    Notes: Summary Nine patients who underwent heart transplantation (one female; average age 48 ± 11, range 19–58 years) were followed in respect to contents of right-sided heart septum, blood and plasma ubiquinone (UQ), plasma α-tocopherol (αT), and plasma free cholesterol (FC). In contrast to healthy persons, substantial inter- and intraindividual variations were observed; individually low values were seen with rejection. Heart muscle UQ in well-treated patients averaged 0.33 ± 0.08, range 0.06–0.58 μg mg−1 (0.38 ± 0.09 μmol g−1 dry weight) and was not different from healthy individuals. Plasma UQ, αT; and FC averaged 0.63 ± 0.33 μg ml−1 (P 〈 0.05 versus sedentary controls), 8.1 ± 4.0 μg ml−1 (P 〈 0.01), and 0.52 ± 0.23 mg ml−1 (P 〈 0.05). Corresponding molar values were 0.73 ± 0.37 (UQ), 2.0 ± 1.1 μmol l−1 (αT), and 1.42 ± 0.54 mmol 1−1 (FC). Blood and plasma UQ values were identical. A saturationlike relationship was found between heart and blood UQ: blood contents below 0.7 μg ml−1 (0.8 μmol l−1) corresponded to markedly lowered heart contents. In four patients in whom blood samples were taken close to a fatal complication it averaged 0.42 μg ml−1 (0.49 μmol l−t, P 〈 0.01). When low heart muscle and blood ubiquinone were present, other variables such as left ventricle cardiac output or cycle ergometer performance was markedly impaired. Plasma UQ and off covaried with a marker of the lipoidal deposit volume, plasma FC. The ratios UQ and αT over FC (N-UQ and N-αT) are alternative means for clinical evaluation. Mean N-αT was relatively more depleted than N-UQ. On an individual basis this was more pronounced for those with low N-UQ than for those with high values.

    Notes: Abstract  The hybridization of total RNA isolated from Nicotiana tabacum pollen tubes with probes for tubulin and actin mRNA revealed a high level of tubulin and actin transcripts in pollen tubes 1.5–18 h old. A weak signal for tubulin and a strong signal for actin mRNAs were still detected in tubes 24 h old. The results suggest that the presynthesized tubulin and actin mRNAs in N. tabacum pollen tubes are very stable or that transcription of genes encoding cytoskeletal proteins may take place during growth of the pollen tubes. The increase of the signal for α- and β-tubulin and actin in the immunoblots of one-dimensional gels between 1.5 and 6 h indicated that the transcripts observed in the northern blots were translated. The increase in the cytoskeletal protein levels correlated well with previous cytological observations which show that in N. tabacum the movement of the vegetative nucleus (VN) and generative cell (GC) from the grain into the tube is most efficient between 3 and 6 h after germination. In the two-dimensional immunoblots it was observed that the signal for three out of four α-tubulin isotypes present in pollen tubes increased between 1.5 and 3 h. The time of the increase of α-tubulins was in good agreement with the start of movement of the VN and GC from the grain into the tube, and the synthesis of the α-tubulins could be a prerequisite for the beginning of the movement. These observations lend support to the idea that the microtubule cytoskeleton, in addition to microfilaments, is involved in the transport of the VN and GC in pollen tubes.

    Notes: Abstract Magnetic susceptibility χ of polycrystalline rhombohedral samarium has been measured as a function of temperatureT between 4 and 300 K. The χ vs.T curve exhibits two peaks: a large one at 15 K and a small one at 106 K. The magnetic susceptibility between the Néel temperatureT N=106 K and 300 K obeys the predictions of the Stewart theory. We also demonstrate hysteretic effects in the magnetic susceptibility aroundT N and indicate that no significant results on critical indices could be obtained from these measurements.

    Notes: Summary In 3-and 12-hour-old pollen tubes ofNicotiana tabacum microtubules (MTs) most resistant to cold-depolymerization were located in the generative cell (GC). The most cold-sensitive MTs occurred in the apical region of the pollen tube. In the basal part of the tube, the cold-resistance of MTs increased, especially in 12-hour-old tubes. The cold depolymerization pattern of MTs with accumulation of tubulin at the cell and nuclear membranes suggested that binding of MTs to membranes might increase their stability. The IIF microscopy and staining with fluorochrome-conjugated phallotoxins indicated that microfilaments (MFs) unlike MTs remained polymerized in cold-treated pollen tubes. Recording of the positions of the vegetative nucleus (VN) and GC in the control and cold-treated pollen tubes in different ages showed clearly that low temperature stopped sharply the movement of the VN and GC into the young tubes while no drastic effects were recorded in the old tubes. The cessation and recovery of the movement correlated well with de-and reassembly of MTs, which indicated that MTs in addition to MFs may be necessary for the movement of VN and GC. The immunoblot results showed that the levels of tubulin and actin monomers in the cold-treated tubes were comparable to those present in the control tubes, which might be a prerequisite for the rapid recovery of the structure and function of the cytoskeleton recorded after the return of the cold-treated pollen tubes to normal temperature.

    Notes: Summary The effects of nocodazole and brefeldin A (BFA) on the growth of dikaryotic hyphae inSchizophyllum commune corresponded with the development of abnormal structures in the apical region of treated hyphae. Microtubules (MTs) were totally depolymerized after 1 h nocodazole treatment, which correlated with strong branch formation in the apical cells. One reason for branching could be the shift in the position of apical vesicles from the center to the side of the tip, observed in some nocodazole-treated hyphae. After 2 h growth in the presence of nocodazole the apical cells had malformed or swollen tips, or tips of normal shape but containing only a few apical vesicles. After 0.5 h treatment with BFA, almost all the leading hyphae had swollen apical parts in which the endoplasmic reticulum (ER) formed an interconnected network and perturbed Golgi particles were found. The orientation of MTs in the BFA-treated hyphae often followed that of the interconnected ER network, which suggested an association between MTs and ER. The results of the experiments with nocodazole suggest that, in filamentous homobasidiomycetes the subtle organization of cytoplasm necessary for the polar growth at the apex is maintained only in the presence of an intact MT cytoskeleton. The BFA experiments indicated that the secretion pathway inS. commune is sensitive to BFA. In addition rapid change in apical morphology in the BFA-treated hyphae emphasizes the importance of correct orientation of components of the secretory pathway for normal apical growth to continue.
