    Notizen: Gene frequencies in samples of aerial populations of barley powdery mildew (Erysiphe graminis f.sp. hordei), which were collected in adjacent barley areas and in successive periods of time, were compared using mobile and stationary sampling techniques. Stationary samples were collected from trap plants in three periods within 1 week at a distance of more than 1000 m from the nearest barley field. At four dates within the same 8-day period, other samples were collected by a mobile spore trap along four sampling routes of a total distance of 130 km around the stationary stand of exposure. The samples were characterized by virulence genotypes defined according to infection types on 12 near-isogenic barley lines, and frequencies of single virulence genes were subsequently calculated. The three samples collected at the stationary site differed significantly with respect to allele frequencies at three loci. The main wind direction was different in the three sampling periods, implying different powdery mildew sources. For the mobile exposure, the differences between routes were not significant for any locus. However, the routes differed most for the loci under direct selection by host resistances genes, indicating a different distribution of source varieties along routes. There was no difference between allele frequencies at different dates, indicating that the proportions of spores from different source varieties were similar at these dates. In conclusion, samples collected by the stationary technique will mainly reflect the source varieties present in the local area, whereas samples collected by the mobile spore trap will mainly reflect sources close to the sampling route. Therefore, sampling sites as well as sampling routes should be defined such that source varieties are representative for the overall varietal distribution in the survey region considered.

    Notizen: A Japanese field isolate (Race I) of Erysiphe graminis f.sp, hordei was tested on 17 barley lines carrying the mlo powdery mildew resistance gene. Race I produced many successful infections with infection type larger than or equal to 2 on six lines (M66, MC20, SRI, SR7, A tem and Totem), On the remaining 11 lines it reacted with infection type 0. Colony numbers on the Mlo-lines were between 26% and 12 9% of the numbers on a susceptible cultivar Manchuria, These numbers were larger than, or similar to those produced by isolate HL3/5, which has the highest recorded infection efficiency on Mlo-resistant cultivars. The interaction between isolates and lines was highly significant. The isolate GE3, from which HL3/5 was derived by selection, gave rise to occasional colonies corresponding to less than 03° o of the number produced on cv. Manchuria.

    Notizen: The effect of the use of different doses of the fungicide fenpropimorph on populations of barley powdery mildew Blumeria (Erysiphe) graminis f. sp. hordei was investigated in a laboratory selection experiment. A sample from the Danish aerial population of powdery mildew was split into populations, and these were kept separately for 31 generations on susceptible barley seedlings treated with fungicide at two concentrations, as well as on a control. Samples from these populations were tested for their resistance to fenpropimorph and their virulence spectra. There was a large amount of environmental variation in the ED50 values used to measure fungicide resistance. In both treated populations, the average level of fungicide resistance increased, this increase being faster and greater in the population treated with the high dose. The diversity of pathotypes of the treated populations decreased, with the decline being more rapid in the population treated with the high dose, where one pathotype dominated the population after 31 generations. This pathotype was apparently not the fittest in the population treated with the low dose. This implies that knowledge of ED50 is not sufficient to predict pathotype evolution under different fungicide treatments. The dominant pathotype in the high-dose treatment may not have been clonal, as there was evidence of two levels of fungicide resistance. The large environmental variation observed in estimated ED50 values for resistance towards fenpropimorph may help to explain why this resistance has evolved at a slower rate than resistance towards other fungicides.

    Notizen: Aerial populations of Blumeria graminis f.sp. hordei were studied in two French and two Danish regions from 1991 to 1999, at a time of year when only winter barley was present. A high frequency of genotypes not able to grow on the spring-sown crop of the previous growing season (denoted ‘spring-avirulent’) was observed in most years and regions. This frequency increased with increasing proportion of winter barley; it was highest in France and decreased in general over the 8-year period. Most of the spring-avirulent genotypes possessed the Va22 virulence gene, matching a resistance that has never been present in barley cultivars grown in Europe. A hypothetical cropping system, including winter- and spring-sown crops with three resistance genes altogether, was constructed to mimic the utilization of host cultivars in the four regions. Results from a mathematical model simulating changes in the composition of the pathogen population in this system, demonstrated that selection solely due to host resistance genes, i.e. without assuming any cost of virulence, might lead to such results as those observed. The changes in frequency of spring-avirulent genotypes and the frequency of unnecessary virulence genes may be predicted from the proportion of the barley area sown with winter barley, the use of resistance genes in the cultivars, the initial composition of the pathogen population, and hitch-hiking due to gametic disequilibria.

    Notizen: Abstract Genetic variability of cultivated and wild barley, Hordeum vulgare ssp. vulgare and spontaneum, respectively, was assessed by RFLP analysis. The material consisted of 13 European varietes, single-plant offspring lines of eight land races from Ethiopia and Nepal, and five accessions of ssp. spontaneum from Israel, Iran and Turkey. Seventeen out of twenty-one studied cDNA and gDNA probes distributed across all seven barley chromosomes revealed polymorphism when DNA was digested with one of four restriction enzymes. A tree based on genetic distances using frequencies of RFLP banding patterns was estimated and the barley lines clustered into five groups reflecting geographical origin. The geographical groups of land-race lines showed less intragroup variation than the geographical groups of spontaneum lines. The group of European varieties, representing large variation in agronomic traits, showed an intermediate level. The proportion of gene diversity residing among geographical groups (FST) varied from 0.19 to 0.94 (average 0.54) per RFLP pattern, indicating large diversification between geographical groups.

    Notizen: Summary A mathematical model is presented and analysed to find the conditions under which changes in gene frequencies can be used in asexually reproducing populations for estimating fitness of single genes, for example, for estimating the fitnesses of unnecessary virulence genes relative to their corresponding avirulence genes. It is concluded that the underlying distribution of relative fitness of clones (genotypes) has to be unimodal and that many populations consisting of a mixture of distinguishable clones then provide the best experimental data for estimating relative fitness of single genes. An improved statistical test procedure, i.e. generalized logistic regression, is suggested for analysing changes in gene frequencies in population experiments with a mixture of distinguishable clones. A population study of Erysiphe graminis f.sp. hordei (Klug-Andersen 1980) provides data to illustrate the procedure in the case where the population consists of a large number of genotypes. A bimodal distribution of “genotypes” possessing the virulence gene is indicated here.
