    Notizen: Background: information concerning the mechanisms underlying recovery from hydronephrosis is limited. The frequency of apoptosis during healing from hydronephrosis was studied using a rat kidney model. Methods: The presence of apoptosis was studied using an in situ DNA 3 end labeling method, electron microscopy, and agarose gel electrophoresis. Results: The degree of apoptosis in both the medulla and cortex gradually increased during ureteral obstruction as shown by in situ DNA 3 end labeling. Release of the ureteral obstruction resulted in a further increase in the degree of apoptosis in the medulla and cortex. The increase in apoptosis in the medulla was transient and lasted for only 4 days following release, while that in the cortex continued for at least 3 weeks. Apoptosis in the glomerulus was not observed. Electron microscopy revealed cells with aggregated chromatin in compact granular masses that abutted the nuclear membrane. Following release of ureteral obstruction, DNA fragmentation characteristic of apoptosis was visible on agarose gel electrophoresis. Conclusion: These results suggest that apoptosis is involved in post-obstructive tubular damage in the rat kidney.

    Notizen: Background: Information concerning varicocele is limited in spite of its clinical importance. This study intends to clarify anatomical features of varicocele. Methods: Spermatic veins were obtained from varicocele patients at surgery. Areas of two distinct portions (extracellular matrix and smooth muscle) were measured by a micro computer imaging device image analysis system on histologic slides with Masson trichrome stain. In addition, we evaluated the shape and arrangement of the smooth muscle fibers by a scanning electron microscope (SEM). Results: Masson trichrome stain revealed that grade of varicocele partly correlated with thickness of the wall and with amount of the smooth muscle fibers. In the patients with grade 3 varicocele, SEM examination showed marked hypertrophy of longitudinal smooth muscle fibers, which were discriminated clearly from the inner circular fibers. Conclusions: This study indicated that morphological changes occurred in the muscle layers of varicoceles in the worst grade of varicocele. These structural changes which result from abnormal venous blood circulation may cause development of varicocele, resulting in male infertility.

    Notizen: Background : Little is known about the mechanisms underlying recovery from hydronephrosis. Using a rat kidney model, we investigated the possible contribution of growth factors during regeneration after hydronephrosis. Methods : After the unilateral ureteral obstruction for 7 days, the obstruction was released by ureterocystostomy. Epidermal growth factor (EGF), hepatocyte growth factor (HGF) and transforming growth factor-β1 (TGF-β1) were studied using immunohistochemistry and semiquantitative reverse transcriptase–polymerase chain reaction (RT-PCR). Results : The immunoreaction of EGF in the medulla of both obstructed and contralateral kidneys increased after releasing the obstruction. The release of ureteral obstruction brought further increased immunoreaction of HGF to both the cortex and the medulla of the contralateral kidney. The immunoreaction of TGF-β1 also increased in the interstitium especially around the blood vessels in the post-obstructed kidney. The expression of HGF and EGF mRNA in both kidneys and TGF-β1 mRNA in the obstructed kidney were increased after releasing the obstruction. Conclusions : These results suggest that various growth factors may be involved in the post-obstructive tubular recovery and interstitial damage in the rat kidney.

    Notizen: An 8-year-old boy was admitted to Ehime University Hospital, Ehime, Japan, for the further investigation of a 5-month episode of gross hematuria accompanied by lower abdominal pain. Magnetic resonance imaging revealed a solid tumor measuring 3 cm in diameter of the bladder wall. Cystoscopy demonstrated a red, wide-based, nodular tumor situated on the dome of the bladder. Histological examination of tissue taken at hot biopsy showed fibrolipoma. In consideration of potential malignancy, a partial cystectomy was carried out after informed consent was given. Histological examination of the resected specimen showed it to be cavernous hemangioma.

    Notizen: Abstract The pathogenesis of nonbacterial prostatitis (NBP) is not understood mainly due to the lack of appropriate experimental models. We developed a new experimental model of NBP by inducing a partial obstruction of the urethra (PUO) in the rat. Male Wistar rats aged 12 weeks were used. PUO was produced by a nylon ligature on the urethra over a rubber tube. The tube was slipped out after the ligature had been tied. Two rats were examined histologically 6 h, 1 day, 3 days and 7 days after PUO. In another group, two rats were killed at 1, 3 and 7 days after the release of the PUO that had been left in place for 3 days. On day 3, another eight rats with PUO and eight control rats had 2 ml of urine in the bladder replaced by the same volume of lucifer yellow (LY; 10 μg/ml, MW 500), microperoxidase (MP; 20 μg/ml, MW 1900), horseradish peroxidase (HRP; 10 μg/ml, MW 40 000), or saline as control, respectively. Lymphocytic infiltration and interstitial edema were noted in the prostate following PUO, being most prominent on day 3. After the release of the PUO, these inflammatory changes gradually disappeared. Only LY was noted within the prostatic stroma of the rats 2 h after bladder instillation. Intraprostatic urinary reflux may be an etiologic factor in NBP. The present study showed that lower urinary tract obstruction caused NBP in the rat. Penetration of prostatic tissue by low-molecular-weight substances in the urine may trigger NBP.

    Notizen: Abstract The role of Fas in the apoptosis of testicular germ cells was investigated in BALB/c mice and Fas-deficient lpr/lpr mice. Spontaneous apoptosis of germ cells was observed in the testes of 40-day-old BALB/c mice, and experimentally induced cryptorchidism increased this apoptosis to such an extent that there was a decrease in the weight of the testis. Flow cytometry and immunohistochemistry using a Fas-specific monoclonal antibody demonstrated expression of Fas on germ cells including spermatogonia, spermatocytes, and spermatids. Furthermore, analysis by flow cytometry suggested that Fas expression on germ cells was increased following cryptorchidism. However, spontaneous and cryptorchidism-induced apoptosis of germ cells were also observed in 40-day-old Fas-deficient lpr/lpr mice. Moreover, testis weight also decreased following cryptorchidism in the mutant mice. The present results may indicate that the expression of Fas on germ cells does not correlate with spontaneous apoptosis or apoptosis induced by cryptorchidism. However, on the contrary, this study shows that Fas are partly involved in cryptorchidism-induced apoptosis, because the decrease in testis weight of lpr/lpr mice was less than that in BALB/c mice.
