    Notizen: This study reports for the first time the presence of marginal cells, probably with stretch receptor function, in the spinal cord of four elasmobranch species, two electric rays, Torpedo marmorata and T. torpedo, the skate Raja undulata and the dogfish Scyliorhinus canicula. In all four species, the marginal cells were located close to the lateroventral surface of the cord and possessed thick dendrites which formed part of characteristic glomerular structures. In vitro retrograde labelling of the spinal cord of the dogfish with horseradish peroxidase (HRP) showed that some of these cells have contralateral projections. Ultrastructural study of normal and retrograde HRP-labelled material showed that the glomerular dendrites of marginal cells give rise to numerous fingerlike structures and are associated with a rich plexus of nerve terminals. Characteristically, these dendrites contain numerous mitochondria. Immunocytochemical studies revealed a rich plexus of somatostatin- and GABA-immunoreactive fibres in the glomeruli. These results strongly suggest that the marginal cells of the elasmobranch spinal cord are stretch receptors homologous to lamprey edge cells and to the marginal nucleus cells of the spinal cord of urodeles and snakes. We discuss the possible role of these cells in the modulation of swimming movements.

    Notizen: Abstract The development of somatostatin-immunoreactive neurons and fibres was studied, using immunocytochemistry, in the brain of the brown trout. Somatostatinergic perikarya were found in many regions including several telencephalic areas, the preoptic nucleus, anterior tuberal and lateral tuberal nuclei, the lateral recess nucleus, dorsal tuberal nucleus, the pre- and pseudoglomerular nuclei, central thalamic nucleus, optic tectum, interpeduncular nucleus, several isthmal and reticular nuclei and the solitary fascicle nucleus. The ventrolateral area of the telencephalon and the nucleus lateralis tuberis are the first immunoreactive nuclei to appear in ontogeny, and cells of some telencephalic areas and of the lateral optic recess nucleus, the latest. Somatostatin-immunoreactive fibre tracts innervate the hypophysis and different regions of the brain. The most richly innervated areas in adults are the dorsolateral telencephalic area and the organon vasculosum laminae terminalis. Two patterns of production of somatostatinergic cells were observed: that of populations in which cell numbers increase over the lifetime of the fish, and that of populations whose cell number is established early in development or even diminishes in adulthood. These results provide interesting contrasts to those previously reported in birds and mammals.

    Notizen: Abstract  Calcium-binding proteins of the EF-hand family are widely distributed in the vertebrate central nervous system. In the present study of the trout brain, immunocytochemistry with a monoclonal antibody against chick gut calbindin-28k and a polyclonal antibody against bovine S100 protein specifically stained ependymocytes and radial glia cells with identical patterns. Western blot analysis of trout brain extracts with the antibodies to S100 and calbindin stained the same low-molecular-weight (10 kDa) protein band. In rat brain extracts, however, the monoclonal antibody to calbindin recognized a major protein band with molecular weight corresponding to that of calbindin-28k. This indicates that the trout protein is a new calcium-binding-like (calbindin-like) molecule that is immunologically related to both S100 and calbindin. Immunocytochemical studies of the trout brain using the antibodies to CaB and S100 showed that ependymocytes were stained in most ventricular regions, except in a few specialized ependymal areas of the ventral telencephalon, epithalamus, hypothalamus (including the paraventricular organ and saccus vasculosus) and brain stem. Immunocytochemistry also indicated the presence of calbindin-like protein in radial glia cells of several regions of the brain (thalamus, pretectal region, optic tectum, and rhombencephalon). Differences in immunoreactivity between neighbouring ependymal areas suggest that this protein may be a useful marker of different territories. All immunoreactive glial cells were nicotin-adenin-dinucleotide-phosphate diaphorase-positive, although this enzymohistochemical reaction is not specific for these glial cells since it reveals oligodendrocytes and some neurons. Immunoreactivity appears at different developmental stages in the different brain regions, with a broadly caudorostral gradient, suggesting that the expression of this protein is developmentally regulated. Comparison of the distribution of the calbindin-like protein with that of glial acidic fibrillary protein indicates that calbindin-like immunocytochemistry is a specific technique for revealing radial glia and ependymocytes in the trout.

    Notizen: Abstract Development of the retinopetal system of the larval sea lamprey, Petromyzon marinus, was investigated following labelling of this system by injection of horseradish peroxidase into the orbit. This study extends our previous report on larval stages and provides a detailed description of the development of this system. We present quantitative and qualitative evidence suggesting that the retinopetal nuclei of Schober's M2-M5 nucleus, the mesencephalic reticular area and the tectum arise sequentially in that order, that the three retinopetal nuclei originate from a common anlage in the ventricular zone of the mesencephalic tegmentum and that the retinopetal cell population increases throughout the larval period. No neuronal death was observed. We also describe and discuss the significance of a transitory phase of retinopetal cell differentiation characterized by the presence of ventricular dendrites. Finally, we compare the development of retinopetal and retinofugal systems.

