    Notes: Abstract The intertidal sea anemone Oulactis muscosa (Drayton) is dioecious and most individuals are sexually mature throughout the year. Biochemical genetic evidence was used to determine the genetic structure of populations and to infer the relative contributions of sexual and asexual reproduction to recruitment. Data were collected for six enzyme-encoding loci from local populations spread along 735 km of the south east coast of Australia. The genetic structure of each of the nine local populations studied was consistent with recruitment by sexually produced individuals. In almost all cases, the observed single-locus genotypic frequencies closely matched those expected for hardy-Weinberg equilibria, however, consistent deficits of heterozygotes were detected for all loci. No apparent subdivision of the population was detected within the sampling area. Low levels of genetic differentiation were found between local populations and standardised variance (F ST ) values were similar to those for other species with widespread planktonic dispersal of larvae.

    Notes: Abstract We surveyed patterns of allelic variation within twelve samples of the pipi Donax deltoides Lamarck from beaches separated by up to 1200 km but connected to varying degrees by the East Australian Current. We used these data to test the prediction that the irregular patterns of water movement would cause genetic differentiation in pipis, so that there would be more genetic variation within and among the more southern regions than the northern regions. We found that six loci were at least moderately variable within all samples, and there were no clear geographic patterns in allelic frequencies. In general, genotype frequencies within samples were consistent with predictions for an outcrossed, sexually reproducing species, and we detected no evidence of population subdivision. Within samples, with the exception of the peptidase loci, single-locus genotype frequencies were in close agreement with expectations for Hardy–Weinberg equilibrium. We observed no significant linkage disequilibrium for any pairwise comparison of loci in any sample. Our hierarchical analysis of genetic variation revealed little variation among all samples (F st = 0.009). Loci showed consistently low levels of subdivision (F st from 0.003 to 0.018). We found almost no variation among the four geographic regions sampled (F rt = 0.001). All variation was therefore attributable to variation among samples within regions (F sr = 0.010). These data imply that larvae are moving between regions and that levels of present or recent gene flow are high, and support the conclusions of other studies which have inferred widespread gene flow for animals dispersing via planktonic, outcrossed larvae in parts of this region. This implies that the East Australian Current is sufficient to produce strong larval connections despite its intermittent nature. If existing levels of population subdivision reflect current levels of gene flow, then these data imply that D. deltoides represents a single fishery on the east coast of Australia.

    Notes: Abstract Acrorhagial fighting occurs between some pairs of adult Actinia tenebrosa, and such fighting almost always results in the locomotory withdrawal of the loser. Adults are also able to attack and kill juvenile conspecifics in laboratory conditions. In laboratory trials, 58 of 89 pairs of adults responded aggressively when placed in contact. Conflicts developed disproportionately often when adults were genotypically different, as judged by electrophoretic examination of 5 polymorphic enzymes, or when adults were markedly different in weight. Neither the sexual maturity nor presence of brooded juveniles affected the probability that conflicts would develop between adults. Weight did not affect the outcome of the conflicts. Presentation of apparently asexually produced juveniles to adults revealed that both the recognition and effector phases of the acrorhagial response are triggered by genotypic dissimilarity and not by size dissimilarity per se. Acrorhagial responses were initiated only on contact with juveniles from other adults. Localised release of asexually-produced brooded juveniles contributes substantially to recruitment within established populations of A. tenebrosa. Aggressive interactions between genotypes thus seem likely to play an important role in intra-specific competition for recruitment space.

    Notes: Abstract In situ and in vitro observations indicate that brooding colonial ascidians commonly display limited larval dispersal, whilst the larvae of most solitary species are assumed to be widely dispersed. We used allozyme data to determine the population genetic consequences of reproduction and dispersal in a broadcast-spawning solitary ascidian and two brooding colonial species along the central and southern coast of New South Wales, Australia. We surveyed genetic variation at 2 to 9 variable loci for samples collected from 6 to 8 local populations of each of the stalked solitary species Pyura gibbosa gibbosa Heller, 1878; the social Stolonica australis Michaelsen, 1927 and the compound Botrylloides magnicoecum Hartmeyer, 1912. Samples from each local population displayed levels and patterns of genotypic diversity that were consistent with expectations for sexually-derived recruitment of both solitary zooids and separate colonies. However, we found clear differences in the structure of the populations of solitary and colonial species. Genotype frequencies within all nine samples of P. gibbosa gibbosa conformed to expectations for random mating (i.e. Hardy–Weinberg equilibria). Moreover, allele frequencies showed little variation among samples [mean standardised genetic variance (F S T ) =0.002], which implies that local populations are strongly connected by larval dispersal. We estimate (via Wright's “island model”) that gene flow (N e m) within this set of local populations is 125 effective migrants per generation, which is very similar to estimates obtained for other broadcast-spawning taxa in this region. In contrast, genotype frequencies within samples of both colonial species were characterised by large and statistically significant deficits of heterozygotes, consistent with expectations for highly limited dispersal of larvae or sperm. Moreover, local populations were highly differentiated (F S T =0.201 and 0.202 for S. australis and B. magnicoecum, respectively) and N e m was estimated to be ∼1.0 in each case. These values of F S T and subsequent estimates of N e m lie within the range of values reported for other New South Wales taxa with direct larval development, and imply that local populations are effectively closed to immigration.

