    Notes: Abstract Aims/hypothesis. The circulating concentrations of endothelin-1 (ET-1), a peptide derived from endothelium, are increased in hypertension and diabetes. Endothelin-1 has recently been shown to be an insulinotropic agent. The mechanism of action of endothelin-1 on the endocrine pancreas has not yet been clarified. Methods. We investigated the action of endothelin-1 on the insulin secretion, the binding of 125I-ET-1 to beta cells as well as its effects on purified beta and non-beta cells from normal rats. The expression of endothelin receptors in alpha- and beta-cell lines and in normal rat islets was also studied. Results. First, we studied the effects of endothelin-1 on insulin secretion from beta-cell lines (INS-1, βTC3 and MIN6). At all endothelin-1 concentrations applied (1 pmol/l to 1 μmol/l) no change in insulin secretion was found. Ligand-binding experiments on βTC3 cells showed no specific binding of 125I-ET-1. A prominent expression of ETA-receptor mRNA in an alpha-cell line (αTC1.9) and in normal rat islets was found whereas no expression was found in INS-1 cells. No influence of endothelin-1(1 μmol/l) on insulin secretion stimulated by glucose was detected from purified beta cells. Endothelin-1-(100 nmol/l) increased, however, both insulin and glucagon secretion from a mixture of purified beta and non-beta cells indicating that alpha cells seem to have a key role for the action of ET-1 on insulin secretion. Conclusion/interpretation. The insulinotropic impact of endothelin-1 is not caused by a direct action on the beta cells but seems to be mediated by a paracrine action, probably secondary to enhanced release of glucagon from the endothelin receptor positive alpha cells.     Notes: Summary Endothelin-1 (ET-1), a potent endothelium-derived vasoconstrictor peptide, is secreted in response to insulin. Elevated circulating ET-1 levels have been found in patients with diabetes mellitus and vascular dysfunction. The question arises whether ET-1 acts as a direct modulator of insulin secretion. To test this, we studied the effects of ET-1 on isolated mouse islets of Langerhans. ET-1 (1 nmol/l–1 μmol/l) dose-dependently stimulated insulin secretion from islets incubated in the presence of 16.7 mmol/l glucose (p 〈 0.05). The effect of ET-1 is glucose-dependent since no potentiation was found at 3.3 mmol/l glucose. Furthermore, ET-1 induced a large, transient increase in glucose-stimulated insulin secretion during islet perifusion in the presence (p 〈 0.001), but not in the absence, of extracellular Ca2 + . The rate of 45Ca2 + -efflux from 45Ca2 + -prelabelled islets was transiently stimulated by ET-1 during perifusion at 16.7 mmol/l glucose in the presence of extracellular Ca2 + (p 〈 0.001). A short-lived increase in 45Ca2 + -efflux was also observed in the absence of extracellular Ca2 + (p 〈 0.05). It is suggested that the effects of ET-1 on insulin secretion are critically dependent on influx via Ca2 + -channels. In addition, ET-1 transiently enhanced 86Rb + -efflux from 86Rb + -prelabelled islets both in the presence (p 〈 0.001) and in the absence (p 〈 0.001) of extracellular Ca2 + suggesting that ET-1 does not elicit insulin secretion by inhibition of the potassium permeability. Our study provides evidence that ET-1 stimulates insulin secretion via a direct effect on the islets of Langerhans. [Diabetologia (1996) 39: 1030–1035]

    Notes: Summary Diabetic patients with polyneuropathy develop motor dysfunction. To establish whether motor dysfunction is associated with muscular atrophy the ankle dorsal and plantar flexors of the non-dominant leg were evaluated with magnetic resonance imaging in 8 patients with symptomatic neuropathy, in 8 non-neuropathic patients and in 16 individually matched control subjects. In the neuropathic patients the muscle strength of the ankle dorsal and plantar flexors was reduced by 41 % as compared to the non-neuropathic patients (p 〈 0.005). Volume of the ankle dorsal and plantar flexors was estimated with stereological techniques from consecutive cross-sectional images of the lower leg. The neuropathic patients had a 32 % reduction in volume as compared with the non-neuropathic patients (p 〈 0.005). To determine the regional distribution of atrophy cross-sectional magnetic resonance images were performed at predetermined levels of the lower leg in relation to bone landmarks. In the neuropathic patients there was an insignificant increase of 3 % of muscle area at the proximal lower leg level, whereas the atrophy was 43 % (p 〈 0.002) at the mid lower leg level and 65 % (p 〈 0.002) distally. Analysis of individual muscles confirmed that the atrophy predominated distally. We conclude that muscular atrophy underlies motor weakness at the ankle in diabetic patients with polyneuropathy and that the atrophy is most pronounced in distal muscles of the lower leg indicating that a length dependent neuropathic process explains the motor dysfunction.     Notes: Abstract –  Predicting species distributions has important implications for the conservation and management of freshwater fishes, particularly in areas such as the Great Plains, USA where human impacts have resulted in extirpations and declines for numerous native species. There are a number of statistical approaches for constructing distributional models; the accuracy of each is likely dependent on the nature of the environmental gradients, species responses to those gradients and the spatial extent of the modelling. Thus, it is important to compare multiple approaches across species and habitats to identify the most effective modelling approach. Using geographical information system (GIS) derived characteristics of stream segments as predictors, we tested the model performance of three methodologies – linear discriminant function analysis, classification trees and artificial neural networks (ANN) – for predicting the occurrence of 38 fish species in a Great Plains river basin. Results showed that all approaches predicted species occurrences with relatively high success. ANN generally were the best models, in that they generated the most significant models (35 of 38 species) and most accurately predicted species presence for the greatest number of species (average correct classification = 81.1%). The importance of GIS variables for predicting stream fish occurrences varied among species and modelling techniques, but were generally strong predictors of species distributions, including the federally endangered Topeka shiner Notropis topeka. In summary, predictive models should be viewed as both competitive and complementary methodologies for establishing quantitative linkages between fish species and their environment. Our study demonstrates the potential utility of such an approach for guiding conservation efforts for stream fishes of the Great Plains, USA.

    Notes: Abstract  The emergence phenology and feeding ecology of annual cicadas in tallgrass prairie are poorly documented. However, these large insects are abundant, and their annual emergence represents a potentially important flux of energy and nutrients from belowground to aboveground. We conducted a study at Konza Prairie Research Natural Area in eastern Kansas to characterize and quantify cicada emergence and associated energy and nutrient fluxes. We established emergence trap transects in three habitat types (upland prairie, lowland prairie, and riparian forest), and collected cicadas every 3 days from May to September. A subset of trapped cicadas was used for species- and sex-specific mass, nutrient, and stable isotope analyses. Five species were trapped during the study, of which three were dominant. Cicadetta calliope and Tibicen aurifera exhibited significantly higher emergence production in upland prairie than in lowland prairie, and were not captured in forested sites at all. T. dorsata emerged from all three habitat types, and though not significant, showed a trend of greater abundance in lowland grasslands. Two less abundant species, T. pruinosa and T. lyricen, emerged exclusively from forested habitats. Nitrogen fluxes associated with total cicada emergence were estimated to be ∼4 kg N ha–1 year–1 in both grassland habitats, and 1.01 kg N ha–1 year–1 in forested sites. Results of stable isotope analyses showed clear patterns of resource partitioning among dominant cicada species emerging from grassland sites. T. aurifera and C. calliope had δ13C and δ15N signatures indicative of feeding on shallowly rooted C4 plants such as the warm-season grasses dominant in tallgrass prairie ecosystems, whereas T. dorsata signatures suggested preferential feeding on more deeply rooted C3 plants.