    Notizen: Meningeal capillaries are unfenestrated. They are made up of endothelial cells that have a pinocytotic caveolae density of 41 ± 11/μm2 and 89 ± 21/μm2 on the abluminal and luminal sides respectively. The total density of intramembrane particles is not significantly different between the luminal and the abluminal membranes; however, the coefficients of partition are significantly different (P〈 0.001). One or two strands of tight junctions occur between adjacent cells but no gap junctions nor desmosomes exist. The density of nuclear pores is less than 3.2/μm2. An abundance of intermediate filaments and free vesicles, some of which are seriated, characterize the cytoplasm. The functional significance of these findings is discussed.

    Notizen: The gross development of the trout inner ear between embryonic and juvenile stages was studied by light microscopy. The otocyst has already formed in 3-4 mm embryos. The semicircular canals begin to separate from the utriculo-saccular cavity in 6 mm embryos, the anterior canal first, then the posterior and the horizontal canal later. The formation of the saccular cavity begins in 7 mm embryos, whereas that of the lagena occurs in 18 mm fry. The first macular primordia appear before the separation of cavities. The anterior and horizontal crests arise from the primordium of the utricular macula, and the posterior crest, macula lagena, and macula neglecta arise from that of the saccular macula. The macula lagena and macula neglecta appear later. The sensory areas of the labyrinth and the number of receptor cells grow continuously between the embryonic and juvenile stages. © 1993 Wiley-Liss, Inc.

    Notizen: The study of the sensory organs of the trout labyrinth by means of electron microscopy show that hair cells differentiate gradually in these organs; all of them produce new cells over a long period. The course of cytodifferentiation follows a similar pattern in all organs. Afferent nerve fibers and terminals are found at approximately the same time that sensory cells are being differentiated; the efferent synapses appear latter in development.     Notizen: Summary The elasmobranch nucleus sacci vasculosi was studied by means of electron microscopy (in the dogfish) and immunocytochemistry (in the dogfish and the skate) by using antibodies against tyrosine hydroxylase, alpha-melanocyte-stimulating hormone, somatostatin, serotonin, and substance P. Ultrastructural study of the dogfish nucleus sacci vasculosi shows the presence of medium-sized cells that possess numerous mitochondria but that have no dense-core vesicles in the cytoplasm or in cell processes. Fibres of the conspicuous tractus sacci vasculosi have a beaded appearance and form conventional synapses with dendrites and cell perikarya of the nucleus sacci vasculosi. The perikarya of this hypothalamic nucleus were not immunoreactive to any of the antibodies tested, and fibres immunopositive to tyrosine hydroxylase, alpha-melanocyte-stimulating hormone, somatostatin, serotonin, and substance P were scarce within this nucleus, in both the dogfish and the skate. Dorsal to the nucleus sacci vasculosi, there are numerous positive neuronal processes in addition to many small neurons that show immunoreactivity to alpha-melanocyte-stimulating hormone, somatostatin and tyrosine hydroxylase. Two types of neuron occur in this dorsal region, displaying dense-core vesicles of either 100–160 nm or 60–100 nm diameter in their cytoplasm; they were identified as peptide-containing and monoamine-containing neurons, respectively. The neuropil of this region has a significantly different ultrastructure from that of the nucleus sacci vasculosi, with many processes containing dense-core vesicles. This group of neurons, located dorsal to the nucleus sacci vasculosi and showing (a) immunoreactivity to neuropeptides or to monoamine-synthesizing enzyme, and (b) cytoplasm with dense-core vesicles, was considered not to be a part of the nucleus sacci vasculosi but rather part of the nucleus tuberculi posterioris. These results support the non-peptidergic and non-aminergic character of the nucleus sacci vasculosi.

    Notizen: Summary The sequence of morphological changes in the retinal pigment epithelium during the metamorphic period of the sea lamprey Petromyzon marinus L. has been investigated using electron microscopy. At early metamorphic stages (stages I and II), photoreceptors are present in a small zone of the retina. During these stages, the lateral surface of the epithelial cells shows zonulae occludentes and adhaerentes. The degree of cell differentiation varies throughout the retinal pigment epithelium. Cells covering the differentiated photoreceptors in the central retina have phagosomes, whereas pigment granules appear only in the retinal pigment epithelium dorsal to the optic nerve head. Most epithelial cells have myeloid bodies; their morphology is more complex around the optic nerve head. At stage III, when photoreceptors develop over the whole retina, the distribution of cytoplasmic organelles is almost homogeneous in the retinal pigment epithelium. Subsequently, the basal plasma membrane of the epithelial cells becomes progressively folded and their apical processes enlarged. In addition, extensive gap junctions develop between retinal pigment cells. In late metamorphic stages, noticeable growth of myeloid bodies occurs and consequently the retinal pigment epithelium resembles that of the adult. This study also describes, for the first time, the presence of wandering phagocytes in the retinal pigment epithelium of lampreys; their role in melanosome degradation is discussed.

    Notizen: Summary The lamellate cells of amphioxus have round nuclei, and cytoplasm with many mitochondria and a large amount of glycogen. Each of these cells projects a highly modified, branched cilium into the central canal, where it characteristically forms lamellar structures. Primary branches and secondary lamellae often contain accessory microtubules that are not derived from the axonema. The functional and evolutionary significance of this cell type is discussed in relation to the ciliary photoreceptors found in other chordates.
