    Notes: We have developed an ultrahigh vacuum plasma-enhanced chemical-vapor deposition system, and deposited high-purity device-quality hydrogenated amorphous silicon films. High sensitivity secondary ion mass spectrometry measurements show that impurity contents in the bulk of the present films are reduced to 2×1015 cm−3 for O, 7–10×1015 cm−3 for C, and 5×1014 cm−3 for N; these impurities are normally present at fairly high levels. Nevertheless, extensive light soaking of the films resulted in a defect density as high as 5×1017 cm−3, which is well above the impurity content. This result excludes those models of photoinduced degradation that postulate one-to-one correlation between light-induced defects and O, C, or N impurity atoms. © 1996 American Institute of Physics.

In an attempt to clarify the developmental mechanisms of lacuna and diffuse degeneration of the white matter in the brain in chronic hypertension, we investigated histologically the cerebral changes and histochemically, as well as biochemically, the lysosomal enzymes in the brain of stroke-prone spontaneously hypertensive rats (SHRSP).2. The most prominent advanced lesions observed in SHRSP were cyst formation in the cortex and subcortical white matter, and diffuse degeneration of the white matter. On the other hand, the early cerebral changes were all related to blood-brain barrier dysfunction. The localization of cystic lesions and degeneration of the white matter corresponded very well with the extent of brain oedema demonstrated by immunostaining for leaked fibrinogen. All lysosomal enzyme activities in the adult SHRSP, both in the cortex and white matter, were higher than those in the controls. Histochemical investigation showed that SHRSP had an increased number of cells, reactive astrocytes and microglial cells, with positive reaction to lysosomal enzymes in the oedematous portion.3. These findings suggest that chronic oedema due to blood-brain barrier dysfunction causes cystic changes as well as diffuse degeneration of the white matter, and that activated lysosomal enzymes in the reactive astrocytes and microglia play an important role in the development of such hypertensive lesions.

    Notes: Summary Cell proliferation in the brain of spontaneously hypertensive rats (SHR) and control Wistar rats of various ages was autoradiographically investigated using [3H]thymidine. The brain of SHR showed an increase in labeled cells. The increase of labeled cells was seen in the early stage of the development of hypertension when there were still no definite morphological changes in the cerebral vessel walls or brain parenchyma. The labeled cells tended to increase in number with the age of the animals. The distribution of these cells corresponded with areas of increased vascular permeability and cerebrovascular lesions, that is, water-shed regions. The labeled cells consisted of endothelial and adventitial cells of the intracerebral arterioles and pial arteries as well as glial cells. Arachnoid cells, subarachnoid cells, medial smooth muscle cells of the pial arteries were also labeled, though less intensely. The significance of labeled endothelial cells in the development of cerebrovascular changes and sequential parenchymal changes is discussed.

    Notes: We used the mouse monocyte/macrophage-like tumor cell line P388D1 to test whether or not interleukin-1 (IL-1) stimulates differentiation of monocyte/macrophage progenitors. Incubation of these cells with recombinant human interleukin-1 (rhIL-1) α and β resulted in their increased adherence, stimulation of nonspecific esterase activity, and increased Fc rosette formation. rhIL-1s inhibited cell growth and stimulated Fc rosette formation in a dose-dependent fashion. The cell growth inhibition due to rhIL-1s depended on the concentration of serum in culture medium. Synergism between rhIL-1 and calcium ionophore A23187 was found for the cell growth inhibition and Fc rosette formation. The presence of ethylene glycol bis- (β-aminoethyl ether) N,N,N,N-tetraacetic acid(EGTA) in the medium abolished the stimulatory effect of rhIL-1 on Fc rosette formation of the cell line. These results demonstrate that rhIL-1s are a potent inducer of the differentiation of the macrophage-like tumor cell line P388D1.
