Summary Two groups of 50 and 20 rats respectively, of both sexes, were fed a diet containing 2.0% and 2.5% phenacetin respectively, for 3–27 weeks. An examination by light microscopy regularly showed an iron-positive and iron-negative pigment in the cells of the proximal renal tubulus. Interstitial nephritis or papillary necrosis has not been found. By electron microscopy the following were observed in the proximal tubular cells: electron-dense pigment corpuscles surrounded by a membrane and partly containing ferritin; large, very polymorphous myelin figures; vacuoles which included cytoplasmatic components, and a distinct increase of the smooth endoplasmatic reticulum. Taking into account the literature, these changes may be explained as follows: By a large storage of iron-positive and iron-negative pigment in phagolysosomes and by malnutrition, some areas of cytoplasma are damaged and then separated by a membrane. In these areas a “digestion” by lysosomal enzymes begins. In this way phospholipids become released which can, as a sequence of disorders of intra-cellular hydration, be transformed to myelin figures.

Zusammenfassung Je zwei Gruppen von 50 bzw. 20 Ratten beiderlei Geschlechts erhielten während 3–27 Wochen phenacetinhaltiges Futter (2,0% bzw. 2,5%). Lichtmikroskopisch findet sich in den Nieren regelmäßig im proximalen Tubulus ein eisenpositives sowie ein eisennegatives Pigment. Eine interstitielle Nephritis oder Papillennekrosen sind nicht nachzuweisen. Elektronenoptisch werden in der proximalen Tubuluszelle beobachtet: 1. Elektronendichte, von einer Membran umgebene Pigmentkörper, welche teilweise Ferritin enthalten; 2. große, sehr polymorphe Myelinfiguren; 3. Vacuolen, welche Cytoplasmabestandteile in sich einschließen; 4. eine deutliche Vermehrung des glatten endoplasmatischen Reticulums. Unter Berücksichtigung der Literatur erscheint folgende Deutung der Veränderungen möglich: Durch die massive Speicherung von eisenpositivem und eisennegativem Pigment in Phagolysosomen und durch die Mangelernährung werden einzelne Cytoplasmabezirke geschädigt und sodann durch eine Membran abgegrenzt. Innerhalb dieser Bezirke setzt eine „Verdauung“ durch lysosomale Enzymsysteme ein. Dadurch werden Phospholipide frei, welche sich als Folge von intracellulären Hydratationsstörungen zu Myelinfiguren umformen können.
