    Notes: A Bragg spectrometer for monitoring impurity radiation of hydrogenlike carbon and oxygen at the tokamak ASDEX Upgrade is presented. The use of fast multistrip proportional counters allows soft x-ray photon counting with count rates above 1 MHz in a simple arrangement. The absolute calibration was performed using a large area x-ray tube with a LiF anode. The concentrations of both impurities are calculated using the coronal model both for ionization and electron impact excitation. The results of the measurements concerning the long term behavior before and after conditioning of the torus walls by boronization as well as the fast plasma processes are presented. © 1996 American Institute of Physics.

    Notes: A new low noise miniaturized multichannel bolometer module for absolute measurements in the VUV and soft x spectral ranges is described. Highly integrated four-channel modules (2×3.3×1.5 cm3) each comprising four independent ac-excited (50 kHz) metal resistor bolometer bridges were successfully tested on a large tokamak (Tore Supra in Cadarache) and on an electron synchrotron (BESSY in Berlin). The bolometer system features a linear response to the absorbed radiation power, a low detection limit (≤1.0×10−6 W cm−2 on Tore Supra with an integration time of τint=10×10−3 s) and a low NEP (≤10×10−9 W on BESSY). The thermal cross-talk between adjacent detectors is negligible (〈0.003) and a low thermal drift ΔUBr/ΔT 〈 10−4 V °C−1 is achieved. It can be operated at a maximum temperature of 150 °C, at high magnetic fields (tested up to B=4.5 T in the laboratory) and survives high nuclear radiation doses. The system offers the possibility of detecting low-power VUV and soft x-radiation with sampling rates of up to 10 kHz on plasma machines and of absolutely calibrating VUV and soft x instruments. Effective suppression of electric, thermal and nuclear radiation interferences is characteristic of the bolometer system.Strain gauge effects, which could affect the behavior of the bolometers at high magnetic fields, are suppressed by the ac-excitation technique.

    Notes: The arrays of silicon diodes recently installed on the Joint European Torus (JET) measure the plasma soft x-ray emission with high temporal and spatial resolution, allowing magnetohydrodynamic (MHD) effects to be investigated. The diagnostic consists of two "pinhole'' cameras which view the same poloidal cross section in orthogonal directions. A data-acquisition system based upon three cascaded digital filters can convert the 200-kHz detector signal analog-to-digital converter (ADC) output to lower sampling rates of 10, 1, and 0.1 kHz. Initially, studies have concentrated on the sawtooth collapse. By means of a trigger system data on the sawtooth crash have regularly been recorded at a sampling frequency of 200 kHz. Using tomographic reconstruction techniques, the soft x-ray emissivity cross section during the crash has been reconstructed. First results during ion cyclotron radio frequency (ICRF) heating show no precursor activity prior to the sawtooth collapse, while during the crash, the hot central plasma is displaced off axis by about 30 cm on a time scale of 50–200 μs, after which a hollow soft x-ray profile forms.

    Notes: The knowledge of IgE-binding epitopes on allergen molecules is important for better understanding allergen–antibody interactions and, thus, for developing new strategies for immunotherapy.Our purpose was to more precisely define the number and structure of IgE-binding epitopes of a paradigmatic major grass pollen allergen.We performed an IgE-binding epitope mapping of rHol l 5, a group V pollen allergen of velvet grass (Holcus lanatus), with overlapping fragments (length between 15 and 186 amino acids), which were expressed in E. coli as MBP fusion proteins. Using sera of 65 grass pollen allergic patients, the fragments were analysed by immunoblotting for IgE reactivity. Specificity of antibody binding was confirmed by competitive blot inhibition assays.At least four different continuous IgE-binding epitopes were identified on small fragments (about 30 amino acids), and at least five different discontinuous IgE-binding epitopes on larger fragments, which were destroyed by further fragmentation. The fragments were differentially recognized by individual patients' sera. By investigating IgE-binding to one of the small fragments in more detail, we found further epitope regions on this fragment. It was noteworthy that IgE reactivity to small fragments was weak compared to large fragments or to the complete molecule. Competitive blot inhibition experiments showed that binding of IgE antibodies to the small fragments was specific but with lower avidity than to the complete rHol l 5.rHol l 5 harbours multiple discontinuous as well as continuous IgE-binding epitopes spread over the whole molecule, which were individually recognized by IgE antibodies from different patients. Low avidity of IgE antibodies to small fragments suggests that the continuous epitope regions do not represent the complete epitope and are most probably parts of discontinuous epitopes.
