    Notes: Abstract The cortical response to stereoscopic stimuli was measured as a function of disparity using visual evoked potentials (VEP). The stereoscopic stimulus consisted of a dynamic random-dot pattern that portrayed a three-dimensional horizontal grating. Disparity of the grating was variable between 0 and 18 arc min, step size being 4.22 arc s. Evoked responses were recorded using a “random-sequence sweep technique.” The VEP amplitude increased approximately linearly with the logarithm of disparity and allowed an estimation of the stereo threshold by linear extrapolation. The evoked potential estimates of the stereo threshold of 16 subjects were compared to psychophysically obtained thresholds. Several threshold-estimation techniques are discussed. Evoked potential and psychophysical threshold estimates had the lowest discrepancy when the VEP amplitude was linearly extrapolated to the logarithmic disparity axis. The difference between the evoked potential estimate and the psychophysical threshold was less than 20% in 56% of all cases; 16 of 18 cases (89%) agreed within a factor of 2.

    Notes: Summary Crude synaptosomal preparations of rat brain were separated by concomitant flotation and sedimentation in a sucrose gradient into fractions containing myelin, nerve endings, and mitochondria, respectively. Two myelin-free nerve ending fractions were obtained which differ in the serotonin content and the activities of the enzymes 5-hydroxytryptophan decarboxylase, monoamine oxidase, choline acetylase, and acetylcholin-esterase. In glucose medium sodium ions stimulate serotonin uptake into the nerve endings while potassium ions in sucrose solution induce the release of this amine.

    Notes: Summary The present study shows that low amounts of applied iron have a potent effect on the ventrolateral striatum. This is reflected by an influence on spontaneous night activity, cognitive behaviour during the water maze navigation task, exploratory activity and in response to postsynaptic apomorphine stimulation. Such functional disturbances could be observed up to months after a single application of either 0.3 μg or 1.5 μg FeCl3. The low dose of iron stimulates while 1.5 μg inhibits the spontaneous dopaminedependent locomotor night and explorative activity. The low concentration of ionic iron injected intrastriatally also increases lipid peroxidation in striatal and hippocampal tissues. These results suggest that the functional integrity of the ventral striatum and the regulation of the iron metabolism are critical for the sensorimotor performance.

    Notes: Summary The effects of the cytokines interferon (IFN)-γ, interleukin (IL)-1, and tumor necrosis factor (TNF)-α on the serotoninergic transmission in the nucleus raphe dorsalis (NRD) were studied after peripheral and central application. The studies were performed in the freely moving rat using differential pulse voltammetry with multicarbon fibre electrodes to study the extracellular levels of the serotonin (5-HT) metabolite 5-hydroxyindoleacetic acid (5-HIAA). The extracellular 5-HIAA levels were not changed in the NRD after peripheral application of rat recombinant IFN-γ, but elevated by the cytokines IL-1β and TNF-α. After intracerebroventricular (i.c.v.) application the cytokines IFN-γ, IL-1β and TNF-α stimulated the serotoninergic transmission in the NRD. Our data suggest that the effect of peripherally elevated cytokine concentrations on the serotonin metabolism in the NRD of the rat is cytokine-dependent. In this respect the T-cell and NK-cell cytokine IFN-γ acts clearly different when compared to the mainly macrophage-derived cytokines IL-1β and TNF-α, and plays a different role in the communication between immune and central nervous system.

    Notes: Summary Sleep deprivation (SD) represents a well-established therapy for major depression. Recent findings suggest that the antidepressive effects of sleep deprivation are mediated at least in part by pro-serotoninergic mechanisms. Furthermore, SD has been demonstrated to modify different host defense activities. We therefore investigated the serotonin (5-HT) content in platelets, platelet density distribution and 5-HT-induced IL-1β release from platelets in 10 healthy men before and after total SD (TSD) as well as after recovery sleep. Blood samples were drawn on 3 consecutive days at 7.00 h, 13.00 h, and 19.00 h, respectively. In addition, the psychophysiological parameters tiredness and wakefulness were assessed. After TSD the normal daily variation of IL-1β release with high morning levels and low evening levels was found to be significantly inverted. The release of IL-1β corresponded positively to the subjectively experienced tiredness of the probands. Analysis of platelet density distribution indicated a significant daily variation of low density platelets with low levels in the morning and high levels in the evening, which was absent after TSD. Our findings favour an increased pro-serotoninergic effect after TSD, which comprises respective variations of the host defense system, but is abolished by consecutive recovery sleep.

    Notes: Summary Plasma membranes were isolated from pig platelets after glycerol facilitated lysis by sucrose density gradient centrifugation. The purity of the membrane fraction was followed by electron microscopy, gel electrophoresis and analysis of acid phosphatase (EC 3.1.3.2) and phosphodiesterases (EC 3.1.4.1). (3H)5-Hydroxytryptamine ([3H]5-HT) was bound to two saturable binding sites of the membranes. The KD value for the high affinity sites was 0.85 nM and for the low affinity sites 0.48μM. With the exception of tryptamines little or no (3H)5-HT was displaced by serotonin antagonists and uptake inhibitors suggesting another type of binding than that of 5-HT1. Apparently, enhancement of binding in the presence of Na+ was due to stimulation of an uptake process. Binding of (3H)ketanserin and (3H)LSD to pig platelet membranes showed the characteristics of 5-HT2 binding sites previously identified in rat brain. Since ketanserin inhibited 5-HT induced aggregation of pig platelets (IC50=14.2 nM), the ketanserin binding sites can be classified as 5-HT2 receptors. The functional properties of these binding sites and their density in pig platelets as compared with brain membranes may motivate studies on 5-HT2 receptors in pig platelets as models for those in nerve endings.
