Summary The isolated portal vein of the rat shows according to anatomic and physiological experiments a constant mechanical spontaneous activity. The influence of val5-angiotensin II has been investigated. This peptide enhances the active tension at a concentration of 10−9 g/ml. The effect remains unchanged after pretreatment of the rats 48, 24 and 12 hours previously by reserpine (3×1,5 mg/kg) and was not influenced by propranolole, phenoxybenzamine, atropine, but completely abolished by iproveratrile. The influence of various cations and temperature was studied. The results show, that the effect of val5-angiotensin II is not caused by an adrenergic mechanism but by direct action on smooth muscle of the portal vein.

Zusammenfassung Die isolierte Pfortader der Ratte gehört zu den Gefäßpräparaten, die in vitro eine mechanische Spontanrhythmik zeigen und auch elektronenmikroskopisch die Charakteristika solcher spontan aktiver glatter Gefäßmuskeln aufweisen. Es wurde der Einfluß von Angiotensin II (Val5-Angiotensin-II-Asp-β-amid) auf die Kontraktilität und Autorhythmik geprüft. Angiostensin II führt in einer Konzentration von 5×10−10 g/ml bereits zu einer signifikanten Zunahme der Kontraktionsamplitude, der Effekt ist bei einer Konzentration von 10−8 g/ml maximal. Diese Wirkung von Angiotensin ist durch Senkung der Inkubationstemperatur auf 5°C ebenso wie die Wirkung anderer Agonisten zu hemmen. Ca2+-Entzug inhibiert ebenso wie Zugabe von Iproveratril die kontraktilitätssteigernde Wirkung des Oktapeptides; Atropin, Phenoxybenzamin und Propranolol sind ohne Einfluß auf die Wirkung von Angiotensin II. Wie die Versuche an reserpinisierten Tieren zeigen, greift die Substanz direkt an der Mukelzelle an und wirkt nicht als indirektes Sympathomimetikum. Sie ist auch nach wiederholter Gabe von Tyramin (10−6 g/ml) noch wirksam und löst ferner keine Tachyphylaxie an dem von uns verwendeten Präparat aus.
