-immunoreactive fibres surrounded mainly CGRP-negative cells. All CGRP-like immunoreactive motoneurons were also choline acetyltransferase (ChAT)- and acetylcholinesterase (AChE)-positive. On the other hand a large population of ChAT- and AChE-positive motoneurons were devoid of CGRP-immunoreactivity. It is probable that CGRP/ChAT/AChE-positive cells surrounded by SP-positive fibres have different functions in motoric nervous system than the CGRP-negative ChAT/AChE-positive cells, which are surrounded by ENK-immunoreactive fibres.

    Notes: Abstract The aim of the present study was to map immunohistochemically the distribution of the glucocorticoid receptor (GR) in rat skin. Nuclear GR-like immunoreactivity (LI) was found in both epidermis and dermis. In the epidermis, the basal cell layer showed an intense immunoreaction; the lower part of the spinous layer was also labelled. In the dermis, the fibroblasts as well as the sweat glands, sebocytes and adipocytes were GR-immunoreactive (IR). In the root sheath of the hair follicle the staining was most intensive in the outer layer. The endothelial cells comprising the smooth muscle cells of the blood vessels, as well as the arrector pili muscle, showed GR-LI. In the peripheral nerves, the immunoreaction was localized to the nuclei of the Schwann cells and in the perineurial fibroblasts. Mast cells did not show nuclear GR-LI. Based on our immunocytochemical findings that several cell types of the skin are GR-IR, the variable physiological and pharmacological effects of glucocorticoids are easier to understand.

    Notes: Summary The development of acetylcholinesterase (AChE) and non-specific cholinesterase (NsChE) activity was studied in the rat neostriatum by the light and electron microscope using three thiocholine methods. The AChE activity was first demonstrable only in the lateral parts of the nucleus, and during the early postnatal development the most intense activity was in the cell bodies, whilst the typical intense staining of the neuropil of adult animals was seen in two-week-old rats. Two types of AChE-containing cells were observed in the neostriatum of rats younger than two weeks and in cultures of newborn rat neostriatal cells. The neuropil of the cultures showed weak activity in the membranes of thin preterminal processes. In the neuropil of old rats, NsChE activity was present in the membranes of nerve cell processes. The capillary endothelial cells of newborn rats contained both AChE and NsChE. During subsequent development, the AChE activity disappeared, whilst for NsChE no change was seen in the distribution of activity seen in newborn or young adult rats less than three months old.

    Notes: Summary Cerium was applied for the ultrastructural, cytochemical localization of adenylate cyclase (EC 4.6.1.1.). The enzyme activity was stimulated with norepinephrine, prenalterol and choleratoxin in the brown fat cells of newborn rats. The final reaction product was observed in the plasmalemmas of the stimulated adipocytes. The precipitate was finely crystalline, easily visible in the electron microscope and in the X-ray microprobe analysis it yielded cerium and phosphate peaks, respectively. The use of cerium offers a new tool valid for the cytochemical localization of adenylate cyclase enzyme related to the membrane receptors.
