    Notes: Abstract Methods for the preparation and analysis of a new renal radiopharmaceutical,99mTc-thiodiglycolic acid (TDGA), are reported. The kit for Tc-TDGA contains a lypophilized acidic (pH 2.5) mixture of 5 mg TDGA and 0.05 mg SnCl2·2H2O. Acetate buffer has been found to be a suitable solvent for paper chromatography of99mTc-TDGA. The results of the quantitative organ distribution studies in rats and rabbits indicated the characteristics of99mTc-TDGA to be intermediate to the renal tubular agent131I-Hippuran and the GFR agent99mTc-DTPA.

    Notes: Abstract The need for defining the assay conditions assumes greater significance for the manufacturer of RIA kits. This stems from the consideration that despite high quality of kit reagents in the kits, it is the definition of optimum conditions of assay that makes them effective and also permits to evaluate the limits of the sensitivity and precision. In this connection we have studied in detail a number of parameters mainly to define the stability of the assay system to changes in the protein concentration, the influence of polyethylene glycol in various ionic media, incubation parameters for systematic error analysis in three separation systems based on dextran coated charcoal, plain polyethylene glycol and polyethylene glycol assisted second antibody. Second antibody in a preprecipitated form was found to be readily adaptable as a reagent. Among the three methods of separation studied, the second antibody employed as a complex with primary antibody assisted by polyethylene glycol had superior separation without any specific criticality, characteristics as judged by the variations at various dose levels over an extended period and provided a practical procedure for routine assays.

    Notes: Abstract Three different radiotracers of testosterone incorporating125I at position 3 or 19 were prepared and tested for usability in radioimmunoassays employing the antisera raised at our laboratory. Histamine was radioiodinated and conjugated to suitable testosterone derivatives. On optimization, tracers with high purity, good immunoreactivity and long stability could be obtained for all the three tracers. However, the usability of a tracer will also depend on the antiserum used and this was also demonstrated using different pairs of antisera and tracers.

    Notes: Abstract A radioimmunoassay procedure to measure estradiol levels in human serum was developed using125I labeled estradiol and antiestradiol antibody raised in-house using the estradiol-6-CMO-BSA as the immunogen. Controlled process serum was used to maintain the serum matrix and sample-standard identity. The assay was validated by analysing several (n=40) samples and comparing with established commercial assays. The assay had a sensitivity of 35 pg/mL and a range of 50 pg/mL to 5 ng/mL. The interassay C.V. were between 2 to 10% while the intraassay C.V. were between 2 and 8%. This assay is sensitive enough for monitoring ovulation and hypersecretions in ovarian tumours and was primarily developed to be used as an in-house assay in the local hospitals.

    Notes: Abstract Tracer specificity plays an important role in the radioimmunoassay (RIA) of steroid homones. In this paper, we describe the preparation and purification of125I-labeled cortisol derivative with a carboxymethyloxime-histamine bridge. The investigation on the method of purification showed that HPLC could be adopted for the routine preparation of a pure, homogeneous tracer. The retention time observed in HPLC for125I-histamine-CMO-cortisol conjugate could be used as an index for qualitative and quantitative assessments.

    Notes: Abstract A novel method to eliminate T3 from gross sera/plasma based on prior treatment with 8-anilino-1-naphthalene sulphonic acid has been standardized. This method is rapid with a minimum loss of protein and uses only small quantities of charcoal. The treated sera have been tested for acceptance in the RIA system for routine assays.