    Notes: Abstract An electrophoretic survey of allozyme variation revealed substantial genetic differentiation within the eastern Australian population ofActinia tenebrosa. This differentiation appears to reflect the effects of both asexual reproduction and limited gene flow among local populations separated by up to 1050 km. Variation was assessed within groups of 27 to 55 adults sampled between September 1985 and December 1988 collected from small areas of shore within each of 24 local populations. All individuals were collected from stable rock platforms, with the exception of Boulder Bay, where some sea anemones were removed from small mobile boulders. High levels of variability were detected for each of seven enzyme-encoding loci. The patterns of genotypic variation detected imply that local populations are maintained by predominantly asexually generated recruitment. Levels of multi-locus genotypic diversity within samples were consistently less than 50% of the level expected for sexual reproduction with free recombination. This was reflected by the detection of relatively low numbers of multi-locus genotypes and significant departures from expectations for single-locus Hardy-Weinberg equilibria within 17 of the 24 local populations. Standardised genetic variances (F ST ), calculated from the genotypes of all individual adults were typically much greater than those expected for marine organisms with widely dispersed larvae. The former values were reduced, but were still extremely large when “clonal” genotype frequencies were substituted into the calculation. These data imply that although widely dispersed larvae may be an important source of initial colonists, levels of gene flow among established local populations are low. Furthermore, cluster analysis revealed a clear subdivision of the population into northern and southern groups. However, this subdivision was largely explained by strong clinal variation at a GPI-encoding locus. For this locus, allele frequencies ranged from fixation of the A allele in samples from the 12 most northern sites to near fixation of the alternative B allele in southern samples. Subdivision of the eastern Australian population is consistent with the predicted off-shore movement of the Eastern Australian Current close to the border between Victoria and New South Wales. However, the split into northern and southern regions, as evidenced by the variation forGpi, could reflect patterns of gene flow and/or other factors such as natural selection or the recent patterns of colonisation.

    Notes: Abstract The encrusting spongeHalisarca laxus forms a seemingly obligate association with the stalked solitary ascidianPyura spinifera. In 1991 we examined spatial variation and short-term temporal variation in this association at three neighbouring sites in southeastern Australia. This sponge dominated the surface of almost all the 500 individual ascidians examined, with mean cover usually exceeding 90%. This pattern was consistent among sites and throughout the year of the study. The domination of a small isolated patch of habitable substratum by a sponge is most unusual, given that they are regarded as relatively poor recruiters. To understand how this association might be maintained, we determined the underlying genotypic diversity of the sponge population using starch-gel electrophoresis.P. spinifera is a clump-forming ascidian and usually occurs in clumps of up to 22 individuals. Electrophoretic surveys, based on six variable allozyme loci, revealed that at a total of five plots within three neighbouring New South Wales populations, single sponge genotypes may cover entire ascidian clumps; although a clump sometimes played host to more than one sponge clone. Allele frequencies (averaged across four loci that appear to conform to Mendelian inheritance) showed little variation among populations (standardised genetic variance,F ST=0.013). Nevertheless, sponge populations were genotypically diverse, with samples from 63 of 172 individual clumps displaying unique “clonal” genotypes. Moreover, multi-locus genotypic diversity within all sites approached the level expected for sexual reproduction with random mating. Taken together, these data imply thatH. laxus produces sexually-derived larvae that are at least moderately widelly dispersed. Given the relatively small size of the patches that this sponge inhabits, we also conclude that these larvae are good colonists and good spatial competitors on their ascidian hosts.

