    Notes: To determine the responses of plants to deficiencies of multiple metals, tobacco plants (Nicotiana tabacum L.) were subjected to treatments that were deficient in combinations of Fe and two other micronutrients, Zn and Mn. The response was measured using macro indices, including plant appearance, FW, chlorophyll concentration, and mineral concentrations, and with a molecular index, the barley (Hordeum vulgare L.) Ids2 promoter/GUS fusion gene system (Yoshihara et al. 2003, Plant Biotech 20: 33–41). Tobacco plants grown in medium with combined deficiencies grew better and had higher chlorophyll concentrations than did plants grown on medium deficient in Fe only, although the measured Fe concentrations in the plant tissues were essentially the same. The Ids2/GUS expression responded to Fe deficiency, but not to Mn or Zn deficiencies in tobacco plants when Fe was present. Tobacco plants grown in medium with combined deficiencies had clearly detectable GUS activity, but the response was significantly lower than that in tobacco plants deficient in Fe only. The Fe-deficiency symptoms were mitigated at both the visible and molecular levels. Although more precise experimental evidence is needed to explain the mitigation mechanism, the balance of minerals was shown to be an important parameter to consider when estimating iron deficiency based on tobacco plant responses.

    Notes: Objectives: To examine the effect of periodic preventive care on the progression of periodontal disease in 24 young adults with Down's syndrome (DS) (mean age±SD: 20.8±5.6 years)Material and Methods: Subjects were divided into two groups: 13 subjects who had frequently visited our clinic (mean interval between visits: 3.7±1.3 months; managed group) and 11 subjects who had not visited our clinic for more than 1 year (mean duration of no visits: 27.5±10.1 months; interrupted group). The progression of periodontal disease in the subjects was evaluated clinically, microbiologically and roentgenologically.Results: Clinical parameters (probing depth, frequency of the presence of a pathological periodontal pocket, alveolar bone loss, frequency of the incidence of pathological bone loss, and modified total PMA (M-PMA)) and benzoyl-dl-arginine-naphthylamide (BANA) scores in the interrupted group were significantly higher than those in the managed group. Subject age showed significant positive correlations with probing depth and alveolar bone loss. BANA scores showed significant positive correlations with probing depth, alveolar bone loss and M-PMA in the interrupted group.Conclusions: The results suggest that periodic preventive care is effective for suppressing the progression of periodontal disease in young adults with DS.

    Notes: Abstract Glutelin is the most abundant storage protein in rice, which is expressed specifically in the endosperm of maturing seed. Glutelin is encoded by about 10 genes per haploid genome, which are clearly divided into two subfamilies (GluA and GluB). Most of them are coordinately expressed during seed maturation in spite of the remarkable divergence in the 5′-flanking regions between members of two subfamilies. In order to identify the common regulatory mechanisms responsible for the endosperm-specific expression, various cis-regulatory elements in the 5′-flanking region of the glutelin GluB-1 gene were characterized by studying the expression of chimeric genes that consisted of the sequentially deleted or mutagenized promoter and a β-glucuronidase (GUS) reporter gene in transgenic tobacco seeds. The essential cis-regulatory elements governing the spatially and temporally specific expression of the glutelin gene expression were located within the first 245 bp of the promoter region of the GluB-1 gene from the site of initiation of transcription. The AACA motif between positions −73 and −61 common to all the six genes for glutelin sequenced to date and is repeated between positions −212 and −200 is implicated in the seed-specific expression. The GCN4 motif between positions −165 and −158 and between positions −96 and −92 that is conserved at homologous sites in all the members of glutelin gene family is also involved in the seed-specific regulation. However, both are required for the high level of seed-specific expression, because deletion of the region containing one set of both elements or substitution mutation of the AACA or GCN4 motif substantially reduced the activity. As a whole, our results suggest the combinatorial interaction of the elements in regulation of the glutelin gene expression.

    Notes: Abstract High iron-content transgenic tobacco plants have been produced by transfer via Agrobacterium tumefaciens of soyabean ferritin cDNA under the control of a CaMV 35S promoter. Immunoblot analysis of protein from transgenic tobacco plants suggested mature ferritin subunits are produced by cleavage of transit peptides. The expressed ferritin was observed in the tissues of leaves and stems. The maximal iron content of transformant leaves was approximately 30% higher than leaves from non-transformants. The increased iron content of each transformant was correlated with increases in ferritin content. These results demonstrate the potential of breeding high iron content crops by introduction of the ferritin gene
