    Notes: Abstract. The current strategy of using transgenic crops expressing insecticidal protein toxins is placing increasing emphasis on the discovery of novel toxins, beyond those already derived from the bacterium Bacillus thuringiensis. Here we review the cloning of four insecticidal toxin complex (tc) encoding genes from a different bacterium Photorhabdus luminescens and of similar gene sequences from Xenorhabdus nematophilus. Both these bacteria occupy the gut of entomopathogenic nematodes and are released into the insect upon invasion by the nematode. In the insect the bacteria presumably secrete these insecticidal toxins, as well as a range of other antimicrobials, to establish the insect cadaver as a monocultural breeding ground for both bacteria and nematodes. In this review, the protein biochemistry and structure of the tc encoding loci are discussed in relation to their observed toxicity and histopathology. These toxins may prove useful as alternatives to those derived from B. thuringiensis for deployment in insect-resistant transgenic plants.

    Notes: Abstract The 5′ untranslated region of HLA-F contains a polypurine tract comprising repeats of tri- and hexa-nucleotide motifs. We have recently demonstrated that this polypurine tract is highly polymorphic by using the polymerase chain reaction. Here, we demonstrate that some of the alleles can be explained by a deletion of approximately 100 by DNA and show that alleles of this novel, highly polymorphic locus are as strongly associated with haemochromatosis as HLA-A3 or D6S105-8. The observed frequency of heterozygosity at HLA-RF is extremely high (95%) and this locus has been found to be informative in pedigrees that are non-informative at HLA-A and D6S105. We also show an example of replication slippage at HL,A-F in one pedigree.

    Notes: Abstract The BglI restriction fragment length polymorphism (RFLP) of the human factor VIII (FVIII) gene is potentially useful in linkage studies in haemophilia A. The sequence at the RFLP locus is not known, therefore it is not amenable to analysis by the polymerase chain reaction (PCR) and Southern blotting is required. We present a novel approach for analysis of the BglI RFLP using the PCR targeted to known sequence downstream in exon 26 of the FVIII gene. Briefly, the size of the genomic restriction fragment carrying the PCR target depends upon whether the RFLP site is present or absent. If fragments of the required size are isolated from a genomic digest and used as substrates in the exon 26 PCR, the generation of a product in one or other fraction indicates the upstream RFLP status. We have called this approach “virtual PCR”, since PCR is used to obtain information about the RFLP without amplifying the locus itself.

    Notes: Twenty seven insulin-dependent diabetic patients received a continuous intravenous infusion of an insulin-glucose-potassium solution whilst undergoing elective surgical or obstetric procedures. The dose of insulin was adjusted according to blood glucose values. It was concluded that the regimen is safe and simple to manage and should result in good control of blood glucose levels.

    Notes: The effects of intravenous morphine (10 mg/70 kg) on the ventilatory response to CO2 were studied in two groups of subjects, young (18-29 years) and old (66-85 years), prior to elective surgery. In both groups morphine caused a signijcant depression of respiration as judged by a reduction in the slope of the CO2 response curve. a reduction in the calculated ventilation at an end tidal CO2 tension of 7.3 kPa, a rise in resting end tidal CO2 and a rise in the CO2 threshold. There were no significant differences between the two groups in the changes produced by the drug, suggesting that acute respiratory depression after a single intravenous injection of morphine is similar in old and young people.

    Notes: Summary The tissues of the domestic hen's reproductive tract have been shown to contain both acidic and neutral mucins. The presence of an egg in the isthmus was always accompanied with an increase in the concentration in the isthmus secretory epithelium of PAS-positive mucins without free acidic groups. Staining of uterine tissue for strongly acidic groups with basic dyes was greater when an egg had been present in the shell gland at the time of death and the same was true for tissues of the upper oviduct. The results are discussed with special reference to the periodic secretion of organic mammillary core and egg shell mucins.