    Notes: Abstract Anthothoe albocincta, a common subtidal anemone along south-eastern Australia, reproduces both sexually through broadcast spawning and clonally through fission. Clones may be distinguished both by their electrophoretic genotypes and the colour of their tentacles and oral discs. Local populations typically consist of many, dense clonal aggregations. However, some clones appear to have locally extensive distributions, forming a series of separate aggregations. The capacity of clones to disperse among local populations is unknown. In this study we used an electrophoretic survey at six allozyme loci to quantify levels of variation among samples from each of 13 local populations and four geographic regions sampled between April 1992 and June 1993. These data revealed that populations of A. albocincta, separated by up to 930 km, were at least moderately subdivided. Levels of geographic variation were high and the average F ST value (standardised genetic variation) was 0.27 based on genotypes of all individual polyps. This value reflects substantial variation both within (F PR =0.13) and among (F RT =0.16) regions. Estimates of gene flow among both neighbouring populations and regions are therefore low (N e m=1.7 and 1.3, respectively). UPGMA (unweighted pair-group method using arithmetic averages) dendrograms suggest that a genetic discontinuity occurs at the very south-east corner of Australia, paralleling reports for two other south-eastern Australian marine invertebrates. In addition, our analyses and theoretical predictions imply that localised proliferation of clonal genotypes may have caused us to underestimate the potential importance of gene flow via larval dispersal. Moreover, the abundance and vast geographic range of this species suggests that widespread dispersal does occur. Collections from three populations covering the peroid December 1992 to June 1993 were examined by crude dissection along with histological sectioning, and showed A. albocincta to be dioecious, with unisexual clones. Eggs within the ovaries of six females sampled over a 3 mo period were small (96±4 μm) and similar to those of related species that produce planktotrophic larvae. In contrast, we found no evidence that clones were dispersed (shared) among neighbouring local populations. An average of only 6% of six-locus genotypes were common to pairs of local populations separated by up to 125 km, this being equal to the percentage expected through sexual reproduction alone. In addition, the percentage of shared genotypes did not decline with increasing geographic separation. These data imply that although asexual reproduction may be used to maintain local populations, the sexual production of genotypically diverse larvae is the primary source of widely dispersed colonists and hence of new clones.

    Notes: Abstract Two of the most abundant corals of the Great Barrier Reef Acropora cuneata and Acropora palifera, can be distinguished reliably by conventional taxonomic methods only if large specimens of both species are collected from the same uniform biotope. In this study gross and microscopic examination of the corallite structures of large coralla, collected from a reef crest biotope, were used to confirm the assignment of these specific names to two electrophoretically distinct groups of corals. Identifications based on genetic and morphological data gathered from small fragments of coralla collected from five different sites were in agreement for two sites within which skeletal data were adequate for taxonomic purposes (i.e. one reef crest and one outer lagoon site). However, specimens collected from the other three sites could be identified reliably only by using the electrophoretic data. These results confirm that small fragments from diverse habitats may not be adequate for morphologically based identification. The application of electrophoretic identifications to the results of an earlier electrophoretic survey of 76 sites on 11 reefs revealed that, although A. palifera and A. cuneata frequently co-occur, they have significantly different habitat distributions. Acropora cuneata is more commonly associated with high energy environments such as the reef crest, whereas A. palifera occurs most commonly in the sheltered reef lagoons and on the deeper reef slopes.

    Notes: Abstract Electrophoresis was used to provide genetic evidence of the mode of production of brooded planulae in each of four species of scleractinian coral collected from the central region of the Great Barrier Reef during September, October and November 1984. Comparisons were made of the multi-locus genotypes of planulae and their broodparents for two ahermatypic (non zooxanthellate) species,Tubastraea diaphana andT. coccinea and two hermatypic (zooxanthellate) species,Acropora palifera andSeriatopora hystrix. For both ahermatypic species, all planulae were found to be genetically identical to their broodparents, including 91 planulae which were heterozygous for at least one locus. These results are consistent with asexual (ameiotic) reproduction. In contrast, non-parental genotypes were detected in the majority of hermatypic broods, which is consistent with expectations for sexual reproduction with at least some outcrossing. These data confirmed that brooded planulae may be produced both sexually and asexually and countered the suggestion that electrophoretic studies of hermatypic corals may be weakened by the contaminating effect of the enzymes of their symbiotic xooxanthellae.
