    Notes: summary Sections of flash-frozen saliva particle mixture were examined by transmission electron microscopy. The saliva samples used consisted of 40% by volume parotid and 60% by volume submandibular sublingual fractions. One mL of the saliva was mixed with 0.1 mL of either hydrophilic or hydrophobic particles of 0.9 μm diameter. The micrographs of the saliva and hydrophilic particles mixes showed the multiple spherical particles in the salivary network structures. Micrographs of the saliva and hydrophilic particle mixes showed the presence of few such objects; when observed, the particles were located only in the electron dense part of the salivary structures.

    Notes: By reflected light microscopy, the thrombi formed on a variety of foreign surfaces have been shown to be composed of isolated cellular aggregates and an interaggregate red cell fibrin mesh. The thrombi viewed by this method were seen in two dimensions as they formed at the foreign surface blood interface. Standard histological evaluation of the thrombi in the third dimension, that is in the depth dimension, revealed columnar shaped cellular aggregates extending from the foreign surface into the blood and a red cell fibrin mesh surrounding the aggregates. The thrombi formed at Epon plate-blood interfaces were viewed with an electron microscope. Plates made of Epon embedding epoxy were used as the foreign surface exposed to blood, and the plates and fixed initial thrombi formed on them were embedded in more Epon. A layer of heavy metal staining, noncellular material was found covering the surface of the plates exposed to blood, with platelets deposited on this layer. Since platelets were not found adherent to the foreign surfaces directly, it is our postulate that the intervening film “conditions” the surface preliminary to platelet adhesion.

    Notes: This investigation developed experimental evidence for the influence of different surface energy states on tissue incorporation of biomedical materials. Implants of two smooth metals, each with three different surface energy states, were placed in the subdermal fascial plane of the backs of New Zealand White rabbits and were allowed healing times of 10 and 20 days. The implant surfaces were thoroughly characterized by physical-chemical criteria prior to surgical placement and again following removal from the tissue capsules generated by the host animals. Quantitative histopathologic analysis, using standard morphometric criteria, of the adjacent tissues revealed up to a threefold increase of fibroblastic-fibrocytic cells against the initially scrupulously cleaned, high-surface-energy materials. The cells were flattened and active, producing tenacious bonds through a thin pre-adsorbed protein-dominated “conditioning” film, that could be broken only by cohesive failure in the tissue itself. In contrast, the lower-surface-energy materials typical of standard dental implants were “walled off” by a cell-poor, nonadhesive capsule with a fibrous interface separated from a thicker “conditioning” film by a lipid-rich mucus zone. The advantages of proper surface treatment to favor the desired degree of biological adhesion are apparent.

    Notes: Using platelet-rich plasma freshly obtained by plasmapheresis, the spread areas of human platelets settled onto substrata of different surface energies were measured to evaluate the hypothesis that initial platelet spreading response is surface dependent. At 37°C, minimal platelet spread areas were found on substrata with critical surface tension between 20 and 30 mN/m (dynes/cm), and increased spreading and morphologic changes occurred on substrata of both higher and lower critical surface tension. This differential response was absent at 25°C. Therefore, measurement of spread platelet area at physiologic temperature rather than number of attached cells is proposed as a superior method of estimating apparent blood compatibility of new biomaterials.

    Notes: Organic-free specimens of 5 metals were prepared and characterized by parameters indicating their relative surface charge, surface texture, and surface energy prior to evaluation of their degree of thromboresistance. For the materials studied, there was no predictive correlation between the pre-implatation electrical rest potentials and ultimate thromboresistance when standard prosthetic rings were placed in the canine inferior vena cava. Surface textural inhomogeneities greater thatn a micrometer or so made thrombus formation a certainty. There was a positive postimplantation correlation between the presence of a thin, labile, proteinaceous deposit on the prosthetic ring lumens and the passivation of the “high-energy” materials towards thrombus accumulation. The metals which did consistently passivate, 316 Stainless Steel and Stellite 21, did so dynamically by acquiring an organic film within 2 hr after implantation. During continued residence in fresh, flowing blood these metals ultimately became dominated by an apparently monodisperse protein component of low critical surface tension. These findings were confirmed when thromboresistant pyrolytic carbon having initial surface properties similar to the metals but lacking reactivity and corrosivity was used as a comparison standard.

    Notes: Glow discharge treated (GDT) inorganic materials are sterile and free of all organic contaminants. Such materials implanted in the canine inferior vena cava often demonstrate significant thromboresistance. Standard Pyrex glass tubing was GDT in argon to provide a surface free energy above 70 dynes/cm, slight surface negativity, and other surface-physical modifications of its interface. Chromic acid cleaned lengths of the same tubing, and short segments whose edges were not fire-polished were used as controls. Upon canine implantation, the in vivo results were similar in independent surgical laboratories. The glass was implanted as 1.4 cm long rings or as 5 cm long tubes in separate medical centers according to different protocols. GDT specimens remained patent; all controls accumulated thrombi. A remarkably pure, low critical surface tension, labile protein coating covered all cylindrical lumens after 2 hr in vivo. This coating diminished within 2 weeks and was essentially absent after 480 days, even though scanning EM showed evidence of a micron thick luminal film, probably eroding glass. No emboli were found in the implant animal's kidney or lungs at any stage.
