    Notizen: We report an ultrastructural study of the reactivity of an IgG monoclonal antibody, which reacts with human dermal elastic fibres. On electron microscopy, immunogold labelling demonstrated the reactivity of HB8 with the mirofibrillar component of the fibres. In contrast, the electrondense granular component which contains elastin was unlabelled with HB8. A fine network of microfibrils from the level of the oxytalan fibres to the large elastic fibres of the deep dermis was labelled with gold particles. No reactivity was detected on collagen fibres. These results demonstrate that HB8 reacts with the microfibrillar glycoproteins of the elastic fibres in normal skin. The precise biochemical nature of the antigens identified by HB8 is still unknown.

    Notizen: BL13, a mouse monoclonal IgGl antibody raised against human B cells, blocked the function of the C3d receptor (CR2) and bound with high affinity (5x 108 L M−1) to CR2 on B lymphoma cells. Following capping with the second antibody BL13 inhibited C3d-dependent rosette formation of Daudi and Raji cells and C3b-dependent CR2-mediated rosette formation with B lymphoma cells, but did not inhibit CRI-mediated rosettes between C3b-bearing cells and peripheral blood lymphocytes. Competitive binding experiments between biotinylated BL 13 or anti-CR2 antibody HB-5 and unlabelled antibodies demonstrated that BL13 bound to an epitope that is distinct from that recognized In HB-5, and closely associated with that recognized by monoclonal antibody ann-B2, BL13 only reacted with some B cells and follicular dendritic cells in germinal centres in human lymph nodes, whereas HB-5 strongly reacted with circulating B cells and bound to most cells in the follicles. These results demonstrate the heterogeneity of antigenically defined CR2.

    Notizen: We have sought to determine whether rheumatoid factors (RF) produced in rheumatoid arthritis (RA) were different from physiological RF produced in normal, healthy adults. RF-secreting clones were established following Epstein Barr virus (EBV) stimulation of peripheral blood lymphocytes. Ten RF-secreting clones were established from seven RA patients and 16 from six healthy controls. All monoclonal RF (MRF), except two in each group, were monoreactive and ten of these were shown to have low to medium affinity for IgG, Fc, irrespective of their origin. A majority (74%) of the MRF bound to protein A, indicating that genes of the VHIII family were preferentially used for synthesizing these autoantibodies. The expression of cross-reactive idiotypes (CRI) by the MRF did not allow distinction between those derived from RA patients and controls. The VHI-associated CRI G8 and VHIII-associated CR1 D12 were expressed at low frequency in both panels of RF. These CRI have been shown to be expressed at high frequency in RF paraproteins. However, the idRQ idiotype was expressed within both panels of RF. A possible distinction between polyreactive and monoreactive MRF appeared to be light chain usage since all (four) polyreactive RF used λ chains while the normal κ/λ ratio was observed for monoreactive RF. The frequency of EBV-activated cells secreting IgM bearing CRI or secreting RF was determined and showed that CRI expression occurred with a higher frequency than did RF, suggesting a dissociation between CRI expression and RF activity.

    Notizen: The effector cell(s) in human antibody-dependent cell cytotoxicity (ADCC). with antibody-coated chicken erythrocytcs as targets, was studied by comparison of cell suspensions from various lymphoid organs and by means of various cell fractionation methods. Effector cells (K) were found mostly in peripheral blood, spleen, and bone marrow but not in tonsils, lymph nodes, and thymus. Effector cells bear Fc receptors and can form EA rosettes with the antibody-coated target cells. About 1.5% peripheral blood lymphocytes can form ‘high-avidity’ EA rosettes with targets coated at low antiserum concentration. Most of the effector cells belong to this small subset, as shown by experiments of selective depletion. Removal of most monocytes, T cells, or B cells from, or addition of T-cell-specific antiserum to, the effector cell suspensions did not affect ADCC. Effector cells in this model of ADCC therefore lack the conventional B- or T-cell markers but at least some of them are likely to bear C3 receptors.

    Notizen: Cells bearing Fc receptors were studied using a rosette technique with chicken erythrocytes coated with rabbit antibodies (EA). By comparison with other markers and by selective depletion of T and B cells it was demonstrated that EA rosettes were formed by about 10% of the monocytes, 7% of the T lymphocytes, and 45% of the B cells. In addition, 26% of EA-rosette-forming cells (EA-RFC) in the peripheral blood carried no other markers than C3 receptors. Marked differences were found in the percentages of EA-RFC in cell suspensions from various lymphoid organs: peripheral blood, 15.2%; spleen, 26.6%; bone marrow, 30.9%; lymph nodes, 2.8%; thymus, 0.3%; and tonsils, 0.5%. It was concluded that Fc receptors were present on subsets of different lymphocyte sub-populations; their presence could not be regarded as a specific marker of one of these subpopulations.

    Notizen: The purpose of the present study was lo examine the phenotype of the cutaneous immunocompetent cells in lichen planus and chronic graft versus host (GVH) reaction infiltrates, by the use of monoclonal antibodies directed against T cell populations and Langerhans cells.Our results in lichen planus suggest an immunological reaction similar to the delayed hypersensitivity reaction, including all the immunocompetent cell sub-populations, with a first stage of antigenic information by Langerhans cells (OKT6 +, BL6 +, HLA-DR+) and helper cells, and a second stage mediated by suppressor/cytotoxic cells.The results from the study of GVH reaction also suggest an effect mediated by suppressor/cytotoxic cells (OKT3+, OKT4−, OK.T8+, HLA-DR+).Our results favour the existence of a lymphocytotoxic process in lichen planus and chronic GVH reaction.

    Notizen: We have investigated lymphocyte subpopulations (six different markers) and T cell functions (mitogen responses and serum thymic factor determination) in twenty patients with psoriasis compared with thirty-five healthy subjects. Normal results were found for B cell markers, but significantly lower numbers of T cells were assessed by E-rosettes and anti HTLA serum. Mitogen responses were normal for PHA but depressed after Con A and PWM stimulation. Lastly, thymic factor levels were significantly higher in psoriasis than in normal subjects of the same age.These findings could be in favour of a T cell subpopulation defect involved in an auto-immune pathogenesis of psoriasis.
