Zusammenfassung Eine einfache Methode zur Visualisierung enzymatisch aktiver Komponenten wird am Beispiel pectinolytisch, amylolytisch und proteolytisch wirksamer Enzyme beschrieben. Die Enzyme werden durch ultradünnschicht-isoelektrische Focussierung bzw. ultradünnschicht Gradientengelelektrophorese getrennt. Auf das Trenngel wird ein 120 μm oder 240 μm dickes, auf Folie polymerisiertes Acrylamidgel aufgerollt, welches ein für das jeweilige Enzym spezifisches und auf optimalen pH-Wert gepuffertes Substrat enthält. Nach einer definierten Reaktionszeit wird es vom Trenngel abgenommen, kurz in Wasser gewaschen, angefärbt und entfärbt. Die Trennschärfe der Focussierung oder Elektrophorese ist in den Abklatschgelen exakt reproduziert und die Nachweisempfindlichkeit gegenüber bisherigen Enzymvisualisierungen deutlich erhöht. Das Trenngel kann ohne wesentliche Bandenverluste zur Proteinanfärbung weiter behandelt werden.

Abstract A simple method for activity staining of pectinolytic, amylolytic and proteolytic enzymes is described. The enzymes are separated by ultrathinlayer isoelectric focusing or ultrathinlayer gradient gel electrophoresis. After successful separation an ultrathin acrylamide gel on foil support containing a specific substrate buffered to optimal pH for the enzyme to be visualized, is pressed onto the surface of the separating gel. After the reaction time the print gel is removed, washed with water, stained and destained. The resolution of the ultrathinlayer isoelectric focusing or ultrathinlayer gradient gel electrophoresis is exactly reproduced in the print gel, the sensitivity in comparison to conventional activity staining methods is improved. The separating gel can be further employed for protein staining without loss of activity.
